This paper outlines the construction of a novel vector system comprising interchangeable terminators, as well as a multiple cloning site (MCS), to facilitate the transformation of the fungal plant pathogen Botrytis cinerea. Previous molecular studies on B. cinerea have relied upon the pLOB1 based vector system (controlled by the Aspergillus nidulans oliC promoter and a region reported to be the B. cinerea tubA terminator). Investigations, however, have revealed that, rather than the genuine B. cinerea tubA terminator, the pLOB1 terminator fragment is from another gene locus within the genome. Because previous studies have found that terminators aide in transcript stability, the main aims of this study were to develop and evaluate both vector systems, pOT (controlled by the A. nidulans oliC promoter and A. nidulans trpC terminator) and pLOB, with a range of exogenous genes, including enhanced green fluorescent protein (eGFP), monomeric red fluorescent protein (mRFP), luciferase (LUC) and β-glucuronidase (GUS). Our investigations demonstrate that pLOB and pOT based vectors are capable of expressing all four reporter genes and may be applied to future molecular studies on B. cinerea and other related ascomycetes. Additionally, this is the first reported expression of mRFP and LUC in B. cinerea.

咀嚼と脳賦活との関わりについての基礎的検討として, 若年者を対象として個体が示す顎顔面骨格構成および咬合形態によって, 咀嚼時の脳血流動態に差異が存在するかどうかについて調べるため, 酸素標識の水とポジトロンCT(PET)を用いて検討を行った.研究対象は, 21∿32歳の健常ボランティア男性6名である.それぞれ頭部X線規格写真の撮影と歯列模型の採得を行い, 顎顔面骨格構成および咬合について検討した.脳血流動態については安静時とガムベース咀嚼時にPETを用いて測定を行った.得られた脳血流画像について関心領域を設定し, ガム咀嚼に伴う局所脳血流(rCBF)の変化を求めて咬合や顔面形態との関連について検討した.その結果, ガム咀嚼による脳血流変化には個体差が大きいものの, 上・下顎骨に調和のとれたおおむね良好な咬合を有する者では, 1次運動感覚領下部(ローランド野), 島, 小脳半球などで明らかな脳血流の増大が認められた.一方, 顎顔面骨格や咬合に種々の問題を有する不正咬合者では咀嚼による脳血流変化が正常咬合者と異なる傾向が認められ, 1次運動感覚領下部や島での明らかな賦活は認められない者もいた.以上のことから, 咀嚼による脳の賦活化部位とその量は個体間でかなり変異に富んでおり, それには個体の示す顎顔面骨格構成および咬合とそれに関わる咀嚼運動様式が関与している可能性が示唆された.

Background:Cardiovascular guidelines include risk prediction models for decision making that lack the capacity to include novel predictors.Methods and Results:We explored a new “predictor patch” approach to calibrating the predicted risk from a base model according to 2 components from outside datasets: (1) the difference in observed vs. expected values of novel predictors and (2) the hazard ratios (HRs) for novel predictors, in a scenario of adding kidney measures for cardiovascular mortality. Using 4 US cohorts (n=54,425) we alternately chose 1 as the base dataset and constructed a base prediction model with traditional predictors for cross-validation. In the 3 other “outside” datasets, we developed a linear regression model with traditional predictors for estimating expected values of glomerular filtration rate and albuminuria and obtained their adjusted HRs of cardiovascular mortality, together constituting a “patch” for adding kidney measures to the base model. The base model predicted cardiovascular mortality well in each cohort (c-statistic 0.78–0.91). The addition of kidney measures using a patch significantly improved discrimination (cross-validated ∆c-statistic 0.006 [0.004–0.008]) to a similar degree as refitting these kidney measures in each base dataset.Conclusions:The addition of kidney measures using our new “predictor patch” approach based on estimates from outside datasets improved cardiovascular mortality prediction based on traditional predictors, providing an option to incorporate novel predictors to an existing prediction model.

Oxidative stress caused by reactive oxygen species is considered a major mediator of tissue and cell injuries in various neuronal conditions, including neurological emergencies and neurodegenerative diseases. Molecular hydrogen is well characterized as a scavenger of hydroxyl radicals and peroxynitrite. Recently, the neuroprotective effects of treatment with molecular hydrogen have been reported in both basic and clinical settings. Here, we review the effects of hydrogen therapy in acute neuronal conditions and neurodegenerative diseases. Hydrogen therapy administered in drinking water may be useful for the prevention of neurodegenerative diseases and for reducing the symptoms of acute neuronal conditions.

The autocorrelation of angle resolved photoemission data from the high temperature superconductor Bi2Sr2CaCu2O8+delta shows distinct peaks in momentum space which disperse with binding energy in the superconducting state, but not in the pseudogap phase. Although it is tempting to attribute a nondispersive behavior in momentum space to charge ordering, a deconstruction of the autocorrelation reveals that the nondispersive peaks arise from the tips of the Fermi arcs, which themselves do not change with binding energy.

A 2-year-old castrated male mongrel dog presented with a well-demarcated fluctuant dermal mass, located on the back of the neck. Grossly along with cystic structures filled with a black greasy fluid, when cut open. Microscopically, the mass was multi-lobulated. The lobules consisted of neoplastic basaloid cells and showed central degeneration, forming multiple central cystic structures filled with dark melanin-pigmented materials. Immunohistochemically, the neoplastic cells were strongly positive for CK14 and partially positive for CK19, but negative for CK7, CK8/18, CD34, S-100, Melan-A and α-SMA. Based on the findings, the present case was diagnosed as a feline-type basal cell tumor characterized by cystic structures filled with abundant black fluid.

ペルーおよび日本国内で採取したラマ47頭,アルパカ27頭および両者の交雑種1頭とそのアルパカへの戻し交雑種1頭の計76頭の血液を用いて,19座位の血液タンパク質・酵素型を電気泳動学的に解析し,以下に示す成績を得た。1)血漿タンパク質 : Albumin, Haptoglobin,血漿酵素 : Alanine aminotransferase, Aspartate aminotransferase, γ-Glutamyltranspeptidase,赤血球タンパク質 : Haemoglobin,ならびに赤血球酵素 : Acid phosphatase, Catarase, Glucose-6-phosphate dehydrogenase, Phosphoglucomutase, Phosphohexose isomeraseの計11座位では多型は認められなかった。2)血漿タンパク質4座位 : Post-albumin(Po),Gc-protein(Gc),Transferrin(Tf)およびγ-globurin field protein(γG),血漿酵素3座位 : Amylase(Amy),Creatine kinase(CK)およびLeucine aminopeptidase(LAP),ならびに赤血球酵素1座位 : EsteraseD(EsD)の計8座位に多型が認められた。これら8座位のうちTfでは6型,PoおよびGcでは4型,γ-G, AmyおよびEsDでは3型,LAPおよびCKでは2型が認められた。ラマおよびアルパカにおけるこれら8座位の総合的な父権否定率は,0.931および0.867であった。3)ラマとアルパカとの間で血液タンパク質・酵素型を比較したところ,Gc, AmyおよびEsDにおいて種間差が認められた。すなわち,GcおよびAmyでは両種で共通な易動度を示すバンド以外に各々種特有の易動度を示すバンドが存在し,またEsDでは両種間でバンドの易動度が異なっていた。4)ラマとアルパカの交雑種,ならびにアルパカへの戻し交雑種の血液タンパク質・酵素型はラマあるいはアルパカと共通であった。5)CKおよびEsDを除く,17座位の遺伝子頻度に基づいて算出したラマとアルパカとの間の遺伝的距離は0.035であった。以上の成績から,血液タンパク質・酵素型の解析はラマおよびアルパカの集団の遺伝子構成を推定する上で有力な指標となることが明確となった。一方,ラマとアルパカが遺伝的に極めて近縁な関係にあることを裏付けるものと判断された。Blood samples of llamas and alpacas were classified by using electrophoretic procedures in the polymorphism at 19 loci. Electrophoretic variation was found for 8 loci, namely plasma proteins : post albumin (Po), Gc protein (Gc) and transferrin (Tf) and γ-globurin zone protein (γG), for plasma enzymes : amylase (Amy), creatine kinase (CK) and leucine aminopeptidase (LAP), and for red cell enzyme : esteraseD (EsD). Synthetic probabilityes of paternity exclusion about the 8 loci for llamas and alpacas were 0.931 and 0.867, respectively. No variants were found for plasma proteins : albumin and haptoglobin, for plasma enzymes : alanine aminotransferase, aspartate aminotransferase and γ-glutamyltranspeptidase, for red cell haemoglobin, and for red cell enzymes : acid phosphatase, catarase, glucose-6-phosphate dehydrogenase, phosphoglucomutase and phosphohexose isomerase. Nei's genetic distance between llamas and alpacas on the 17 loci (except CK and EsD) was 0.035. Preliminary estimate of the genetic distance measure may suggest that llamas and alpacas are more likely related as subspecies than as separate species.

<p>Agassiz's desert tortoise (<i>Gopherus agassizii</i>) reaches the southern edge of its range in the Sonoran Desert of California. The reproductive ecology of this wide-ranging species is understudied here compared to populations in the adjacent Mojave Desert. Understanding potential geographic variation in reproductive ecology is important for effective management of conservation-reliant species like <i>G. agassizii</i>. We studied the fecundity and clutch phenology of female <i>G. agassizii</i> at two study sites in the Sonoran Desert region of Joshua Tree National Park over five years (1997–1999–2015–2016) spanning two of the strongest El Niño events on record and an epic drought. Across all years, mean clutch size was 4.3±1.5 eggs, mean clutch frequency was 1.78 clutches/female/year, and mean X-ray egg width was 36.51±1.56 mm, all of which are comparable to other published studies both in the Sonoran and Mojave deserts of California. Our results generally support earlier published findings that <i>G. agassizii</i> utilize a bet-hedging strategy of consistently producing small clutches almost every year, even during times of low annual primary productivity. A regionally warmer climate in the Sonoran Desert of California appears to have an effect on the timing of egg production, as the earliest dates of egg visibility in our study (April 6) were approximately two weeks earlier than the earliest dates reported for <i>G. agassizii</i> in the Mojave Desert. Shelled eggs were no longer visible in tortoises after mid-June in all years but the El Niño year 1998, when eggs were visible until mid-July.</p>

As a part of the Science Exchange Program between Nagoya University and Russian Academy of Sciences, heligmosomid nematodes of the small mammals captured in the adjacent area of Akademgorodok City, West Siberia, Russia was investigated in summer, 1994. The host materilas of the survey were 27 individuals of the Soricidae and 60 individuals of the Muridae, and its specific names were shown below; Sorex araneus, S. caecutiens, S. isodon, S. minutus, S. tundrensis, Apodemus agrarius, A. peninsulae, Clethrionomys glareolus, C. rufocanus, C. rutilus, Microtus agrestis, M. arvalis, M. gregalis and M. oeconomus. Among them, Longistriata depressa was obtained from S. araneus, S. isodon, S. minutus and S. tundrensis, and Heligmosomoides neopolygyrus was obtained from A. agrarius and A. peninsulae, respectively. However, there was no heligmosomid obtained from the genera Clethrionomys or Microtus. This is the first host record of L. depressa from S. isodon and S. tundrensis, and the locality record of H. neopolygyrus from West Siberia.

Four microalgal strains, NN-G, TB-G, CC-B, and SC-B isolated from green water of tilapia culture ponds in the Philippines were cultivated in 10ml of ESS liquid medium at 23℃ with a 12h light-12h dark cycle. The cell forms of NN-G and TB-G were green small round and green large oval, respectively, while both CC-B and SC-B had brown cubic or quadrangular cells. Predominant bacteria isolated from the four microalgal cultures were commonly restricted to Moraxella-Acinetobacter group, Pseudomonas and Flavobacterium. Strains NN-G and TB-G contained chlorophyll b, chlorophyll a, violaxanthin and zeaxanthin, while strains CC-B and SC-B possessed chlorophyll c, chlorophyll a, and fucoxanthin as major pigments. Phylogenetic analysis based on SSU rDNA sequences demonstrated that strains NN-G, TB-G, CC-B and SC-B have close taxonomic relationships with Chlorella spp., Tetraselmis spp., Chaetoceros spp., and Chaetocertos spp., respectively.

Cardiovascular magnetic resonance is a well-established tool for the quantification of focal fibrosis. With the introduction of T1 mapping, diffuse myocardial processes can be detected and quantified. In particular, infiltration and storage disorders with large disease-related changes, and diffuse fibrosis where measurement is harder but the potential impact larger. This has added a new dimension to the understanding and assessment of various myocardial diseases. T1 mapping promises to detect early disease, quantify disease severity and provide prognostic insights into certain conditions. It also has the potential to be a robust surrogate marker in drug development trials to monitor therapeutic response and be a prognostic marker in certain diseases. T1 mapping is an evolving field and numerous factors currently preclude its standardization. In this review, we describe the current status of T1 mapping and its potential promises and pitfalls. [Purpose] To present the case of the dramatic reduction in pain, disability, and neurologic symptoms following the reduction of forward head translation and increased cervical curvature in a patient suffering from post-surgical radiculopathy. [Subject and Methods] A 52-year-old male mechanic presented with chronic neck pain, unilateral paresthesia along the C5 and C6 dermatome distributions and diminished unilateral grip strength for 12 years following a C5–C6 cervical discectomy and fusion. Outcome measures included the neck disability index, the numerical pain rating scale, and the Zebris cervical range of motion system. Radiographs and computerized posture analysis revealed excessive forward head posture. Initial traditional ‘symptom-relief’ chiropractic rehabilitation was provided, followed by CBP® structural rehabilitation of head and neck posture with a 2.5 year follow-up. [Results] The initial traditional chiropractic rehabilitation did not improve posture or disability scores. CBP methods resulted in radiograph-verified postural alignment improvements corresponding with clinically significant improvements in the patient’s neurologic condition, pain and disability scores. These results were maintained at a 2.5 year follow-up with minimal treatment. [Conclusion] Patients with post-surgical axial symptoms and/or radicular complaints should be screened for altered cervical alignment and anterior head translation. Future studies should attempt to duplicate these positive results in a trial with long-term follow-up.
