Three-dimensional earth sciences information is being made available in digital formats because such information is useful and understandable to various users. In this study, we develop a three-dimensional hydrogeological model of Tsukuba City and the surrounding region, and develop a method for estimating the hydraulic conductivity of aquifers in combination with the three-dimensional hydrogeological model and other related hydrogeological information with the primary objective of proposing a method for evaluating groundwater resources. The three-dimensional hydrogeological model was developed using a geological map and drilling data with data interpolated using a geostatistical method. The model developed reproduced the data used well, and it was found to be accurate for practical use. Moreover, this model could be used for various types of visualization, from which information could be extracted and applied immediately to numerical simulations; this provided information that was useful and understandable for evaluating groundwater resources. The proposed method for estimating the hydraulic conductivity of aquifers was developed from hydraulic conductivities obtained from various drilling surveys, which were grouped together taking geology and stratigraphy into consideration based on a three-dimensional hydrogeological model, and using a statistical analysis. This method can be used to rapidly evaluate trends in hydraulic conductivity by integrating data distributed over a broad area. In addition, if a columnar section of an evaluated point is obtained, this method can be used to determine the actual hydraulic conductivity of aquifers. Therefore, these methods were found to be effective for evaluating groundwater resources.

Gene silencing mediated by RNA interference (RNAi) is expected to have a beneficial impact on the treatment of many diseases because of its potency, selectivity and versatility. To maximize the potential of RNAi effectors such as small interfering RNA and microRNA in clinical therapy, the development of a practical delivery system is required, especially for systemic administration. Recent studies demonstrated that chemical modification of these small RNAs and/or encapsulation of them into lipid nanoparticles is a promising strategy to achieve targeted delivery via systemic administration. In this review article, we introduce recent progress of the research on systemic delivery systems for RNAi therapeutics and consider crucial elements for the design of lipid nanoparticles as a small RNA vector.

Diatom assemblages in a rice paddy inundated by the 2011 Tohoku Earthquake tsunami were studied. We collected diatom samples from a rice paddy in Sabusawa Island in Miyagi Prefecture on 14 August 2011. The rice paddy was intruded by seawater during the high tide of the spring tide as a result of destruction of the coastal levee and the land subsidence after the earthquake. The salinity of the paddy was slightly higher than that of the coastal seawater, and naturally, rice was not cultivated at that time. In total 117 diatom taxa (including 22 unidentified ones) belonging to 50 genera were observed. The diatom assemblages consisted of fresh, brackish, and marine water species, and moreover, extinct Miocene marine diatoms. The dominant species was Tabularia parva on Zannichellia palustris and in a floating algal floc, and Navicula phylleptsoma on the surface soil. Halamphora luciae was subdominant in all samples. Judging from the past literature, these diatoms all tolerate changes in salinity.

Objective Shewanella bacteremia is an uncommon but potentially fatal disease. Although hepatobiliary diseases have been proposed to be risk factors for various Shewanella infections, little is known about the features of Shewanella bacteremia in patients with hepatobiliary diseases. This study aims to characterize the presentation and risk factors of Shewanella bacteremia in patients with hepatobiliary diseases. Methods We retrospectively investigated the clinical features, microbiology and outcomes of patients with Shewanella bacteremia who were admitted to a tertiary medical center between January 2001 and December 2010. All isolates were confirmed to the species level using 16S rRNA sequencing analyses. The English language medical literature was searched for previously published reports. Results Fifty-nine cases of Shewanella bacteremia, including nine at the hospital, were identified, 28 (47.4%) of which involved underlying hepatobiliary diseases, representing an important risk factor. In 12 of the 28 cases, the infections involved the hepatobiliary system; with a tendency towards an Asian origin. In our case series of nine patients, Shewanella haliotis was isolated in five patients. The majority of our patients lived in coastal areas, consumed seafood regularly and developed bacteremia during the summer season. Conclusion It is recommended that the possibility for Shewanella infection be considered in patients with bacteremia and also underlying hepatobiliary diseases, particularly if patients present with hepatobiliary infections, a history of seafood, or development of the disease during the summer.

理論化学の研究対象において金属表面や金属錯体が主要になりつつある現在,相対論的密度汎関数法(RDFT)の重要性は増している.この方法は,多電子系の電子状態を金属を含む大規模系の反応や物性を定量的に取り扱えるため,大規模金属系の理論研究においてもっともすぐれた理論の1つであることは間違いない.RDFTは量子電気力学にもとづく確かな土台をもつ理論であり,化学における相対論的計算の主要理論となっている.本レビューでは,RDFTの基礎を説明した後,最新の2成分RDFTに至るまでの具体的な道筋を明らかにする.さらには,RDFTが将来的に進むであろう道筋も示すことも目的としている.

アズキ熱水抽出物(アズキ煮汁)の抗腫瘍活性及びその作用機構の一つであるアポトーシス誘導について検討を行った.アズキ熱水抽出物をDIAION HP-20で処理した後,蒸留水,40%エタノール,60%エタノール,80%エタノールと順に溶出溶媒を切り換え,各溶出画分を得た.これらの溶出画分を用いてヒト胃癌細胞(KATO III cells)の形態学的変化,増殖抑制作用及びアポトーシス誘導により生じるDNAフラグメントの検出を行った.その結果,40%エタノール溶出画分に小球状のアポトーシス小体が観察され,さらにアポトーシス誘導により生じるDNAの断片化を示した.また,40%エタノール溶出画分によるアポトーシス誘導についてDNA断片化の濃度及び培養時間依存性に関して検討した.その結果,アポトーシス誘導は濃度及び培養時間依存的であることが判明した.また,40%エタノール溶出画分によるヒト正常細胞に対する影響は観察されなかった.以上より,40%エタノール溶出物による抗腫瘍活性機構にはアポトーシス誘導が関与していることが示唆された.

A new enzyme, D-tagatose 3-epimerase, was found in Pseudomonas sp. ST-24 during the course of studies on D-sorbose fermentation. This new enzyme catalyzes epimerization of keto-sugars, for example between D-tagatose and D-sorbose, and between D-fructose and D-psicose. It was shown that this enzyme epimerizes the configuration at the C-3 position of these substrates. This epimerase didn't act on D-fructose 6-phosphate and D-ribulose 5-phosphate. The enzyme has been purified from cells grown on a medium containing 1% D-glucose and 0.05% D-tagatose, and it appeared homogeneous on electrophoresis. The enzyme has a molecular weight of about 68, 000 by gel filtration and consists of two subunits identical in molecular weight (about 33, 000 by SDS-PAGE). The maximum activity at 30°C was obtained at pH 7-9, and the enzyme was stable from pH 7-11. The optimum temperature was around 60°C, and it was stable up to 60°C for 10 min.

4種の従属栄養培養および独立栄養培養によって得たE. gracilis細胞の凍結乾燥品をペプシンとトリプシンによる人工消化実験に用いた.E. gracilis細胞はすべて高い人工消化率を示し,特に光照射下に従属栄養で培養したGreen株(GL細胞)は両酵素による高い消化率を示した.GL細胞を主とし,ネズミ飼育試験でユーグレナ・タンパク質の栄養価を求めたところ,真の消化率で僅かに劣ったが,PER,生物価,NPUにおいてGL細胞はカゼインに比肩しうる値を示し,すぐれた栄養価を持つタンパク質であることがわかった.熱エタノール処理したGL細胞は栄養価低く,貯蔵多糖をあらかじめ除去したGL細胞は無処理のGL細胞と変らない栄養価を示した.光照射下培養したBleached株細胞はGL細胞よりやや低い生物価を示した. ネズミ飼育試験は大阪市立環境科学研究所神戸保氏の,またューグレナ大量培養は当研究室中野長久博士の協力と助言を得た.厚く感謝する.本研究の費用の一部は文部省科学研究費に依った.記して謝意を表する.

重原子系電子状態計算手法の一つとして,1成分L-S基底を多電子配置関数として用いるスピン軌道配置間相互作用法(SOCI法)を開発・応用してきた.特に計算効率を高くするには,群論を効果的に用いることが重要である.まずU (2n)群の生成演算子の結合定数をGUGA法によるセグメント積で表現した.さらに偶数電子系に対しては,通常のMs固有関数形のスピン関数の代わりに,時間反転操作に対して不変である「実数スピン関数」を用いることにより,SO相互作用を含むハミルトニアン行列を実対称行列に変換し,さらに二重群の意味で特定の既約表現ブロックだけの計算を可能とした.奇数電子系においても,計算効率を維持したまま,直接法による大規模SOCI計算を可能とする方法を用いている.

Xylitol is widely used as a sweetener in foods and medications. Xylitol ingestion causes a small blood glucose rise, and it is commonly used as an alternative to high-energy supplements in diabetics. In previous studies, a xylitol metabolite, xylulose-5-phosphate, was shown to activate carbohydrate response element binding protein, and to promote lipogenic enzyme gene transcription in vitro; however, the effects of xylitol in vivo are not understood. Here we investigated the effects of dietary xylitol on lipid metabolism and visceral fat accumulation in rats fed a high-fat diet. Sprague-Dawley rats were fed a high-fat diet containing 0 g (control), 1.0 g/100 kcal (X1) or 2.0 g/100 kcal (X2) of xylitol. After the 8-week feeding period, visceral fat mass and plasma insulin and lipid concentrations were significantly lower in xylitol-fed rats than those in high-fat diet rats. Gene expression levels of ChREBP and lipogenic enzymes were higher, whereas the expression of sterol regulatory-element binding protein 1c was lower and fatty acid oxidation-related genes were significantly higher in the liver of xylitol-fed rats as compared with high-fat diet rats. In conclusion, intake of xylitol may be beneficial in preventing the development of obesity and metabolic abnormalities in rats with diet-induced obesity.

In this study, a simple and sensitive method for the determination of arsenic in water samples was developed. The method is based on the formation of micro particles of Ethyl Violet and molybdoarsenate, which gives an apparently homogeneous blue color to the solution. The absorption of the excess dye gradually decreases due to its conversion to a colorless carbinol species under strongly acidic conditions. Consequently, the sufficiently low reagent blank enables the spectrophotometric determination of arsenic with the detection limit of 4 µg l-1. The coefficient of variation for the spectrophotometry at 50 µg l-1 was 3.5% (n = 5). Furthermore, it is possible to detect concentrations as low as 10 µg l-1 of arsenic visually. Our method will be useful as a simple, rapid, and cost-effective field test of arsenic, requiring no complex apparatus or skilled laboratory support.

Rhenium- or manganese-catalyzed transformations based on the cleavage of inactive bonds, such as carbon–hydrogen and carbon–carbon bonds are discussed. Such transformations are recognized as key methods to realize efficient and powerful reactions. We also reveal novel regio- and/or stereodefined cycloaddition reactions using a rhenium or manganese catalyst. Most of the catalytic activities are typical of rhenium and manganese carbonyl complexes.

食品中の放射能を高速に測定する装置を開発中である。従来は、検出効率の低さから測定精度の確保に多大な測定時間を要していた。測定の高速化には、アンフォールディング手法を適用するが、測定値の信頼性を確保するには、測定ばらつきによる誤差を抑制する必要がある。そこで、品質工学を用いて安定な出力が得られる装置を設計した。

Physical exercise-mediated production of reactive oxygen species has been shown to cause oxidative stress, particularly in contracting skeletal muscles. Growing evidence indicates that exercise-induced oxidative stress plays an indispensable role in upregulating signaling pathways required to promote not only skeletal muscle, but also whole body adaptation to physical exercise. It is becoming increasingly clear that exercise-related beneficial adaptations are strongly regulated by exercise-induced oxidative stress, consistent with hormesis theory. According to the hormesis hypothesis, exercise-induced mild to moderate oxidative stress through reactive oxygen species generation stimulates favorable exercise-related physiological adaptations. Additionally, repeated exposure to oxidative stress induced by physical training can trigger various hormesis-based adaptations (i.e., hormetic adaptive responses), including activation of antioxidative defense mechanisms. This brief review provides an overview of several conceptual frameworks related to exercise-mediated hormetic adaptive responses rather than a detailed critique of individual reports.

The effect of tetrapod-shaped alpha tricalcium phosphate granules (Tetrabones® [TB]) in combination with basic fibroblast growth factor (bFGF)-binding ion complex gel (f-IC gel) on bone defect repair was examined. Bilateral segmental defects 20-mm long were created in the radius of 5 dogs, stabilized with a plate and screws, and implanted with 1 of the following: TB (TB group), TB and bFGF solution (TB/f group), and TB and f-IC gel (TB/f-IC group). Dogs were euthanized 4 weeks after surgery. Radiographs showed well-placed TB granules in the defects and equal osseous callus formation in all the groups. Histomorphometry revealed that the number of vessels and volume of new bone in the TB/f-IC group were significantly higher than those in the other groups. However, no significant differences in neovascularization and new bone formation were observed between the TB/f and TB groups. Furthermore, no significant difference in the lamellar bone volume or rate of mineral apposition was observed among groups. These results suggest that increased bone formation might have been because of the promotion of neovascularization by the f-IC gel. Therefore, the combinatorial method may provide a suitable scaffold for bone regeneration in large segmental long bone defects.
