Rice is an important staple crop and fungal blast disease destroys about 10–30% of its global produce, annually. Although genetic manipulation has largely been employed in crop-improvement programmes and agricultural biotechnology, the ease of transformation of several recalcitrant indica cultivars continues to be a challenge. HR-12 and CO-39 are two indica cultivars that are commonly used in breeding programmes, but are susceptible to biotic threats like fungal blast and sheath blight disease. Here in this study, we have optimised a rapid and reproducible transformation protocol for the said cultivars, having compared both the tissue-culture and in-planta methods of transformation. Murashige & Skoog basal media supplemented with maltose and 2.5 mg l−1 2,4-D induced efficient callogenesis in HR-12, while maltose with 3 mg l−1 2,4-D gave optimum results in case of CO-39. The media containing 0.5 mg l−1 NAA, 3 mg l−1 BAP, and 1 mg l−1 kinetin yielded a maximum regeneration efficiency of 62% and 65% in HR-12 and CO-39, respectively. The studies with Agrobacterium tumefaciens, LBA4404 strain harbouring pCAMBIA1303 suggested that although these cultivars demonstrated successful gene-transfer, they failed to regenerate efficiently, post-transformation. Alternatively, our modified in-planta piercing and vacuum infiltration-based protocol resulted in 33–35% transformation efficiency in less than half the time required for tissue-culture based transformation method. As per our knowledge, it is among the highest obtained from existing piercing-based direct transformation protocols in rice, and can also be implemented in genetically manipulating other recalcitrant varieties of rice.

The commercial use of genetically modified (GM) crops requires prior assessment of the risks to the environment when these crops are grown in the field or distributed. Assessments protocols vary across countries and GM crop events, but there is a common need to assess environmental biosafety. In this study, we conducted an environmental risk assessment in a confined field of GM tomato plants that can produce miraculin, a taste-altering protein that causes sour tastes to be perceived as sweet, for practical use in Japan. The evaluation was conducted for 1) competitiveness (the ability to compete with wild plants for nutrients, sunlight, and growing areas and prevent their growth) and 2) the production of toxic substances (the ability to produce substances that interfere with the habitat and growth of wild plants, animals, and microorganisms). Investigations of plant morphology and growth characteristics as well as tolerance to low temperature during early growth and overwintering for assessment endpoints related to competitiveness showed no biologically meaningful difference between GM tomato and non-GM tomato. In addition, harmful substances in plant residues and root secretions were assessed by the plow-in method, succeeding crop test and soil microflora tests, and it was determined that GM tomato does not exhibit an increase in harmful substances. Based on these results, it was concluded that GM miraculin-accumulating tomato is comparable to conventional tomato and is unlikely to have unintended adverse effects in the natural environment of Japan.

A NAC domain transcription factor, VND-INTERACTING2 (VNI2) is originally isolated as an interacting protein with another NAC domain transcription factor, VASCULAR-RELATED NAC-DOMAIN7 (VND7), a master regulator of xylem vessel element differentiation. VND7 directly or indirectly induces expression of a number of genes associated with xylem vessel element differentiation, while VNI2 inhibits the transcriptional activation activities of VND7 by forming a protein complex. VNI2 is expressed at an earlier stage of xylem vessel element differentiation than VND7. Here, to investigate whether VND7 also affects VNI2, a transient expression assay was performed. We demonstrated that VND7 downregulated VNI2 expression. Other transcription factors involved in xylem vessel formation did not show the negative regulation of VNI2 expression. Rather, MYB83, a downstream target of VND7, upregulated VNI2 expression. By using the deletion series of the VNI2 promoter, a 400 bp region was identified as being responsible for downregulation by VND7. These data suggested that VND7 and VNI2 mutually regulate each other, and VNI2 expression is both positively and negatively regulated in the transcriptional cascade.

Dalbergia congestiflora Pittier is a woody plant species grown in Mexico and Central America and widely used as timber wood and medicinal material. Since D. congestiflora is an endangered species, an in-vitro micropropagation technique is needed for mass propagation of D. congestiflora plantlets. Nodal segments of D. congestiflora stem cuttings grown in greenhouse conditions were disinfected with an appropriate protocol and in vitro established on Murashige and Skoog medium (MS) supplemented with 0.05 mg l−1 benzylaminopurine (BA). The explants showed 10% contamination with 90% survival, and the initial shoot was regenerated in 90% of them. Axillary buds of 45-day-old initial shoots were cultured on MS containing BA (0, 0.05, 0.1, 0.5, 1, 1.5 and 2 mg l−1) singly or in combination with α-naphthaleneacetic acid (NAA) (0, 0.1, 0.5 and 1 mg l−1). A higher shoot number (9.6 shoots/explant) was obtained on MS with 1 mg l−1 BA and 0.1 mg l−1 NAA. Rooting was investigated using half-strength MS, 2% sucrose and different concentrations of indole butyric acid (IBA) (0, 0.1, 0.5 and 1 mg l−1). After 30 days of culture, developing shoots were elongated and rooted in culture medium without IBA, with production of 3.2 roots/shoot. Micropropagated plantlets of D. congestiflora were successfully transplanted and acclimatized to a mixture of peat moss and perlite (2 : 1) with 100% relative humidity in greenhouse conditions with 80% survival at 30 days of culture. This micropropagation protocol will contribute to the conservation of D. congestiflora, and assure the mass propagation for sustainable usage of this species.

Potato (Solanum tuberosum L.) is a major global food crop. Contemporary potato production largely utilizes micropropagation to produce healthy seed potatoes. The micropropagation of potatoes is widely achieved through nodal explants using the conventional Murashige and Skoog (MS) medium. Currently, effective culture media that can facilitate rapid propagation are increasingly required for new cultivars that have been developed to possess improved traits. In this study, we evaluated the effect of enhanced meso nutrients (CaCl2.2H2O, MgSO4, and KH2PO4) in MS medium on the growth of S. tuberosum. The cultivars used in this study were representative of Japanese, European, and Peruvian lines. Enhanced meso nutrients improved the overall quality of all cultivars, as indicated by longer shoots and larger leaves with dark color, compared with MS medium only. Shoots grown on enhanced mesos were approximately 1.5 times longer than on MS medium. Quantitative ion analysis revealed that plantlets with improved shoot length and leaf quality in most cultivars had increased calcium, magnesium, potassium, and phosphorus uptake than plantlets on MS medium. The results suggest that the reduced iron uptake on 3.0×MS, compared with 2.0× or 2.5×MS mesos, reduced plant growth. This study revealed for the first time that mesos concentrations higher than MS medium concentrations, complemented by enhanced calcium, magnesium, potassium, phosphorus, and iron uptake, play a significant role in improving the in vitro growth of potato.

Objective To investigate the risk factors for the development of Pneumocystis jirovecii pneumonia (PCP) in patients with rheumatoid arthritis (RA) undergoing methotrexate (MTX) therapy. Methods This single-center retrospective cohort study included consecutive patients with RA who received MTX for at least one year. The study population was divided into PCP and non-PCP groups, depending on the development of PCP, and their characteristics were compared. We excluded patients who received biologic disease-modifying anti-rheumatic drugs (DMARDs), Janus kinase inhibitors, and anti-PCP drugs for prophylaxis. Results Thirteen patients developed PCP, and 333 did not develop PCP. At the initiation of MTX therapy, the PCP group had lower serum albumin levels, a higher frequency of pulmonary disease and administration of DMARDs, and received a higher dosage of prednisolone (PSL) than the non-PCP group. A multivariate Cox regression analysis revealed that the concomitant use of PSL [hazard ratio (HR) 5.50, p=0.003], other DMARDs (HR 5.98, p=0.002), and serum albumin <3.5 mg/dL (HR 4.30, p=0.01) were risk factors for the development of PCP during MTX therapy. Patients with these risk factors had a significantly higher cumulative probability of developing PCP than patients who lacked these risk factors. Conclusion Clinicians should pay close attention to patients with RA who possess risk factors for the development of PCP during MTX therapy.

Objective Peripherally inserted central catheters (PICCs) are widely used in patients with hematologic malignancies. However, the risks of PICC-related complications during chemotherapy for acute myeloid leukemia (AML) are not fully understood. Methods We conducted a retrospective review of 128 adult patients with AML who received induction therapy by way of PICC insertion between 2012 and 2019. Results The median duration of PICC insertion was 30 days. The incidence rate of catheter-related bloodstream infection (CRBSI) was 2.4% at 30 days, and women were more likely to suffer from CRBSI than men. Local reactions at the insertion site were observed in 56 patients; however, these events did not predict CRBSI. The incidence rates of catheter-related thrombosis (CRT) were 1.6% at 30 days. Obesity put patients at an increased risk for CRT. Unexpected PICC removal occurred in 59 patients, and women were at a higher risk of catheter removal than men. Conclusion Low PICC-related complication rates, possibly associated with high rates of catheter removal, were observed during intensive chemotherapy for AML. Women and obese patients require careful monitoring of their PICC. Procedures to achieve appropriate PICC removal without increasing the complication rate need to be considered.

In this study, we provide a time-dependent mechanical model, taking advantage of molecular dynamics simulations, quasiharmonic analysis of molecular dynamics trajectories, and time-dependent linear response theories to describe vibrational energy redistribution within the protein matrix. The theoretical description explained the observed biphasic responses of specific residues in myoglobin to CO-photolysis and photoexcitation on heme. The fast responses were found to be triggered by impulsive forces and propagated mainly by principal modes <40 cm−1. The predicted fast responses for individual atoms were then used to study signal propagation within the protein matrix and signals were found to propagate ~8 times faster across helices (4076 m/s) than within the helices, suggesting the importance of tertiary packing in the sensitivity of proteins to external perturbations. We further developed a method to integrate multiple intramolecular signal pathways and discover frequent “communicators”. These communicators were found to be evolutionarily conserved including those distant from the heme.

We investigated sex ratio in seven populations of the Japanese pond turtle, Mauremys japonica, inhabiting ponds in the central region of Aichi Prefecture, Japan. The sex ratio, calculated as a numerical proportion (%) of males to total individuals sexed, was highly variable among the populations, ranging from no more than 20.0% to no less than 92.9%. Since M. japonica shows a temperature-dependent sex determination (TSD) with eggs producing more males under a relatively low incubation temperature and more females under a higher temperature, we hypothesized that the observed remarkable inter-population variation in sex ratio reflects that in the nest temperature. Our field observations indicated that the nesting sites of male-biased populations were largely shaded by wood, thus supposedly under a relatively low ambient temperature, while those of female-biased populations were located in open environments with direct exposure to solar heat. These results offer a circumstantial support to the above hypothesis. Remarkably skewed sex ratios observed in some populations might reflect the effect of substantial habitat fragmentation and severe nesting site limitation caused by recent artificial land development and changes in agricultural land use. If this is actually the case and the effect continues to operate, many of the M. japonica populations may eventually disappear.

On 26 April 1999, three persons were accidentally exposed by high dose 60Co irradiation. They suffered from severe (one case) or moderate (two cases) hemopoietic form of acute radiation sickness (ARS). As part of the comprehensive treatment, strict reverse isolation and granulocyte-macrophage colony-stimulating factor (GM-CSF) therapy were applied. All the patients recovered after an appropriate treatment for 83 days. In our experience, the correct diagnosis and effective treatment at an early stage proved to be helpful to the patients in pulling out from the critical stage of acute radiation sickness. To avoid menstruation by the female patient just in the critical stage, we modified her menstruation cycle by testosterone. In our view, GM-CSF should be given as early as possible with enough dosage for promoting early hematological reconstruction. The experience obtained from the medical management of these patients is valuable for the treatment of such patients in the future.

サイクリック系3段ADCを用いた画素サイズ1.1μmの3,300万画素240枚/秒3次元積層構造CMOSイメージセンサを開発した.裏面照射型で3次元積層構造を,ハイブリッドスタッキング技術を用いることで,画素部とアレイ状に配置したAD変換器を画素エリア内部で接続した.3段パイプラインのサイクリック-サイクリック-逐次比較AD変換器により,変換時間周期を0.92μsに高速化した.3段AD変換器の構成とハイブリッドスタッキング技術により,3,300万画素において240枚/秒の高フレームレートを初めて実現した.画素速度7.96ギガ画素/秒の高速読み出しを実現しつつ,ランダムノイズ3.6電子とセンサ消費電力3.0ワットを達成した.

[Purpose] To investigate whether thoracic spine mobilization added to stabilization exercises increases the muscular strength and range of motion of the thoracic vertebrae of chronic low-back pain patients. [Subjects] This study enrolled 20 patients with chronic low back pain, who were divided into two groups. Ten subjects were randomly selected for the stabilization exercise group and the remaining 10 subjects received thoracic spine mobilization in addition to performing the stabilization exercises. [Methods] The patients performed stabilization exercises and received thoracic spine mobilization for 12 weeks. The range of motion and isometric muscular strength of the vertebrae of all subjects were measured before and after the intervention. [Results] In the comparison of muscular strength before and after the intervention, the change in muscular strength of the trunk flexors in the stabilization exercise group was 16.0±7.4 Nm, and that of the thoracic spine mobilization group was 34.2±7.6 Nm, a significant difference in each group. In the post-intervention intergroup comparison, the muscular strength of trunk flexors in the stabilization exercise group was 111.1±16.9 Nm, while that of the thoracic spine mobilization group was 125.9±11.3 Nm, a significant difference. Also, the muscular strength of the trunk extensors in the stabilization exercise group was 148.9±31.8 Nm, while that of the thoracic spine mobilization group was 182.9±37.2 Nm, a significant difference. The thoracic spine flexion in the stabilization exercise group was 29.8±9 degrees, while that of the thoracic spine mobilization group was 38.7±6.9 degrees, a significant difference. However, there was no significant difference in lumbar flexion values between the two groups. [Conclusion] Thoracic spine mobilization added to a stabilization exercise increased the muscular strength of patients with chronic low back pain.

本論文では,日本統治時代における台湾の日本人住宅・宿舎の庭園の保存・修復の今後に資することを目的に,残存事例が台湾でも数多い台北の日本人住宅・宿舎の庭園について調査した。その結果,以下のことを明らかにした。庭園規模は,建物面積の2~7倍が理想とされ,現地調査では多くの庭園が2倍程度であり,平均では2.2倍であったこと。庭園配置は,台北市内の風向きと住宅への通風が考慮された結果,南北方向,あるいは南西・北東方向に前庭・主庭を配置する傾向があったこと。庭園構成は,芝生が太陽の輻射熱を緩和し,植栽は建物の壁体への直射日光を緩和する役割を担い,ベランダ,テラス,パーゴラが防暑のための特徴的な施設であったこと。外囲いは住宅敷地内の通風を考慮して生垣が推奨されたこと。床下通風の観点から床は高床に設定され,沓脱には自然石ではなく人造石が多用されたこと。

Retrograde long waves in the higher latitudes of Northern Hemisphere can episodically attain large amplitudes and sustain coherent phase propagation for 2-3 weeks. The potential influence of such waves on extended-range weather forecast has been conjectured but not systematically quantified. Using a set of ensemble reforecast data, this study examines the predictability associated with an extraordinary retrograde-wave episode in the 1979-80 winter. Quantified by the anomaly correlation of 500 hPa geopotential height in the 40°N-70°N latitudinal band, increased week-2 predictability is found within the sub-period with the presence of coherent retrograde waves. Some individual forecasts made within the retrograde-wave event exhibit the behavior of “return of skills”. The results suggest a future investigation into the relation between the uptick of anomaly correlation in week-2 and detailed dynamics of the retrograde waves.

There is a need to develop ITER instrumentation and control (I&C) systems with high reliabilities. Interlock systems that activate machine protection functions are implemented on robust wired-logic systems such as programmable logic controllers (PLCs). We herein propose a software tool that generates program code templates for the control systems using PLC logic. This tool decreases careless mistakes by developers and increases reliability of the program codes. A large-scale engineering database has been implemented in the ITER project. To derive useful information from this database, we propose adding semantic data to it using the Resource Description Framework format. In our novel proposal for the ITER diagnostic control system, a guide words generator that analyzes the engineering data by inference is applied to the hazard and operability study. We validated the methods proposed in this paper by applying them to the preliminary design for the I&C system of the ITER edge Thomson scattering system.
