α-Ketoglutarate (α-KG) also known as 2-oxoglutarate (2-OG) is an intermediate metabolite in the tricarboxylic acid (TCA) cycle and is also produced by the deamination of glutamate. It is an indispensable cofactor for a series of 2-oxoglutarate-dependent oxygenases including epigenetic modifiers such as ten-eleven translocation DNA demethylases (TETs) and JmjC domain-containing histone demethylases (JMJDs). Since these epigenetic enzymes target genomic DNA and histone in the nucleus, the nuclear concentration of α-KG would affect the levels of transcription by modulating the activity of the epigenetic enzymes. Thus, it is of great interest to measure the nuclear concentration of α-KG to elucidate the regulatory mechanism of these enzymes. Here, we report a novel fluorescence resonance energy transfer (FRET)-based biosensor with multiple nuclear localization signals (NLSs) to measure the nuclear concentration of α-KG. The probe contains the α-KG-binding GAF domain of NifA protein from Azotobacter vinelandii fused with EYFP and ECFP. Treatment of 3T3-L1 preadipocytes expressing this probe with either dimethyl-2-oxoglutarate (dimethyl-2-OG), a cell-permeable 2-OG derivative, or citrate elicited time- and dose-dependent changes in the FRET ratio, proving that this probe functions as an α-KG sensor. Measurement of the nuclear α-KG levels in the 3T3-L1 cells stably expressing the probe during adipocyte differentiation revealed that the nuclear concentration of α-KG increased in the early stage of differentiation and remained high thereafter. Thus, this nuclear-localized α-KG probe is a powerful tool for real-time monitoring of α-KG concentrations with subcellular resolution in living cells and is useful for elucidating the regulatory mechanisms of epigenetic enzymes.

<p>With urgent demand of elderly care in Taiwan, family as major care provider has confronted with various challenges in the process of social changes. This study intends to explore attitudes toward elderly parental support by focusing on the relative influence of individual resources, cultural norms and family care demand. Drawing from the Taiwan Social Change Survey, parental support is indicated by "living arrangement" and "financial support" which is further delineated by "from parent's perspective" versus "from children's perspective." We first present the trend of changing attitudes on parental support from 1991 to 2016. A consistent picture shows that parental autonomy in living arrangement and in living expenses has been a preferred type from the parent's perspective, while dependence on children, especially on sons, is considered a better choice from the children's viewpoints. The impact of filial norm is suspected to affect children's stand and thus produces the opposing patterns. Multivariate analyses point out cultural norms having the most salient effects, followed by individual resources and much less by care demand at home. Filial piety contributes to the expected attitudes on co-residence between generations and financial provision to parents from both perspectives. Future studies need to examine the emerging change on gender norms in elderly parental support.</p>

Background: About 50% of lung cancer deaths in Taiwan are not related to cigarette smoking. Environmental exposure may play a role in lung cancer risk. Taiwanese households frequently burn mosquito coil at home to repel mosquitoes. The aim of this hospital-based case-control study was to determine whether exposure to mosquito coil smoke is a risk for lung cancer.Methods: Questionnaires were administered to 147 primary lung cancer patients and 400 potential controls to ascertain demographic data, occupation, lifestyle data, indoor environmental exposures (including habits of cigarette smoking, cooking methods, incense burning at home, and exposure to mosquito coil smoke ), as well as family history of cancer and detailed medical history.Results: Mosquito coil smoke exposure was more frequent in lung cancer patients than controls (38.1% vs.17.8%; p<0.01). Risk of lung cancer was significantly higher in frequent burners of mosquito coils (more than 3 times [days] per week) than nonburners (adjusted odds ratio = 3.78; 95% confidence interval: 1.55-6.90). Those who seldom burned mosquito coils (less than 3 times per week) also had a significantly higher risk of lung cancer (adjusted odds ratio = 2.67; 95% confidence interval: 1.60-4.50).Conclusion: Exposure to mosquito coil smoke may be a risk factor for development of lung cancer.

Threshhold dose for organoleptic changes by gamma irradiation in shell eggs is at 0.5 kGy, whereas frozen egg products is at 2.4 kGy and dehydrated egg products is at 3 kGy respectively. For the elimination of Salmonella, necessary dose is estimated to be 1 kGy for shell eggs and to be 2 kGy for dehydrated egg products from survival fraction of S. Typhimurium or S. Enteritidis. Contamination of Salmonella occur significantly in liquid egg products and for the reason of threshhold dose of organoleptic changes, irradiation treatment should be applied to dehydrated condition with 2-3 kGy.

[Purpose] The purpose of this study was to examine the effects of hollowing and bracing exercises on cross-sectional areas of abdominal muscles. [Subjects] Thirty healthy female adults participated in this study. The exclusion criteria were orthopedic or neurologic diseases. [Methods] The subjects of this study were assigned randomly to one of two groups, each with 15 people. Each group performed a 60-minute exercise program, one performed a bracing exercise, and the other performed a hollowing exercise, with both groups performing the exercise three times a week for six weeks. [Results] The changes in cross-sectional areas after the bracing exercise showed statistically significant differences in the left rectus abdominis and both internal and external obliques. The changes in cross-sectional areas after the hollowing exercise showed statistically significant differences in the left and right transversus abdominis and left rectus abdominis. [Conclusion] Performing bracing exercises rather than hollowing exercises is more effective for activating the abdominal muscles.

Torrential rainfall associated with linear precipitation systems occurred in Northern Kyushu, Japan, during July 5–6, 2017, causing severe damage in Fukuoka and Oita Prefectures. According to our statistical survey using ground rain gauges, the torrential rainfall was among the heaviest in recorded history for 6- and 12-h accumulated rainfall, and was unusual because heavy rain continued locally for nine hours. The predictability of precipitation associated with linear precipitation systems for this event was investigated using a cloud-resolving numerical weather prediction model with a horizontal grid interval of 1 km. The development of multiple linear precipitation systems was predicted in experiments whose initial calculation time was from several hours to immediately before the torrential rain (9:00, 10:00, 11:00, and 12:00 Japan Standard Time on July 5), although there were some displacement errors in the predicted linear precipitation systems. However, the stationary linear precipitation systems were not properly predicted. The predictions showed that the linear precipitation systems formed one after another and moved eastwards. In the relatively accurate prediction whose initial time was 12:00 on July 5, immediately before the torrential rainfall began, the forecast accuracy was evaluated using the 6-h accumulated precipitation (P6h) from 12:00 to 18:00 on July 5, the period of the heaviest rainfall. The average of the P6h in an area 100 km×40 km around the torrential rainfall area was nearly the same for the analysis and the prediction, indicating that the total precipitation amount around the torrential rainfall area was predictable. The result of evaluating the quantitative prediction accuracy using the Fractions Skill Score (FSS) indicated that a difference in location of 25 km (50 km) or greater should be allowed for in the models to produce useful predictions (those defined as having an FSS ≥0.5) for the accumulated rainfall of P6h ≥50 mm (150 mm). The quantitative prediction accuracy examined in this study can be basic information to investigate the usage of predicted precipitation data.

Pituitary adenomas are benign tumours that can cause an individual various clinical manifestations including tumour mass effects and/or the diverse effects of abnormal pituitary hormone secretion. Given the morbidity and limited treatment options for pituitary adenomas, there is a need for better biomarkers and treatment options. One molecule that is of specific interest is the signal transducer and activator of transcription 3 (STAT3), a transcription factor that plays a critical role in mediating cytokine-induced changes in gene expression. In addition, STAT3 controls cell proliferation by regulating mitochondrial activity. Not only does activation of STAT3 play a crucial role in tumorigenesis, including pituitary tumorigenesis, but a number of studies also demonstrate pharmacological STAT3 inhibition as a promising treatment approach for many types of tumours, including pituitary tumours. This review will focus on the role of STAT3 in different pituitary adenomas, in particular, growth hormone-producing adenomas and null cell adenomas. Furthermore, how STAT3 is involved in the cell proliferation and hormone regulation in pituitary adenomas and its potential role as a molecular therapeutic target in pituitary adenomas will be summarized.

Background:The COVID-19 pandemic has challenged healthcare systems, at times overwhelming intensive care units (ICUs). We aimed to describe the length and rate of ICU admission, and explore the clinical variables influencing ICU use, for COVID-19 patients with known cardiovascular diseases or their risk factors (CVDRF).Methods and Results:A post hoc analysis was performed of 693 Japanese COVID-19 patients with CVDRF enrolled in the nationwide CLAVIS-COVID registration system between January and May 2020 (mean [±SD] age 68.3±14.9 years; 35% female); 199 patients (28.7%) required ICU management. The mean (±SD) ICU length of stay (LOS) was 19.3±18.5 days, and the rate of in-hospital death and hospital LOS were significantly higher (P<0.001) and longer (P<0.001), respectively, in the ICU than non-ICU group. Logistic regression analysis revealed that clinical variables reflecting impaired general condition (e.g., high C-reactive protein, low Glasgow Coma Scale score, SpO2, albumin level), male sex, and previous use of β-blockers) were associated with ICU admission (all P<0.001). Notably, age was inversely associated with ICU admission, and this was particularly prominent among elderly patients (OR 0.97, 95% confidence interval 0.95–0.99; P=0.0018).Conclusions:One-third of COVID patients with CVDRF required ICU care during the first phase of the pandemic in Japan. Other than anticipated clinical variables, such as hypoxia and altered mental status, age was inversely associated with the use of the ICU, warranting further investigation.

Epithelial–mesenchymal transition (EMT) plays a crucial role in metastasis of epithelial tumors; however, it is challenging to detect EMT by cytology. In the present study, EMT was visualized by fluorescence-immunocytochemistry (FICC). Air-dried smears from epithelial tumors of dogs (n=22) and cats (n=9) were stained using mouse monoclonal anti-E-cadherin and rabbit monoclonal anti‐vimentin antibodies. Enzymatic immunohistochemistry (IHC) revealed that 51.6% (8/22 in dogs, 8/9 in cats) of the cases showed EMT. In dogs, FICC could detect EMT in 62.5% (5/8) of those cases. In cats, FICC could detect EMT in 100% (8/8) of the cases. In conclusion, the present FICC method could successfully detect EMT using conventional air-dried cytology smear slides.

The size of canine focal liver lesions (FLLs) is known to be one of the predicting criteria for malignancy. However, there are discrepancies for the measurement of maximum lesion size, resulting in contradicting results among studies and incidences of false positive outcomes. Thus far, the morphometric changes of FLLs for distinguishing malignancy from benignancy remains undocumented. This study aimed to investigate morphometric characteristics of FLLs using computed tomography (CT). CT images of 40 dogs with histopathological confirmation of 49 liver lesions, including 39 hepatocellular carcinomas and 10 nodular hyperplasias were retrospectively reviewed. The morphometric parameters including size (long and short axis diameters measured on transverse image), shape (measured by long to short axis (L/S) ratio), volume, and surface appearance of a liver lesion were evaluated using univariate and stepwise multivariate analyses, respectively. The results of univariate analysis showed that long and short axis diameters, L/S ratio, volume, and surface appearance of a lesion were significantly different between hepatocellular carcinomas and nodular hyperplasias. Multivariate analysis revealed that short axis diameter (>3.30 cm; odds ratio (OR): 36.1, 95% confidence interval (CI): 3.36–387.05, P=0.0031) and L/S ratio (>1.23; OR: 18.1, 95% CI: 1.61–205.12, P=0.0191) were independent predictors of malignancy, with the area under the curve of 0.9154. These results suggest that the combination of short axis diameter and L/S ratio is a promising tool for predicting liver malignancy with outstanding discriminating ability.

Sitosterolemia is an inherited metabolic disorder characterized by increased levels of plant sterols, such as sitosterol. This disease is caused by loss-of-function genetic mutations in ATP-binding cassette (ABC) subfamily G member 5 or member 8 (ABCG5 or ABCG8, respectively), both of which play important roles in selective excretion of plant sterols from the liver and intestine, leading to failure to prevent absorption of food plant sterols. This disorder has been considered to be extremely rare. However, accumulated clinical data as well as genetics suggest the possibility of a much higher prevalence. Its clinical manifestations resemble those observed in patients with familial hypercholesterolemia (FH), including tendon xanthomas, hyper LDL-cholesterolemia, and premature coronary atherosclerosis. We provide an overview of this recessive genetic disease, diagnostic as well as therapeutic tips, and the latest diagnostic criteria in Japan.

Felis catus papillomavirus (FcaPV), especially type 2 (FcaPV2) is considered as one of the causative agents in squamous cell carcinoma (SCC) in cats. However, our previous study detected FcaPV3 and FcaPV4, but not FcaPV2 in feline SCCs collected in Japan, suggesting that the prevalence of FcaPV2 in SCC may vary depending on geographic locations. To evaluate this hypothesis, two conventional PCR reactions targeting E1 and E7 genes were performed to detect FcaPV2 in feline SCC samples collected in Taiwan and Japan. While 46.9% (23/49) of feline SCC cases from Taiwan were PCR positive for FcaPV2, only 8.6% (3/35) cases from Japan were positive. Our result suggests that the prevalence of FcaPV2 in feline SCCs may depend on the region.

Eisenman (2013) argues that the importance of design innovation increases in the early and late stages of an industry and is lowest in the middle stage. In response, this study empirically examines her proposition through an examination of mobile phones during the feature phone era (1999–2008) when various types of designs were being generated using design patent data. We analyze the number of mobile phone design patents registered before the current advent of smart phones as well as examining the yearly averages of forward citations (the number of times a particular design patent is cited in later design patents) and backward citations (the number of times a particular design patent cites previous design patents). Further, to shed light on interactions with an accumulation of design-related technology, the study also analyzes the number of design-related patent applications per year. Results reveal that while there was an increase in the number of design patents registered in the late stage of the industry, especially between the years of 2007–2008, the average number of forward citations drops below the average number of backward citations from 2003 onward. In other words, although at the late stages of the industry many design innovations emerged, most of these were incremental changes that followed past designs. In contrast, design-related patent applications reached a peak in 2004 and fell thereafter. It is hypothesized that this trend was influenced by the growing number of design-related patents that facilitated the easy generation of new variations in design. These results show the dilemma: Although design innovations continue to be generated over time by accumulating design-related technology, creating genuinely impactful design innovation becomes more difficult as time progresses. The study therefore demonstrates the difficulty of realizing Eisenman (2013)’s theoretical proposition.

Background:Coronavirus disease 2019 (COVID-19) reportedly causes venous thromboembolism (VTE), but the status of this complication in Japan was unclear.Methods and Results:The VTE and COVID-19 in Japan Study is a retrospective, multicenter cohort study enrolling hospitalized patients with COVID-19 who were evaluated with contrast-enhanced computed tomography (CT) examination at 22 centers in Japan between March 2020 and October 2020. Among 1,236 patients with COVID-19, 45 (3.6%) were evaluated with contrast-enhanced CT examination. VTE events occurred in 10 patients (22.2%), and the incidence of VTE in mild, moderate, and severe COVID-19 was 0%, 11.8%, and 40.0%, respectively. COVID-19 patients with VTE showed a higher body weight (81.6 vs. 64.0 kg, P=0.005) and body mass index (26.9 vs. 23.2 kg/m2, P=0.04), and a higher proportion had a severe status for COVID-19 compared with those without. There was no significant difference in the proportion of patients alive at discharge between patients with and without VTE (80.0% vs. 88.6%, P=0.48). Among 8 pulmonary embolism (PE) patients, all were low-risk PE.Conclusions:Among a relatively small number of patients undergoing contrast-enhanced CT examination in Japanese real-world clinical practice, there were no VTE patients among those with mild COVID-19, but the incidence of VTE seemed to be relatively high among severe COVID-19 patients, although all PE events were low-risk without significant effect on mortality risk.

Background:The impact of chronic kidney disease (CKD) on long-term outcomes following acute myocardial infarction (AMI) in the era of modern primary PCI with optimal medical therapy is still in debate.Methods and Results:A total of 3,281 patients with AMI were enrolled in the J-MINUET registry, with primary PCI of 93.1% in STEMI. CKD stage on admission was classified into: no CKD (eGFR ≥60 mL/min/1.73 m2); moderate CKD (60>eGFR≥30 mL/min/1.73 m2); and severe CKD (eGFR <30 mL/min/1.73 m2). While the primary endpoint was all-cause mortality, the secondary endpoint was major adverse cardiac events (MACE), defined as a composite of all-cause death, cardiac failure, myocardial infarction (MI) and stroke. Of the 3,281 patients, 1,878 had no CKD, 1,073 had moderate CKD and 330 had severe CKD. Pre-person-days age- and sex-adjusted in-hospital mortality significantly increased from 0.014% in no CKD through 0.042% in moderate CKD to 0.084% in severe CKD (P<0.0001). Three-year mortality and MACE significantly deteriorated from 5.09% and 15.8% in no CKD through 16.3% and 38.2% in moderate CKD to 36.7% and 57.9% in severe CKD, respectively (P<0.0001). C-index significantly increased from the basic model of 0.815 (0.788–0.841) to 0.831 (0.806–0.857), as well as 0.731 (0.708–0.755) to 0.740 (0.717–0.764) when adding CKD stage to the basic model in predicting 3-year mortality (P=0.013; net reclassification improvement [NRI] 0.486, P<0.0001) and MACE (P=0.046; NRI 0.331, P<0.0001) respectively.Conclusions:CKD remains a useful predictor of in-hospital and 3-year mortality as well as MACE after AMI in the modern PCI and optimal medical therapy era.

「六諭」及與其相關的道徳話語在中國明清時代的庶民教化政策中扮演著重要的角色,而且與宗族社會形成息息相關。在十八世紀初期,《六諭衍義》這一主要解釋「六諭」的書亦傳來日本,對日本徳川時代的庶民教化政策亦〓了相當重要的作用。該書被施于訓點在日本出版之外,以室鳩巣所注釋的《六諭衍義大意》爲首之許多「六諭」道徳話語的異本亦陸續被出版。故,本文首先従教育思想史的觀點來考察與「六諭」相關的諸文本是如何輸入日本且出版的。然而在中國明清時代使用的《六諭衍義》與在徳川時代主要被使用的《六諭衍義大意》一書的内容並不盡相同。所以,本文對《六諭衍義》與《六諭衍義大意》的内容進行比較,試圖探討《六諭衍義》與鳩巣所注釋的《六諭衍義大意》之内容差異與日中社會體係差異之間的關係。並且希望透過以上的檢討,來思考在東亞各國間之道徳話語的流傳與演變的這一問題。
