Adenosine 5’-triphosphate (ATP), the major energy currency of the cell, is involved in many cellular processes, including the viral life cycle, and can be used as an indicator of early signs of cytopathic effect (CPE). In this study, we demonstrated that CPE can be analyzed using an FRET-based ATP probe named ATP indicator based on Epsilon subunit for Analytical Measurements (ATeam). The results revealed that as early as 3 hr, the virus infected cells showed a significantly different Venus/cyan fluorescent protein (CFP) ratio compared to the mock-infected cells. The ATeam technology is therefore useful to determine the early signs of ATP-based CPE as early as 3 hr without morphology-based CPE by light microscopy, and enables high throughput determination of the presence of microorganisms in neglected samples stored in laboratories.

雄のネッタイイエカがチオテパで処理され, 1日あたり15万匹から30万匹が5カ月半にわたってデリー近郊の村に放逐された。村の周囲3km幅の地域にあるネッタイイエカ幼虫の生息場所は殺虫剤で処理され, 実験期間中成虫の発生を抑えた。放逐蚊はすべて螢光色素でマークされた。マーク雄と非マーク雄の比はつねに24 : 1より大きかった。不妊雄の交尾競争力がわかっているので, 村内では不妊雄による交尾率が非常に高いことが期待された。野生の雌をつかまえてそれに産ませた卵をしらべた結果, 最初の3週間に野外集団に高いレベルの不妊性をもたらしたことが証明された。しかしその後は不妊性のレベルは低かった。3kmの幼虫生息場所皆無帯は, 周辺からのすでに受精した雌の侵入をはばむことができなかったことを示すいくつかの証拠がある。実験対象とした村の野生のネッタイイエカの密度と比較対照とした別の二つの村の密度を検討すると, 高い不妊性を示した期間は実験対象とした村のネッタイイエカ密度は一時的に減少したように思われる。

A determination of forest characteristics across broad areas is of great concern to the forest industry in the southern United States, as timber supply decisions can be based on opportunities, or lack of thereof, across all wood procurement areas. This is important in areas such as the southern United States, where the land ownership distribution is highly fragmented and where no general comprehensive source of forest data exists other than the low-intensity USDA Forest Service FIA surveys. In an effort to describe forest characteristics along the lower Coastal Plain of the State of Georgia (USA), we utilized a time series of Landsat data and an algorithm that assesses an integrated forest Z score. The methodology was used to create disturbance maps for over 30 years that represent the year of disturbance for specific locations. The overall accuracy was 52% when all years were considered, and approximately 70% from 1991 forward. Preliminary findings showed moderate levels of accuracy when determining ages for current forests, most of which are even-aged nature stands. Further modifications to the process were necessary to adapt to the unique conditions of study region. The modeling process also prompted several areas for future refinement, including improvement of the temporal resolution of the analysis by using all the available Landsat imagery and detection of the regeneration that normally occurs several years after disturbances.

The autocorrelation of angle resolved photoemission data from the high temperature superconductor Bi2Sr2CaCu2O8+delta shows distinct peaks in momentum space which disperse with binding energy in the superconducting state, but not in the pseudogap phase. Although it is tempting to attribute a nondispersive behavior in momentum space to charge ordering, a deconstruction of the autocorrelation reveals that the nondispersive peaks arise from the tips of the Fermi arcs, which themselves do not change with binding energy.

Reelin is a secreted protein that antagonizes the deposition and toxicity of amyloid β peptide (Aβ). Therefore, augmentation of Reelin activity may ameliorate Alzheimer’s disease (AD). We have recently reported that a disintegrin and metalloproteinase with thrombospondin motifs 3 (ADAMTS-3) cleaves and inactivates Reelin in the mouse brain. In the present study, we investigated the effect of reducing ADAMTS-3 on deposition of Aβ by crossbreeding drug-inducible ADAMTS-3 conditional knock-out (cKO) mice with “next-generation” AD model mice. We found that reducing ADAMTS-3 inhibited deposition of Aβ significantly in AppNL-F mice, which produce human wild-type Aβ. On the other hand, reducing ADAMTS-3 had no effect in AppNL-G-F mice, which produce the Arctic mutant Aβ (E22G) that forms protofibrils more efficiently than does wild-type Aβ. Thus, the findings suggest that the administration of an inhibitor against ADAMTS-3 will prevent the progression of AD pathology caused by deposition of wild-type Aβ.

1999年9月10日に長野市内でツキノワグマ1個体(メス、成獣)を捕獲、お仕置き、放獣し、ラジオテレメトリーによる行動圏調査を開始した。同年12月25日現在、最外郭法で求めた行動圏は2.86km2となり、標高460m~890mの中山間地域(里山地域)に限られていた。環境選好性はアカマツ群落、落葉果樹園で高く、カラマツ植林で低かった。なお、捕獲場所の果樹園は行動圏には含まれていなかった。

野生のネズミ科動物の内部寄生虫については, 比較的多くの報告があるが, ヒマラヤ地方に生息するヒマラヤアカネズミApodemus gurkhaおよびシッキムハタネズミMicrotus sikimensisを宿主とする寄生虫の報告は皆無である。そこで, 1994年11月および1996年3月, ネパール・ヒマラヤ地方Myagdi districtにおいて採集されたヒマラヤアカネズミ14個体およびシッキムマツネズミ9個体について内部寄生蠕虫類の検査をした。その結果, ヒマラヤアカネズミからはHeligmosomoides neopolygyrus, Heligmonoides sp., Syphacia agraria, Heterakis spumosa, Rictularia cristataおよびCatenotaenia sp.が, また, シッキムハタネズミからはCarolinensis minutus, Syphacia montana, Aonchotheca murissylvatici, Trichuris sp., Anoplocephalidae gen.sp.およびTaeniidae gen.sp.(嚢尾虫)が検出された。今回検出された寄生虫のうち, Heligmonoides sp.についてはヒマラヤアカネズミに宿主特異的な新種である可能性が高いが, 他の種はユーラシア大陸の他地域に産するアカネズミ属あるいはハタネズミ属の寄生虫と共通であることが判明した。

Cardiovascular magnetic resonance is a well-established tool for the quantification of focal fibrosis. With the introduction of T1 mapping, diffuse myocardial processes can be detected and quantified. In particular, infiltration and storage disorders with large disease-related changes, and diffuse fibrosis where measurement is harder but the potential impact larger. This has added a new dimension to the understanding and assessment of various myocardial diseases. T1 mapping promises to detect early disease, quantify disease severity and provide prognostic insights into certain conditions. It also has the potential to be a robust surrogate marker in drug development trials to monitor therapeutic response and be a prognostic marker in certain diseases. T1 mapping is an evolving field and numerous factors currently preclude its standardization. In this review, we describe the current status of T1 mapping and its potential promises and pitfalls. Ⅰ.はじめに もやもや病は,両側内頚動脈終末部に慢性進行性の狭窄を生じ,代償的に脳底部に異常血管網が形成される原因不明の疾患である11).一方,川崎病は乳児および幼児において原因不明の系統的血管炎を主体とする疾患であり6),活動期に稀ながら脳梗塞を合併する4,7,12,15,17).川崎病活動期のもやもや病合併の報告はないが,川崎病罹患歴のあるもやもや病の報告がある1,3,8,9,13,14).今回,川崎病の既往があるもやもや病3例を経験したので,文献的考察を交えて報告する.

アフリカ豚コレラ(ASF)強毒ウイルスTengani株をVero細胞に馴化してから, そのまま継代培養した場合と, クローニングしてから更に高継代した場合の両方共, 病原性, 免疫原性, 及び感染性の異ったSubpopulationを含むウイルスが容易に産生された. 第27代継代ウイルスの99%以上は接種豚体内で増殖の出来ない非病原性ウイルスであって, 明らかに自然界に存在しない実験室に於ける人工産物であった. 新Subpopulationの病原性の減弱は, 全く均一に起きる訳ではなく, 弱毒化した第27代継代ウイルスを接種した豚個体内では, 血中に連続的に夫々病原性及び免疫原性の異るSubpopulationの出現を見, ある時期に採取した血液材料から強い病原性を有するウイルスが分離出来た. 最初にクローニングを行ってから23代継代培養したウイルスの99.9%は, 接種豚にSubclinicalな感染を起す弱い病原性を示すウイルスであったが, 耐過豚を親株であるTengani株で攻撃した所, 完全な感染防御を示さなかった. Tengani株ウイルス中には, Lisbon '60株と共通の免疫原性を有するSubpopulationと, Tengani株固有のSubpopulationを保有する事が明らかとなった.

In skeletal muscle, resting cytosolic Ca2+ concentration ([Ca2+]cyto) homeostasis is exquisitely regulated by Ca2+ transport across the sarcolemmal, mitochondrial and sarcoplasmic reticulum (SR) membranes. Specifically, skeletal muscle contractile function is critically dependent on effective SR Ca2+ handling. However, more recent studies have revealed that mitochondria are essentially involved in [Ca2+]cyto homeostasis during and following muscle contractions. We recently provided substantial support for the mitochondria as a major site for Ca2+ sequestration in skeletal muscle during recovery from fatiguing tetanic contractions under in vivo conditions. This review provides an overview of the role of skeletal muscle mitochondria in [Ca2+]cyto regulation in vivo during and following muscle contractions.

Background:Physical activity is associated with lower risk of coronary and cerebrovascular disease but its potential role in prevention of aortic valve stenosis (AVS) is unclear.Methods and Results:We investigated whether physical activity influences AVS risk in a cohort of 69,288 adults. During a mean follow-up of 15.3 years, 1,238 AVS cases were diagnosed. No associations were observed between AVS and walking/bicycling (≥1 h/day vs. almost never: hazard ratio 0.92, 95% CI 0.74–1.15) or exercise (≥4 hs/week vs. <1 h/week: hazard ratio 1.18, 95% CI 0.97–1.43).Conclusions:Physical activity did not reduce the incidence of AVS.

This paper presents a summary of some of the observational and numerical studies on the climatology and possible change mechanisms of the outer-core wind structure of a tropical cyclone (TC), which has been generally referred to as size, a term also to be used in this review although various definitions have been given in the literature. In all the ocean basins where TCs exist, TC size has been found to vary with season, year, decade, latitude and longitude. Such variations are related to those in the synoptic flow patterns in which the TCs are embedded. Several factors have been identified to be responsible for changes in TC size, which include environmental humidity, vortex structure, sea surface temperature and planetary vorticity. Each of these factors can modify the transport of lower tropospheric angular momentum into the TC and hence cause changes in its size. The paper ends with a discussion of outstanding issues in the study of the outer-core wind structure of a TC.

We have performed systematic angle-resolved photoemission spectroscopy (ARPES) on single-layered cuprate superconductor Bi2Sr2CuO6+delta to elucidate the origin of shadow band. We found that the shadow band is exactly the c(2×2) replica of the main band irrespective of the carrier concentration and its intensity is invariable with respect to temperature, doping, and substitution constituents of block layers. This result rules out the possibility of antiferromagnetic correlation and supports the structural origin of shadow band. ARPES experiments on optimally doped La1.85Sr0.15CuO4 also clarified the existence of the c(2×2) shadow band, demonstrating that the shadow band is not a unique feature of Bi-based cuprates. We conclude that the shadow band is related to the orthorhombic distortion at the crystal surface.
