Liquid chromatographic/(LC) analysis is an effective instrument for pharmaceutical analysisand pharmaceutical science. Various methods have been utilized in attempts to optimize LC analytical conditions. ln order to obtain a high degree of separation, the optimum conditions were determined by the use of dynamic S/N ratio as an index for the evaluation of the separation process. An L18 orthogonal array was used in this experiment. lt was demonstrated that S/N ratio was an appropriate scale to indicate the separation of some peaks in a LC. Also, the appropriate types of S/N ratio to show the degree of separation in a LC was discussed.

Generally, a liquid chromatography (LC) is widely used in the field of a chemical and a pharmaceutical analysis. On account of many advances of a modern technique of the instrumental analysis and the appearance of high resolution column, a chemical analysis and a quantitative analysis are done more accurately than before. However, it seems that the determination of the analytical condition is mostly performed empirically. ln development of a new drug research, LC is an indispensable tool for the analysis of the chemical components, and so it is important to get the analytical conditions which is able to performed high degree of the separation. This paper describes that the parameter design was conductedto optimize the analytical condition of a LC. The experiments were conducted on the L₁₈ orthogonal array. Evaluation was done by the S/N ratio and sensitivity in comparison withthe current condition. The optimum condition was determined from the control factor levels which gave the large values of a S/N ratio and sensitivity S. As a result, an improvement was estimated as 3.32db in a S/N ratio. lt seems that this method can be widely applicableto the determination of the analytical conditions of a LC.

We developed safe and simple closed system for preparing ointments containing cytotoxic drugs using a polyethylene bag.β-Carotene (β-C) and fluorescein sodium (FL) were used as model drugs in the present study. All preparation procedures were carried out in a polyethylene bag. The uniformity of drug content in the ointment was obtained by rolling the contents more than 20 times with a roller in both model drugs. The contents of each model drug in the ointment prepared by a closed system were consistent with those by the conventional method using a glass mortar. The time required to prepare the ointment in a closed system was much shorter than that using the conventional method. In addition, no contamination was observed in the closed system, whereas some contamination of the cyto-safe sheet and gloves with FL were observed with the conventional method. Based on the present findings, a closed system using a polyethylen bag is thus considered to be a useful method for preparing ointments containing cytotoxic drugs.

Kinetic analysis of thermal decomposition was carried out using thermogravimetry (TG) and differential scanning calorimetry (DSC). The linear equations of n order, phase-boundary, nucleation growth and diffusion-controlled reaction under linear heating conditions were derived for the estimation of the rate constant and the activation energy. The method to determine the reaction mechanism was shown by evaluation of linearity. The equations were applied to the TG and DSC curves of dehydration of calcium oxalate monohydrate and elimination of carbon monoxide from calcium oxalate. The dehydration was fit to two-dimensional phase-boundary reaction and the activation energy was calculated as 85 kJ/mol. But the dehydration at a high-speed heating rate proceeded as apparent three-dimensional diffusion-controlled reaction. The elimination of carbon monoxide from calcium oxalate was fit to phase-boundary reaction and the activation energy was calculated as 196 kJ/mol for three-dimensional phase-boundary reaction and 182 kJ/mol for two-dimensional phase-boundary reaction.

Compatibility of enteric-coated Futraful E Granules (FTEG) with 60 commercial preparations was investigated. In combination with FTEG, changes in appearance (wetness, coagulation, coloration and liquefaction) under the worst condition (30°, RH 92%) were observed in only 3 of the 60 preparations: neophylline, theocolin and sodium bicarbonate. In thin-layer chromatography, decomposed compounds producedin combination of FTFG with the 3 preparations were identified as5-fluorouracil. In the dissolution test, FTEG was not dissolved in1st fluid (J. P. IX) in the same combination. Therefore, the combination of FTEG with 2 of the 3 preparations should be avoided, but the combination with the remaining 57 preparations is possible if attention is paid topackaging, duration of administration and other conditions.

By use of X-ray diffractometry, thermal analysis and infrared spectroscopy, the crystal forms of N-(2, 6-dimethylphenyl)-Δ8-dihydroabietamide were investigated. Two polymorphic forms (form I and II), two solvates (cyclohexane and CCl4), and an amorphous form were identified. Heating of form II induced a solid transformation to form I. The transition of form I and II to the amorphous form was observed by grinding. Each solvate contained 9.0% cyclohexane and 16.0% CCl4 respectively, i.e., drug : solvent=2 : 1. The activation energy of the desolvation was determined from TG curves by using Ozawa's method, and calculated as 72.2 kcal/mol for the cyclohexar solvente, and 35.5 kcal/mol for the CCl4 solvate.
