We established a DNA watermarking system for discriminating transgenic plants. The system contains an encryption algorithm based on a binary system, genetic transformation and a detection algorithm for encrypted DNA watermark sequences using a DNA dot plot. The encryption algorithm converted character strings into nucleic acid sequences through binary digits, and the sequence was designed to be resistant to transition mutations to decipher codes completely. Moreover, the encrypted sequences were capable of taking specific nucleotide sequences in using the algorithmic redundancy of the corresponding DNA. Genetic transformation enables labeling plant genomes with DNA watermarks. The detection algorithm allows finding traces of sequence changes in DNA watermarks, complementing the error protection function of the encryption algorithm. To validate the effectiveness of our DNA watermarking system, we introduced a DNA watermark to the tobacco genome and detected the DNA watermark in PCR products amplified from the genome. This indicates that DNA watermark technology is useful for introducing artificial genetic markers in plant organisms, in particular when several transgenic host plants and transgenes are used. The source codes of the Perl scripts are available in this report.

In the field of forensic odontology, not only personal identification using oral conditions, including dental treatment marks and DNA typing, but also species identification, age or sex estimation using cranial or partial bones, and time after death may also be applicable as estimation methods. Among these many tasks, one of the most difficult is species identification of fragmented calvarium. This is because the calvarium has poor morphological features, except that it is a flat bone, and few reports have described how to differentiate it. In this study, a simple identification method for turtle shell (carapace and plastron) whose morphological characteristics closely resembled those of the calvarium was applied. As a result, in an enlarged image obtained using a stereoscopic microscope, the characteristics of each suture pattern could be confirmed. In the decalcified and non-decalcified bone specimens, the difference in the inner/outer laminar structure and the specific structural difference of the cancellous bone-equivalent part were confirmed. Furthermore, most of the features could be obtained in the destructive inspection when it was examined using micro CT imaging to determine whether discrimination by nondestructive inspection was possible or not. No significant difference in ingredients was found using the Electron Probe X-ray Micro Analyzer. The calvarium was more calcified than the carapace but less calcified than the plastron. From these results, we suggest that micro CT imaging is effective for discriminating between the calvarium and turtle shell in a short time.

We investigated the water contents in commercial semi-solid preparations used for pressure ulcer (PU) treatment using near-IR spectroscopy (NIRS) and compared the results with those measured using the Karl Fischer (KF) method. The aim of this study was to determine a standard method and select the appropriate topical preparation with the optimal moisture for PU treatment. The water absorption properties of bases and formulations were evaluated with a time-dependent factor using Transwell as the model membrane. KF and NIRS were applicable as measurement methods of the water content in semi-solid formulations. NIRS was shown to be a useful, simple, nondestructive tool that is more advantageous than the KF method. The water absorption characteristics tested using Transwell revealed that the rate of and capacity for water absorption are determined not only by the absorption ability of the polymer base but also by other factors, such as the osmotic pressure exerted by additives. KF and NIR measurements can be used to choose external skin preparations to control the amount of water in PU treatment.

Direct seeding saves time and labour in the cultivation of rice. However, seedling establishment is often unstable, and yields are lower than in transplanting. Anaerobic germination (AG) is a key trait for improvement of direct seeding of rice. We established a simple and reliable method of evaluating AG in rice breeding. We germinated seeds in distilled water or deoxygenated water and measured coleoptile length several days later; compared the results of each method with survival rate in flooded soil; and used the anoxic water method for QTL analysis and for testing cultivars. Coleoptile elongation in anoxic water and survival rate in flooded soil were significantly correlated (r = 0.879, P < 0.01). A significant QTL, likely to be a major gene (AG1), was found in chromosome segment substitution lines and in a backcrossed F2 population derived from tolerant and sensitive lines. Diverse rice genetic resources were classified into tolerant or sensitive accession groups reflecting their ecotypes. Our study revealed that anoxic water evaluation method saves space and time in a stable environment compared with flooded soil evaluation. It is applicable to QTL analysis and isolation of genes underlying anaerobic germination.

Gene drive is a powerful system that can spread a desirable genetic trait into an entire species and/or population of a certain region,bypassing Mendelian rules of inheritance. Recently, one of the genome editing technologies, CRISPR/Cas, has been developed, making it easier to use gene drive in many different organisms. However, gene drive has potential risks that impact genetic diversity when organisms produced by CRISPR/Cas-based Gene Drive Technology (CCGDT) are accidentally released; therefore, a high degree of prudence is required when CCGDT is used.In Japan, a Working Group on Gene Drive has been established in the Academic Association for Promotion of Genetic Studies (AAPGS), and a Statement on the Handling of Gene Drive was issued to the public including research institute across Japan, after comprehensive and extensive discussions by the working group in order to reduce risks posed by CCGDT. A national-wide survey on CCGDT was implemented. The survey revealed that those in managerial positions including members and secretariats of institutional review boards on recombinant DNA, and biosafety officers are conscious of CCGDT, and efforts to grasp experimental plans involving CCGDT are made by utilizing an application form for recombinant DNA experiment. In contrast, potential risks of CCGDT are not understood by many researchers. All stakeholders need to disseminate potential risks and preventive measures regarding CCGDT to all researchers who may wish to use this technology. Researchers should use CCGDT upon understanding its potential risks and taking necessary measures. This report refers to how safety measures for CRISPR/Cas-based gene drive technology which has potential risks to ecological system has been discussed and results of national-wide questionnaire survey on gene drive in Japan based on the poster presentation at the Asian Conference on Safety & Education in Laboratory 2018.

両大戦間期の西洋のモダン・アートにおいては、新しい映像機器の発達によって示された自然の世界に、新たな造形的モデルを見出す傾向がみられた。本論文では、フランスにおける1930年代の芸術界における前衛芸術を主な対象として、こうした傾向がどのような作品を生み出したかについて考察した。1920年代以前に機械の美学を提唱していたフェルナン・レジェ、ル・コルビュジェとアメデ・オザンファンは、1930年代に、「自然」をテーマとする有機的で不定形な形態をそれぞれ絵画に導入している。それは社会・文化的危機を迎えた1930年代という時代状況で、未来の社会に向けての統一的ヴィジョンを示すための一つの解決法をもたらす方法として選び取られたのであり、とくに彼らの「オブジェ」のとらえ方は、シュルレアリスムの「ビオモルフィスム」とは根本的に異なるものである。さらに1930年代のフランスで非具象表現を目指して組織された「アブストラクシオン・クレアシオン」の活動にも、有機的な形態を造形へと取り込んだ画家たちがいる。彼らは、カメラのような機械を自然や生へと到達するための装置とみなし、非具象的なイメージを、物理的な世界へと結びつけようとした。これらの試みには科学と芸術を同一の地平で扱おうとした、先駆的な試みをみることができる。

Chemical investigation of field-collected fruit bodies of the myxomycete Cribraria meylanii resulted in the isolation of a naphthoquinone pigment, cribrarione C, and its structure was elucidated by spectral data as 2,5,6,7-tetrahydroxy-1,4-naphthoquinone (1). This compound (1) had been synthesized previously, while it was isolated here for the first time as a natural product, and its NMR and MS data are described in this study.

Background:Ventricular septal defect (VSD) after myocardial infarction (MI) is a rare but fatal complication. We report patients’ characteristics and operative outcomes after surgical repair of post-MI VSD using a national database of Japan.Methods and Results:This was a retrospective review of the Japan Adult Cardiovascular Surgery Database (JCVSD) to identify adults (age ≥18 years) who underwent surgical repair of post-MI VSD between 2008 and 2014. The primary outcome was operative death. We identified 1,397 patients (671 male [48%], 74.1±9.3 years old) undergoing surgical repair of post-MI VSD among 288,736 patients undergoing cardiac surgery enrolled in the JCVSD during the same period. Of these, 1,075 (77.0%) were supported preoperatively with an intra-aortic balloon pump. Surgical status was urgent in 391 (28.0%) and emergency/salvage in 731 (52.3%). Concomitant coronary artery bypass grafting was performed in 475 (34.0%). Overall 30-day and operative mortalities were 24.3% and 33.0%, respectively. Operative mortality varied according to surgical status: 15.6% in elective, 30.9% in urgent, and 40.6% in emergency/salvage cases. Multivariable analysis identified advanced age and emergency/salvage status as being strongly associated with increased odds of operative death.Conclusions:Post-MI VSD remains a devastating complication in Japan as well as in the USA and Europe.

In human medicine, computed tomography (CT) is the gold standard for visceral fat measurement. Research shows that the visceral fat area (VFA) of the umbilical slice is significantly correlated with the visceral fat volume (VFV). In veterinary medicine, however, few studies have evaluated visceral fat using CT. This study aimed to evaluate the visceral fat in dogs using CT images, and determine if the slice significantly correlated with VFV to simplify visceral fat measurements. This retrospective study includes data on 90 dogs that underwent whole-body CT scans for diagnostic purposes. VFV was calculated as the product of VFA and thickness in each CT slice; the correlation between VFV and VFA was analyzed at the level of each lumbar vertebra. Visceral fat percentage (VF%) was calculated as the ratio of the product of VFV and fat density to the body weight. Visceral fat area percentage (VFA%) was calculated as the ratio of VFA to the body area, and its correlation with the VF% and the body condition score (BCS) was analyzed. VFA was highly correlated with VFV at the level of each lumbar vertebra, with the highest correlation (r=0.964) at the L3 level. VFA% was significantly correlated with VF% (r=0.930) and weakly correlated with BCS (r=0.523). This study demonstrates that it is sufficient to use only the L3 slice for visceral fat evaluation and that the evaluation can be based on VFA% of the L3 level.

Dasatinib-related chylothorax is a rare adverse event, and the mechanism underlying its occurrence is still not fully understood. We herein report the case of a 73-year-old woman with chronic myeloid leukemia (CML) who developed dasatinib-related chylothorax refractory to conventional treatments, except for steroids. To the best of our knowledge, this is the first case of dasatinib-related chylothorax which was successfully controlled by combining diuretics with the Japanese herbal medicine "Goreisan." "Goreisan" is known to inhibit aquaporin channels and regulate the water flow. Our findings showed that "Goreisan" is an effective treatment option for uncontrollable dasatinib-related chylothorax.

[Purpose] The Gait Judge System measures the plantar flexion resistive torque acting on the angle of the ankle joint, as well as the ankle joint itself, of the ankle-foot orthosis using a 1,000 Hz sampling frequency. This pilot study aimed to determine the characteristics of plantar flexion resistive torque acting on the double Klenzak ankle joint of the ankle-foot orthoses worn by healthy individuals. [Participants and Methods] Participants were eight healthy young adults (3 male, 5 female; mean age, 26.8 years old; mean height, 165 cm.; mean body weight, 56.3 kg). Plantar flexion resistive torques and angles of the ankle joint in gait cycles were measured with the Gait Judge System. Speed of gait was calculated using a ruler attached on the floor and the Gait Judge System video. We classified waveforms according to the existence of second peaks in the gait cycle. The correlations between parameters related to the plantar flexion resistive torque and the speed of gait were evaluated using Pearson’s simple correlation analysis. [Results] The plantar flexion resistive torque showed two peaks: the first peak was at the loading response, measured at 17.4 Nm, and the second peak was at the pre-swing phase, measured at 10.9 Nm. However, the second peak was absent in three of the participants. The normalized second peak and the second peak/first peak ratio had a strong, positive correlation with the speed of gait. [Conclusion] The Gait Judge System revealed typical waveforms according to the parameters set in this study.

本稿では、まず小学校体育における「体力を高める運動」にかかわり、最近の実践報告や研究動向から、「体力を高める運動」の問題を浮き彫りにした。そこでは、「体力を高める運動」の実践報告はきわめて少なく、教材の開発もあまり進められていないことが明らかになった。また、その背景には小学生には受けいれられにくいとされる必要充足機能が強調されており、その内容はトレーニング的で量的な体力を問題にする数値主義に基づき、自己の体力の高まりに着目した個人主義的な立場から「体力を高める運動」が位置づけられていることが明らかになった。加えて、最近の研究では、「体力を高める運動」の運動の特性、学習観、身体観といった枠組みから展開されていることが明らかになったが、実際にどのように運動の内容構成を行っていけばよいのかという教材開発の提示までには至っていないことが浮き彫りとなった。そこで、「体力を高める運動」における、①運動特性の捉え方、②学習観の捉え方、③身体観の捉え方の三点について検討した結果、運動の特性は欲求充足機能を前面に取り上げながら、結果として必要充足機能に結びつけるという表裏一体のものとして捉えることが肝要であると述べた。また、学習観は個人主義的な学習観から関係主義的な学習観へシフトすることが重要であると述べ、身体観は一人称的・三人称的身体から二人称的身体へと転換することで、新たな体育教育をひらく可能性があると考察した。こうした視点を持ちながら、「体力を高める運動」の新たな内容構成に基づく教材開発を行った結果、欲求充足と必要充足の機能の両方を重視しながら教材を作成するために、カード(A6版)形式を採用し、カードの表面には欲求充足の観点が分かるように、「運動の中心的なおもしろさ」をイラストと文章で表記し、カードの裏面には体力の四つの要素(体の柔らかさ・巧みな動き・動きを持続する能力・力強い動きを高めるための運動)のどれと結びついているかという観点で示した。また、仲間と共に楽しみながら行える運動を行うことができるという観点から30の運動を選定し、仲間に働きかけたり、仲間から働きかけられたりすることにより生まれる世界を大切にする二人称的な身体から運動を取り上げた。

本研究の目的は、まんがと映画の関係について、実際にまんがを映像化して研究資料自体を制作することと、作成した資料をもとに異なるメディウムである両者について検証を試みることである。すでにまんがと映画の関係については、多くの先行研究により様々な角度から検証が行われているが、それらを踏まえつつ新たな論点を探求することも視野に入れる。本論では、大塚英志原作、藤原カムイ作画『アンラッキーヤングメン』(角川書店、2004~6)の一部を逐語訳的に映像化した上で、すでに一部逐語訳的映像化を行った石ノ森章太郎『龍神沼』(講談社、1961)の映像と適宜比較検証することで特徴を明らかにするとともに、1960 年代と2000 年代の映画的手法における表現の変化についても考察していく。The objective of this case study is to compare and verify the relationship between the manga and the film created with manga as a base. The research about this process is carried out by many researchers previously. So, including the viewpoint of other researchers, I will add my own views to find a new approach to the issue of these studies.In this paper, I am converting the part of manga written by Eiji OHTSUKA and Kamui FUJIWARA titled "UNLUCKY YOUNGMEN" and compare it to the manga based film" RYUJINNUMA" previously converted by myself which was written by Shotaro ISHINOMORI in 1961. Comparing and verifying these two films conversion process will find out the difference between the changes of the method of film in manga from 1960s' to 2000s'.

To develop potent ligands for the vitamin D receptor (VDR), we designed and synthesized a series of vitamin D analogues with and without 22-alkyl substituents. These analogues exhibited agonistic, partial agonistic, or antagonistic activity. To elucidate the mechanism of action of the analogues, we conducted crystal structure analyses of the ligand-binding domain (LBD) of VDR complexed with the analogues. The VDR-LBD/agonist complex exhibited precise interactions, which clearly explained VDR agonism. The VDR-LBD/partial agonist complex showed two conformers (agonist and antagonist binding conformers) in a single crystal, demonstrating that partial agonism could be explained by the sum of the agonistic and antagonistic activities. Antagonist binding to the VDR-LBD structure was elucidated using both crystal structure analysis and in-solution structural analyses with the small-angle X-ray scattering (SAXS)-molecular dynamics (MD) and hydrogen/deuterium exchange coupled with mass spectrometry (HDX-MS) methods. Several antagonist-binding structures were detected. We found that the antagonist binding structures differed depending on the structure of the antagonist itself, and those structures clearly explained the VDR antagonism. Furthermore, the apo VDR-LBD structure without the ligand in the ligand-binding pocket was revealed and found to have an entrance to accommodate the ligand. Thus we elucidated the mechanisms of action of agonists, partial agonists, and antagonists based on structural changes (differences) in the receptor protein induced by ligand binding.

The purpose of this work is to study the electric properties of graphite and the synthesis of a few layers graphite intercalation compounds (GIC). First, intercalation of Li between graphite layers was done by soaking the substrates into a 20 ml n-butyllithium solution. Second, intercalation of FeCl3 into the graphite was done by heating FeCl3 powder and graphite in the quartz tube. For Li intercalation, the appearance of an extra Bragg reflection at 23.7º indicates that Li atoms were intercalated between the graphene sheets in the specimen we grew. For FeCl3 intercalation, the (00l) Bragg reflection was caused by the FeCl3-GIC. Probably Iron Chloride was intercalated in graphite. In the Raman spectra of graphite and FeCl3-GIC, the G, D and 2D peaks were confirmed. Additional peaks only in the specimen of FeCl3-GIC were observed at 1605 cm-1 and 3415 cm-1. It is speculated that the peak is related to the intercalation of FeCl3 into the graphite.
