A free piston double diaphragm shock tube with 70 × 70mm cross-section at test section was newly developed for the purpose of investigating nonequilibrium phenomena behind a shock wave. This paper presents the performance of the shock tube and the measurement system newly developed. Experimental investigation to clarify its characteristics was conducted for various operational parameters, such a rupture pressure of the first diaphragm and initial pressures of the low pressure tube, the high pressure tube, and the compression tube. Based on the characteristics experiment, a performance map of the shock tube was obtained by changing the operation parameters. The result shows that the shock tube is capable of generating the shock layer corresponding to the super orbital reentry flight conditions. The newly developed measurement system enables us to obtain the spatial distribution of spectra behind a shock wave with high spatial and time resolution. A description of the measurement system and typical examples of the measured spectra are presented.

2015年8月現時点において産学連携関連の研究は、大規模データ分析を用いたものを初めとするマクロ的な視点で論じている研究が多い。それとは対照的に、実際的・現場的ミクロな視点を持ったものは少ない。そこで本論文は私がデータ分析システムを開発・販売する会社(AIC社)を起業した経験をまとめた。内容としては起業を計画するところから、製品化断念までの実体験をまとめた。また失敗の原因の考察、考察から見えてきた製品化成功のための一仮説を掲載している。

A single-flux-quantum (SFQ) arithmetic logic unit (ALU) was designed and tested to evaluate the effectiveness of introducing dynamically reconfigurable logic gates in the design of a superconducting logic circuit. We designed and tested a bit-serial SFQ ALU that can perform six arithmetic/logic functions by using a dynamically reconfigurable AND/OR gate. To ensure stable operation of the ALU, we improved the operating margin of the SFQ AND/OR gate by employing a partially shielded structure where the circuit is partially surrounded by under- and over-ground layers to reduce parasitic inductances. Owing to the introduction of the partially shielded structure, the operating margin of the dynamically reconfigurable AND/OR gate can be improved without increasing the circuit area. This ALU can be designed with a smaller circuit area compared with the conventional ALU by using the dynamically reconfigurable AND/OR gate. We implemented the SFQ ALU using the AIST 2.5kA/cm2 Nb standard process 2. We confirmed high-speed operation and correct reconfiguration of the SFQ ALU by a high-speed test. The measured maximum operation frequency was 30GHz.

Individual differences of drug-metabolizing enzymes are important determinants for the metabolic fate of chemicals. This article focuses on polymorphic human flavin-containing monooxygenase 3 (FMO3) and dietary-derived trimethylamine. Malodorous trimethylamine is generally converted to odorless trimethylamine N-oxide by liver microsomal FMO3. Trimethylaminuria is caused by functional disorder of FMO3. In this study mutations of the FMO3 gene were examined in self-reported Japanese trimethylaminuria subjects that showed low FMO3 metabolic capacity in urine tests. Nine novel polymorphisms in the FMO3 gene were discovered in self-reported Japanese volunteers. Functional analyses of recombinant FMO3 proteins suggested that these FMO3 gene mutations were one of the causal factors for decreased FMO3 function resulting in trimethylaminuria. Inter-individual variations of FMO3-mediated microsomal oxygenation activities, levels of FMO3 protein and FMO3 mRNA, and its modification in liver microsomes from Japanese samples were observed. Both genetic polymorphisms in the 5′-upstream of the FMO3 gene and some hormonal changes related to menstruation may be causal factors for inter- and/or intra- individual expression levels of FMO3. To assess the palliative cares, it was found that absorbed levels of trimethylamine in vivo would be possibly controlled by selection of precursor foods like fish containing a variety of trimethylamine amounts. These lines of evidence suggest that individual differences of FMO3 are important determinants for the metabolic fate of dietary-derived trimethylamine.

Inflammatory bowel disease (IBD) is a refractory disease of the gastrointestinal tract that is believed to develop in genetically susceptible individuals. Glycosylation, a type of post-translational modification, is involved in the development of a wide range of diseases, including IBD, by modulating the function of various glycoproteins. To identify novel genes contributing to the development of IBD, we analyzed single nucleotide polymorphisms (SNPs) of glycosylation-related genes in IBD patients and identified MAN2A1, encoding alpha-mannosidase II (α-MII), as a candidate gene. α-MII plays a crucial, but not exclusive, role in the maturation of N-glycans. We also observed that intestinal epithelial cells (IECs), which establish the first-line barrier and regulate gut immunity, selectively expressed α-MII with minimal expression of its isozyme, alpha-mannosidase IIx (α-MIIx). This led us to hypothesize that IEC-intrinsic α-MII is implicated in the pathogenesis of IBD. To test this hypothesis, we generated IEC-specific α-MII-deficient (α-MIIΔIEC) mice. Although α-MII deficiency has been shown to have a minimal effect on N-glycan maturation in most cell types due to the compensation by α-MIIx, ablation of α-MII impaired the maturation of N-glycans in IECs. α-MIIΔIEC mice were less susceptible to dextran sulfate sodium-induced colitis compared with control littermates. In accordance with this, neutrophil infiltration in the colonic mucosa was attenuated in α-MIIΔIEC mice. Furthermore, gene expression levels of neutrophil-attracting chemokines were downregulated in the colonic tissue. These results suggest that IEC-intrinsic α-MII promotes intestinal inflammation by facilitating chemokine expression. We propose SNPs in MAN2A1 as a novel genetic factor for IBD.
