Gold nanoparticles (GNPs) have been reported to exhibit a variety of biological effects including anti-inflammatory and anti-oxidant activities. The extent of an in vitro glycation reaction mixture of collagen and glycolaldehyde was assayed to investigate the inhibition of glycolaldehye-derived advanced glycation end products (glycol-AGEs) formation with GNPs in collagen, which is a major protein component of the human dermis. GNP-treated collagen showed significantly less glycation (56.3±4.2%) than an untreated glycation control. Moreover, GNP-treated glycation in a collagen lattice model significantly decreased the AGEs distribution in the model system. Taken together, these results suggest that GNPs have the potential for use in the prevention of glycation-induced skin aging.

Wide-ranging varieties and/or strains of bulb onions (Allium cepa Common onion group) and shallots (A. cepa Aggregatum group) were utilized to understand the variation in chemical compounds responsible for their taste. The bulb samples of 10 F1 commercial onion varieties (seven short-day and three long-day varieties) from Japan and 12 shallot landraces from abroad (Vietnam: three landraces; Indonesia: nine landraces) were collected as plant materials once a year in 2014 and 2015. The contents of S-alk(en)yl-L-cysteine sulfoxides, total flavonoids, and soluble sugars—including fructose, glucose, sucrose, and fructans—were determined to find differences between bulb onions and shallots, as well as to detect variations among varieties and/or landraces. While a principal component analysis (PCA) based on the results from both 2014 and 2015 could clearly discriminate shallots from bulb onions from a phytochemical perspective, bulb onions mainly had higher monosaccharides than shallots. By contrast, shallots produced more disaccharides than bulb onions. In most cases, regression analyses using the numerical data of the chemical compounds found in bulb onions and shallots suggested year-year correlations between 2014 and 2015. The flavonoid and PeCSO (S-(1-propenyl)-L-cysteine sulfoxide: isoalliin) contents in shallots were higher than those detected in bulb onions, which indicated the stronger pungent and bitter taste could be attributable to excess amounts of these compounds in this tropical plant.

In this article, experimental tests were conducted to study the characteristics of the surge wave and its effects on the failure process of a glacier dam. The results indicated that two obvious surges were monitored by pore pressure transducers (PPTs) when the blockage slid into water. The surge wave attenuated exponentially near the plunging location and then attenuated slowly when it propagated downstream. When the surge wave reached the glacier dam it climbed up along the dam and flew over. Sequential propagating surges exerting on the dam were one of the main factors causing the failure of the glacier dam. The failure mechanism of glacier dam triggered by the surge waves under different initial water supply conditions was primarily analyzed in this article.

The present study investigated how impacts of the inclusion of radiosonde observations conducted locally in the early summer of 2012 over the Kuroshio and Kuroshio Extension (KE) can spread over time across the North Pacific basin to influence the predictability of synoptic and large-scale tropospheric circulation. For that purpose, observing system experiments (OSEs) were performed where each of two extra sets of radiosonde data, one obtained over the East China Sea in mid-May and the other over the KE in early July, was added to an atmospheric ensemble data assimilation system for comparison with the corresponding analyses without those data. The experiments show that the impact of the extra data assimilated propagates eastward mainly due to advection by the subtropical jet (STJ) in May and July. The strong STJ in May allows the upper-tropospheric impact to travel across the basin only within two days. Under the weaker STJ, the corresponding impact in July tends to remain within the western Pacific, until it eventually reaches the eastern portion of the basin. Assimilation of the extra radiosonde data over the Kuroshio or KE can lead to a decrease of pressure over the Gulf of Alaska in both May and July.  Additional forecast experiments based on the OSEs for May revealed that the pressure decrease over the Gulf of Alaska can be traced back to the west of the Alaska Peninsula and to the east of Japan over three days. The impacts that originate on different dates via different paths merge over the central North Pacific, reinforcing the cyclone over the Gulf of Alaska. This study presents examples where the impacts of atmospheric observations over the western boundary current can propagate across the ocean basin through the westerlies to influence the forecast skill in distant regions.

Epidemiological and pathological studies on Helicobacter spp. in feline stomachs in Japan were conducted using genus- and species-specific (H. felis, H. bizzozeronii, H. heilmannii sensu stricto[s.s.] and H. pylori) polymerase chain reactions (PCRs), ureAB gene sequencing and histopathology. PCR results showed that 28 of 56 cats were infected with Helicobacter spp., and H. heilmannii s.s. was the most prevalent species by both PCR (28/28) and ureAB gene sequencing (26/28). Some of the sequences showed high similarities with those from human patients with gastric diseases (99%). There were no significant differences between Helicobacter spp.-positive and -negative cats in the severity of chronic gastritis (P=0.69). This is the first extensive epidemiological study on feline gastric Helicobacter spp. in Japan.

The observation operators in the local ensemble transform Kalman filter (LETKF) were improved to enable use of observations in the vicinity of the poles in the data assimilation system composed of the atmospheric general circulation model for the Earth Simulator (AFES) and the LETKF. The improved observation operators allow to assimilate the observations located south (north) of southernmost (northernmost) Gaussian grid latitudes. An algorithm for searching the nearest observations from an analyzed grid for error covariance localization was also modified to efficiently assimilate observations near the poles.The new algorithms were incorporated into the LETKF, and the impacts of routine radiosonde observations at the South Pole during the periods of July 2012 and January 2013 were assessed. The radiosonde observations suppressed an artificial expansion of the analysis ensemble spread which occasionally caused numerical instability in the upper troposphere and the lower stratosphere over the Antarctic regions. The analysis was also improved in the Antarctic regions.

Background:Pulmonary vein isolation (PVI) using the 2nd-generation cryoballoon (CB2) for the treatment of atrial fibrillation (AF) has demonstrated encouraging acute and mid-term results. However, follow-up data on outcomes beyond 1 year are sparse. We investigated the 3-year outcome after PVI using the CB2.Methods and Results:100 patients with paroxysmal (PAF, 70/100 [70%] patients) or persistent AF (pAF, 30/100 [30%] patients) underwent CB2-based PVI in 2 experienced centers in Germany. Freeze-cycle duration was 240 s. After successful PVI a bonus freeze-cycle of the same duration was applied in the first 71 patients but was omitted in the following 29 patients. Phrenic nerve palsy occurred in 3 patients (3%); 2 patients were lost to follow-up. After a median follow-up of 38 (29–50) months, 59/98 (60.2%) patients remained in stable sinus rhythm (PAF: 48/70 (69%), pAF: 11/28 (39%) P=0.0084). In 32/39 (77%) patients with arrhythmia recurrence, a second ablation procedure using radiofrequency energy was conducted. Persistent PVI was noted in 76/125 (61%) PVs. After a mean of 1.37±0.6 procedures and a median follow-up of 35 (25–39) months, 77/98 (78.6%) patients remained in stable sinus rhythm (PAF: 56/70 (80%), pAF: 20/28 (71%), P=0.0276).Conclusions:CB2-based PVI resulted in a 60.2% single-procedure and a 78.6% multiple-procedure success rate after 3 years. Repeat procedures demonstrated a high rate of durable PVI.

Hypertension is a major risk factor for cardiovascular diseases and is detrimental to several organs including the liver and kidneys. The flaxseed-derived polyunsaturated fatty acids including the omega-3 and omega-6 essential fatty acids have been shown to blunt the effects of hypertension. It is however, unclear whether the flaxseed, which is rich in these essential fatty acids, could improve the liver and kidney dysfunctions observed in the hypertensive condition. To test this, functional markers of the liver and kidneys, including aspartate aminotransferase (AST), alanine aminotransferase (ALT), blood urea nitrogen (BUN), uric acid (UA), creatinine, and renin were examined in hypertensive male Wistar rats fed a flaxseed diet. Normotensive rats maintained on a standard diet were rendered hypertensive with a daily administration of cyclosporin A (CYS) (25 mg/kg) for 4 weeks. Subsequently, hypertensive rats were either fed a standard diet alone or a flaxseed-supplemented standard diet (FLX; 10% W/W) for 8 weeks. Compared to normotensive rats, standard diet-fed hypertensive rats had significantly elevated blood pressure, altered lipid profile, and increased plasma levels of tissue markers measured immediately following the CYS treatment and thereafter at 4 and 8 week intervals. On the other hand, rats fed the FLX-supplemented diet had significantly lower blood pressure, an improved lipid profile and decreased tissue marker levels measured after 4 and 8 week durations. The data demonstrate for the first time the favourable effects of FLX in improving liver and kidney functions in the hypertensive condition. These effects are likely to be mediated by the alpha-linolenic acid (ALA) and linoleic acid (LA) contents of flaxseed oil due to its demonstrated ability to lower the blood pressure.

近年,良質な睡眠を得るための睡眠環境に関する研究が多く報告されている.しかしながら,起床環境に関する研究はあまり報告されていない.本研究は快適な起床環境の構築を目的とし,前段階として被験者6人に対して起床音圧の差異(60dB or 80dB)が起床状態に与える影響を調査した.生体に対する影響について覚醒度:AAC,交感神経活動:LF/HF唾液中クロモグラニンA,活動性と気分の状態:GVA作業能率:作業能率課題解答率を用いて評価した.結果は低音圧(60dB)において覚醒度,括動度が高く交感神経活動が優位であり,高音圧(80dB)では気分が良い傾向が示された.

Background: Aging is associated with adverse outcomes in patients with cardiac diseases. Whether elderly patients hospitalized with heart failure (HF) had increased risks for mortality and rehospitalization compared with younger patients during the long-term follow-up was examined. The predictors of these adverse outcomes were also identified. Methods and Results: The Japanese Cardiac Registry of Heart Failure in Cardiology (JCARE-CARD) studied prospectively the characteristics and treatments in a broad sample of 2,675 patients hospitalized with worsening HF and the outcomes were followed up. The majority of elderly patients were female, had lower body mass index (BMI), a higher rate of ischemic, valvular, and hypertensive heart disease as etiologies of HF, a lower estimated glomerular filtration rate (eGFR), lower hemoglobin, and higher left ventricular ejection fraction values. Even after adjustment for covariates, the elderly patients were associated with higher risks of adverse outcomes. The predictors for all-cause death were: lower eGFR, lower BMI, male sex, sustained ventricular tachycardia or fibrillation (VT/VF), and the use of diuretics at discharge. Conclusions: Among patients hospitalized with HF, elderly patients had a worse prognosis than younger patients. Lower eGFR, lower BMI, male sex, sustained VT/VF, and diuretic use were independent predictors for all-cause death in these patients with higher risk. (Circ J 2011; 75: 2403-2410)

Seed size is one of the most important traits in leguminous crops. We obtained a recessive mutant of blackgram that had greatly enlarged leaves, stems and seeds. The mutant produced 100% bigger leaves, 50% more biomass and 70% larger seeds though it produced 40% less number of seeds. We designated the mutant as multiple-organ-gigantism (mog) and found the mog phenotype was due to increase in cell numbers but not in cell size. We also found the mog mutant showed a rippled leaf (rl) phenotype, which was probably caused by a pleiotropic effect of the mutation. We performed a map-based cloning and successfully identified an 8 bp deletion in the coding sequence of VmPPD gene, an orthologue of Arabidopsis PEAPOD (PPD) that regulates arrest of cell divisions in meristematic cells. We found no other mutations in the neighboring genes between the mutant and the wild type. We also knocked down GmPPD genes and reproduced both the mog and rl phenotypes in soybean. Controlling PPD genes to produce the mog phenotype is highly valuable for breeding since larger seed size could directly increase the commercial values of grain legumes.

Many musical methods and teaching materials for preschool children are currently introduced and published in Japan. Although this phenomenon reflects that the importance of musical education for infants is extensively admitted, it is the fact that the materials whose purpose is unclear for the early childhood education are increasing these days. On the other hand, it is reasonable that the old nursery songs and traditional music of Japan are payed attention as a reaction against for attaching to occidental music since the Meiji era in the history of school education. This study argued about odaly method which was established by a hungarian pedagogist Kodaly Zoltan and considerd how to teach the Japanese nursery songs to infants along the musical purpose.

Although it is established that the composition of the human intestinal microbiota changes with age, transition of the intestinal microbiota of animals with age has not been well studied. In the present study, we collected fresh fecal samples from dogs of 5 different age groups (pre-weanling, weanling, young, aged, senile) and analyzed the compositions of their intestinal microbiota with a culture-based method. The results suggested that the composition of the canine intestinal microbiota also changes with age. Among intestinal bacteria predominant in dog intestines, lactobacilli appeared to change with age. Both the number and the prevalence of lactobacilli tended to decrease when dogs became older. Bifidobacteria, on the other hand, was not predominant in the intestine of the dogs. We also identified lactobacilli at the species level based on 16S rRNA gene sequences and found that the species composition of Lactobacillus also changed with age. It was further suggested that bacteria species beneficial to host animals may differ depending on the host species.

Hydrosol prepared from the flowers of Rosa damascena (rose water) has been traditionally used for various health-related issues, including skin troubles such as erythema, itchiness, swelling. For the care of these skin troubles caused by microbial infection, both antimicrobial and antiinflammatory effects are required. Here, we investigated the effects of rose water on the growth of Candida albicans and methicillin-resistant Staphylococcus aureus (MRSA), which cause skin infections, and on the function of neutrophils, which play a major role in the regulation of inflammatory reactions. To assess its modulatory effects on neutrophils, the effects of rose water against neutrophil adhesion response were evaluated. Rose water inhibited mycelial growth of C. albicans at a concentration of ca. 2.2%, and reduced viability of MRSA within 1 h. Rose water suppressed neutrophil activation induced by lipopolysaccharide (LPS), tumor necrosis factor alpha (TNF-α), and N-formyl-Met-Leu-Phe (fMLP) at 5–15%. It also reduced the LPS- and TNF-α-induced cell surface expression of the adhesion-related molecule, cluster of differentiation (CD) 11b, but did not affect the migratory capacity of neutrophils with or without chemoattractant. These results suggest that rose water may reduce the pathogenicity of microbes, and attenuate neutrophil stimulation, which is involved in inflammatory responses. These findings suggest that rose water has a potential effect to inhibit skin inflammation caused by microbes.

This study was conducted in order to establish a health management method for the elderly in a community through follow-ups of bone mineral density (BMD) measurement results over a 1-year period based on BMD measurements performed by pharmacists and a guidance program. Regarding BMD measurement results, the percent young adult mean (%YAM: mean BMD in healthy persons of the same sex aged between 51 and 82 years old) significantly increased in Period I, during which the intervention by pharmacists was performed (6 months after the start of measurements), but significantly decreased in Period II, during which this intervention was not performed (between 7 and 12 months after the start of measurements). Based on these results, lifestyle improvements were effective in Period I regardless of sex or age; however, it may be important to maintain an improved diet and subject motivation in the future. The results of this study suggest that community pharmacists play an important role in community medicine through positive intervention for local residents' health support.

This study aimed to develop a label-free, sensitive, selective, and environment-friendly fluorescent peptide probe His-His-Trp-His (HHWH) for determining the concentration of copper ion (Cu2+) in aqueous solutions. The results demonstrated that the designed HHWH has a high selectivity and sensitivity for monitoring the concentration of free Cu2+ via quenching of the probe fluorescence upon a binding of Cu2+. The fluorescence intensity of the HHWH had a linear relationship with the concentration of Cu2+ between 10 nM and 10 μM, and the detection limit was 8 nM. Furthermore, HHWH could be regenerated with sulfide ions at least five times. The concentrations of Cu2+ in three different real water samples were detected using this probe, and the results were consistent with the one detected using an atomic absorption spectrometer. Thus, HHWH can be used as an accurate and feasible fluorescent peptide probe for detecting Cu2+ in aqueous solutions.
