Accurate DNA quantification is required for the short tandem repeat typing of forensic samples. Various commercial DNA quantification kits for forensic DNA analysis have recently been released. However, DNA quantification may fail if too much PCR inhibitor is extracted with the target DNA sample. Therefore, this study experimentally evaluated the degree of the alteration of DNA quantification results of various commercial DNA quantification kits (Takara RR281, Quantifiler Duo DNA Quantification Kit, Plexor HY System, Investigator Quantiplex HYres Kit, Quantifiler Trio DNA Quantification Kit, and Quant-iT dsDNA HS Assay Kit) in the presence of various concentrations of PCR inhibitors such as hematin, humic acid, indigo carmine, melanin, and tannic acid. Compared to the other kits, the DNA quantification abilities of the Investigator Quantiplex HYres Kit and Quant-iT dsDNA HS Assay Kit were not negatively affected by high concentrations of hematin. The DNA quantification abilities of the Quantifiler Trio DNA Quantification Kit and Quant-iT dsDNA HS Assay Kit were not negatively affected by high concentrations of PCR inhibitors containing humic acid, indigo carmine, or melanin. The DNA quantification abilities of the Investigator Quantiplex HYres Kit and Quantifiler Trio DNA Quantification Kit were not negatively affected by high concentrations of tannic acid. Furthermore, the DNA quantification ability of the Quantifiler Trio DNA Quantification Kit was more reproducible than those of the other kits. Moreover, this kit was able to assess the degree of DNA degradation by comparing quantified both the short amplicon (80 bp) and long amplicon (130 bp). However, the short amplicon was not negatively affected by high concentrations of PCR inhibitors in contrast to the long amplicon. These results indicate forensic DNA analysts should select the appropriate DNA quantification kits that are unaffected by PCR inhibitors and carefully interpret DNA quantification results.

視床痛は視床出血や視床梗塞後に伴う難治性中枢性疼痛として知られている。しかし,薬物療法や外科的治療法が試みられているが確実な治療法がない。我々は難治性の視床痛6例に漢方治療を試み,症状の軽減した4例を経験した。症例は年齢27才から70才の男性4例,女性2例であった。診断は右視床出血3例,左視床出血1例,右視床梗塞2例で,発症後から漢方治療までの期間は6カ月から12年と幅があった。改善した4例では症状がほぼ消失から4割程度減弱した。改善例は全例,烏頭・附子含有方剤が用いられていた。抑肝散加陳皮半夏が精神症状の強い2例で有効であった。また,駆瘀血剤のみで十分改善を認めた症例もあった。症状が長期にわたる症例では,有効例でも症状は残存した。さらに難治例は症状が固定しており,強い麻痺等を伴う症例であった。

Intracellular and extracellular mechanical forces affect the dynamics of actin cytoskeletons, however, the underlying biophysical mechanisms how forces are transduced into changes in the actin dynamics remain largely unknown. We find that activity of the actin severing protein ADF/cofilin negatively depends on tension in the actin filament through tension-dependent binding/unbinding of cofilin to/from the actin filament, thereby relaxed actin filaments are severed whereas tensed ones are not. Here we review the latest knowledge on this phenomenon and discuss its biological impact.

構造生物学は、タンパク質の構造情報を明らかにすることで創薬に貢献してきた。この20年間はX線を用いた構造解析が中心であり、とくにキナーゼに代表される酵素では、合成化学者が構造情報をもとに活性向上を指向したStructure-Based Drug Discovery(SBDD)を精力的におこなってきた。 近年注目を集めているクライオ電子顕微鏡により、さらに構造生物学の幅がひろがろうとしている。クライオ電子顕微鏡はチャネルやトランスポーター、複合体など、X線結晶構造解析での構造取得が難しかった分子量が大きく、かつ、複数コンホメーションを取り得る生体分子の構造解析を得意としている。これらのターゲットクラスにはhERGなど毒性に関与するタンパク質が多くあり、構造情報が得られることでオフターゲット回避もSBDDを用いて効率的におこなえるようになると期待できる。 クライオ電子顕微鏡を用いた構造解析例は年々増えている一方で、残念ながら日本では絶対数としてクライオ電子顕微鏡の数が少なく企業が個別に利用できる環境にはない。毒性や動態で課題となるタンパク質は共通であることから、当社をはじめとする製薬企業では非競合領域での産官学連携組織を立ち上げて、毒性・動態関連タンパク質の構造解析に共同で取り組んでいる。本講演ではこの連携について紹介し、毒性領域へのさらなる応用について議論したい。

2011年東日本大震災が発生した以降,災害記録を残すべく,自然災害デジタルアーカイブが数多く構築された。証言記録,行政文書,写真記録,動画など数百万点にも及ぶ災害記録を今後の防災・減災に活かすためには,災害記録がどのようなものであるかを知ることが重要である。本稿では,自然災害デジタルアーカイブで公開されている数百万点に及ぶ災害記録を活かすために,災害記録の活用方法やその注意点について述べる。さらに,東日本大震災デジタルアーカイブを中心に,近年の様々な自然災害デジタルアーカイブについても紹介し,利用方法についても合わせて概説する。

In dieser kurzen Abhandlung möchte ich, nachdem ich den Narren in den Fastnachtspielen des Nürnberger Prokurators Jakob Ayrer im Vergleich mit dem bei Hans Sachs beobachtet habe, nach des ersteren grundsätzlichem Talent für die Dichtung fragen. Zuerst muß man zu ermessen versuchen, welchen Einfluß die englischen Komödianten auf Ayrer ausübten, wobei wir die Gattung der Spiele, ohne seine Komödien und Tragödien zu berücksichtigen, ausschließlich auf die Fastnachtspiele beschränken. Im Jahre 1597 hatte Ayrer in seiner Heimatstadt Nürnberg die Gelegenheit, den Aufführungen der englischen Truppe, die von Thomas Sackeville geleitet wurde, beizuwohnen. Ayrer führte die Rolle der lustigen Person auf der deutschen Bühne ein und nannte sie Jann Posset. Es ist klar, daß der Name des Narren Jann Posset aus John Bouset herzuleiten ist, den der englische Darsteller Sackeville schuf. Was Jann Posset als Narren betrifft, so geht es in den Fastnachtspielen Ayrers um Wortspiele, Prügelszenen, die Figur des Pantoffelhelden, den Auftritt aus dem Klosett sowie um Essen und Trinken. Jann Posset muß offenbar zu dem Typus _"clown" gerechnet werden.<br>Ein Narr namens Jeckel tritt in den Fastnachtspielen von Hans Sachs zweimal auf: er kann Freiheiten und Privilegien zur Genüge genießen, weil er außerhalb der sozialen Normen steht, da er keine Vernunft besitzt. Jeckel ist, wie sein Hut und Stab beweist, der von Fürsten oder dem Adel gedingte Hofnarr. Daher übernimmt Ayrer meiner Meinung nach die Rolle des Narren keinesfalls von Hans Sachs, so daß in der Tat Jann Posset mit Jeckcl nichts zu tun hat.<br>Jann Posset stellt auf der deutschen Bühne keine Nebenrolle, sondern die manchmal den Prolog seines Stückes sprechende Hauptfigur dar. Auch daß er ein geborener Bauer war und dazu sein Name aus der Eßkultur stammt, darf man nicht vergessen. Bouset war ein damals in England beliebtes Getränk. Ayrer scheint zwar die Welt das Karnevals zu entwickeln, diese wurzelte aber eigentlich tiefer noch in den Stoffen der englischen Komödianten. Die Hauptsache ist, daß Ayrer die Komik auf eine moralische Lehre, die er aus seiner alltäglichen Weltanschauung bezog, reduzierte und diese wie in folgendem Beispiel äußerte:<br>Also erfehrt mancher mit schaden, <br>Der tracht nach grossem stand und ehrn, <br>Darinn sich thut sein vnglück mehrn.<br>Ayrer meinte, ein Bauer solle ewig ein Bauer bleiben und dieser dürfe nicht nach einem andern, höhern Stand streben. Kein Zweifel, daß Ayrer nicht nur die wahre Funktion des Narren, sondern auch darüber hinaus die der Fastnacht nicht richtig verstand. Wenn er den Nutzen der Narrheit erkannt und seinen Blick auf die Ambivalent des Narren gerichtet hätte, hätte er durch den Narren die verkehrte Welt, die für die Fastnacht sehr bezeichnend ist, verwirklichen und darstellen können.<br>Seine einzige Methode ist die der Nachahmung, da er keine eigene Sprache besitzt. In diesem Sinne hat G. Höfer recht: _"Ayrer hat keine Sprache. An Stelle eines Sprachausdrucks steht bei ihm der bloße dürre Bericht." Mit juristischen Augen steht er alles an, was um ihn herum vorgeht. Von Haus aus fehlte es Ayrer an Auffassungsvermögen, vor allem an Talent für die Dichtung überhaupt. Ayrer hatte zudem keinen einzigen Nachfolger, so daß mit ihm das Fastnachtspiel dem Ende zugehen mußte.

2013 年の調査では日本の高校生の理科の有用感は米中韓と比較し低いが,一方で天文や宇宙開発への関心は高い。そこで,これらと関連し,理科の有用感も増す可能性のある教材として人工衛星電波受信実験に着目した。 受信実験の中では万有引力による運動やドップラー効果など高校物理で学ぶ多くの内容がわかりやすい形で現れ,物理や数学と科学技術との繋がりを実感できる。我々は科学教室と高校授業で実践を行い,理科の有用感と物理内容の理解の向上に対する効果を検証した。結果,多様な学生を対象とする高校授業の実践において効果が確認できた。

<p></p><p>&emsp;Loudspeaker sounds, both announcements for army doctors and radio programs, are frequently inserted in Robert Altman's film <i>M*A*S*H</i> (1970). Accompanied by close-up shots of a loudspeaker, these sounds are undoubtedly diegetic; that is, they can be heard by the characters. However, none of the characters react to them, and the announcer is never seen. These sounds are located in a domain that cannot be grasped by the binary opposition of the diegetic and the non-diegetic.</p><p></p><p>&emsp;This paper considers that while the loudspeaker sounds are backgrounded within diegesis, they are simultaneously foregrounded outside of the diegesis to which the spectator belongs. This activity, the collecting sounds ignored by the characters and manipulating them for emphasis, is due to the "cinematic narrator."</p><p></p><p>&emsp;This cinematic narrator's activity is mimicked by the protagonists; they eavesdrop on and broadcast a sexual act of Houlihan and Burns. By sharing narrative authority with the cinematic narrator, they create and narrate an episode, which retells Houlihan as "Hot Lips." This paper demonstrates that Hot Lips' story, narrated by the protagonists, merges into the whole story of the cinematic narrator. Thus, two different narrative levels coexist without formal boundaries, as in Mikhail Bakhtin's concept of "hybrid utterance."</p><p></p>
