The degradation behavior of three benzodiazepines (BZPs)—lormetazepam (LMZ), lorazepam, and oxazepam—with hydroxy groups on the diazepine ring in artificial gastric juice and the effect of storage pH conditions on drug degradability were monitored using an LC/photodiode array detector (PDA) to estimate their pharmacokinetics in the stomach. Although the three BZPs degraded in artificial gastric juice, none could be restored, despite increasing the storage pH, implying that the degradation reaction was irreversible. As for LMZ, we discussed the physicochemical parameters, such as the activation energy and activation entropy involved in the degradation reaction as well as the reaction kinetics; one of the degradation products was isolated and purified for structural analysis. In the LMZ degradation experiment, peaks corresponding to degradation products, (A) and (B), were detected through the LC/PDA measurements. Regarding the degradation behavior, we hypothesized that LMZ was degraded into (B) via (A), where (A) was an intermediate and (B) was the final product. Although the isolation of degradation product (A) was challenging, degradation product (B) could be isolated and was confirmed to be “methanone, [5-chloro-2-(methylamino)phenyl](2-chlorophenyl)-” based on structure determination using various instrumental analyses. The compound exhibited axis asymmetry as determined using single-crystal X-ray structure analysis. Because the formation of degradation product (B) was irreversible, it would be prudent to target the final degradation product (B) and LMZ for identification when detecting LMZ in human stomach contents, such as during forensic dissection.

Crystal structures of the Ln2Zr2O7 (Ln = Nd and La) were refined by the Rietveld analysis of powder neutron diffraction (ND) data at room temperature. La2Zr2O7 had a pyrochlore (P)-type structure of almost completely ordered for oxygen vacancy. On the other hand, Nd2Zr2O7 had the P-type structure for the oxygen vacancy distributed between the O2 (8b) and O3 (48f) sites. The Rietveld refinement results of powder X-ray diffraction (XRD) data of Ln2Zr2O7 (Ln = Eu, Nd and La) indicated that the Ln3+ and Zr4+ ions exist in ordered states in 16c and 16d sites, respectively. The refined lattice parameters (a), occupancies (g) for oxygen sites (O1, O2 and O3) and x-positional parameters for O3 (48f) site of La2Zr2O7 and Nd2Zr2O7 by using powder XRD data were in almost agreement with those by using powder ND data. The refined ND and XRD data of Ln2Zr2O7 (Ln = Eu, Nd and La) system revealed that the O3 (48f) site occupancy (g48f) decreases with increasing O2 (8b) site occupancy, while O1 (8a) site occupancy keeping 1.0. The change in oxide-ion conductivity (σ) in Ln2Zr2O7 (Ln = Eu, Nd and La) system can be related to a change in the product of site occupancy (g48f) and the oxygen vacancy rate (1-g48f) in the O3 (48f) site, g48f(1-g48f).

Objectives: The Gunma score is used to predict the severity of Kawasaki disease (KD), including coronary artery aneurysm (CAA) as a cardiac complication, in Japan. Additionally, the characteristic ratio of ventricular repolarization (T-peak to T-end interval to QT interval [Tp-e/QT]) on a surface electrocardiogram reflects myocardial inflammation. This study aimed to determine whether the Tp-e/QT can be used to predict CAA in children with KD.Methods: We analyzed chest surface electrocardiograms of 112 children with KD before receiving intravenous immunoglobulin therapy using available software (QTD; Fukuda Denshi, Tokyo, Japan).Results: The Tp-e/QT (lead V5) was positively correlated with the Gunma score (r=0.352, p<0.001). The Tp-e/QT was larger in patients with CAA (residual CAA at 1 month after onset) than in those without CAA (0.314±0.026 versus 0.253±0.044, p=0.003). A receiver operating characteristic curve analysis was performed to assess whether the Gunma score and Tp-e/QT could predict subsequent CAA. The area under the curve of the Gunma score was 0.719 with the cutoff set at 5 points. The area under the curve of the Tp-e/QT was 0.892 with a cutoff value of 0.299. The fit of the prediction models to the observed probability was tested by the Hosmer–Lemeshow test with calibration plots using Locally weighted scatterplot smoothing (LOESS) fit. The Gunma score (p=0.95) and Tp-e/QT (p=0.95) showed a good fit.Conclusions: The Tp-e/QT is a useful biomarker in predicting coronary aneurysm complications in KD.

This study is to investigate and clarify the details about mineralogical characteristics of iron sand used for Japanese classical iron-making furnace “Tatara”, that produces different kinds of iron product, natural steel and cast iron. Though the product difference is known experientially to be attributed to the difference of iron sand kind “Masa” and “Akome”, scientific reason is vague and their characterization is necessary. Specimen was prepared by separating the particles with the similar character by sieving and magnetic separation. Chemical analysis, XRD, mineral texture, EPMA, chromaticity and diffusive reflectance, Raman spectroscopy and specific gravity clarified that main difference between Masa and Akome is in the composition of particles mixed with different mineral characters in term of solid solution TiO2% in iron oxide and quantity of hematite that is weathered magnetite. CO gas reduction test showed that such mineral characters affect the initial temperature of reduction and mineral kinds formed on the way of reduction.

Aims: MND-2119 is a novel once-daily dose self-emulsifying formulation of highly purified eicosapentaenoic acid ethyl ester (EPA-E) and is approved as an antihyperlipidemia agent in Japan. It has improved absorption and achieves higher plasma EPA concentrations at Cmax than conventional EPA-E. In the JELIS trial, concomitant use of EPA-E with statin therapy significantly reduced atherosclerotic cardiovascular disease (ASCVD) risks. As a potential mechanism of action of EPA, endogenous formation of EPA-derived anti-inflammatory metabolites is receiving greater attention. This study aims to investigate the endogenous formation of EPA-derived anti-inflammatory metabolites following single and multiple administrations of MND-2119. Methods: Healthy adult male subjects were randomly assigned to a nonintervention (control) group, MND-2119 2-g/day group, MND-2119 4-g/day group, or EPA-E 1.8-g/day group for 7 days (N=8 per group). Plasma fatty acids and EPA-derived metabolites were evaluated. Peripheral blood neutrophils were isolated, and the production of EPA-derived metabolites from in vitro stimulated neutrophils was evaluated. Results: After single and multiple administrations of MND-2119 2 g/day, there were significant increases in plasma EPA concentration, 18-hydroxyeicosapentaenoic acid (18-HEPE), and 17,18-epoxyeicosatetraenoic acid compared with those of EPA-E 1.8 g/day. They were further increased with MND-2119 4 g/day administration. In neutrophils, the EPA concentration in the MND-2119 2-g/day group was significantly higher compared with that in the EPA-E 1.8-g/day group after multiple administration, and 18-HEPE production was positively correlated with EPA concentration. No safety issues were noted. Conclusions: These results demonstrate that MND-2119 increases the plasma and cellular concentrations of EPA and EPA-derived metabolites to a greater extent than conventional EPA-E formulations.

Background: Recent innovations in information and communication technology have made it possible to assess diet using web-based methods; however, their applicability in the general population remains unclear. Hence, we aimed to examine the applicability of a web-based 24-hour dietary recall (24HR) tool to large-scale epidemiological studies by determining the sampling rate and characteristics of randomly selected participants from a Japanese cohort study.Methods: In total, 5,013 individuals were recruited from a cohort of 21,537 individuals, and 975 agreed to participate in this study. The participants selected either self-administered web-based dietary 24HR (self-administered 24HR) or interviewer-administered telephone-based 24HR (interviewer-administered 24HR) as the method for the dietary assessment and answered questions regarding the acceptability of the system.Results: The response rate of the 975 participants was 19.4%, corresponding to approximately 4.5% of the total study sample. About half of them chose the self-administered 24HR (46.9%). The median time required for the self-administered and interviewer-administered 24HR was 25 and 27 minutes, respectively. In the self-administered 24HR, older people, regardless of sex, tended to require a longer time, and approximately 60% of the participants rated the ease of use of the system as “somewhat difficult” or “difficult.”Conclusion: Characteristics of the participants in this study were not systemically different from those of the entire study sample. Improvements in the approach to entering cooking details and the dish name selection may be necessary for better acceptability in order to be accepted as a self-administered dietary recall tool.

Aim: We investigated the relationship between small dense low-density cholesterol (sdLDL-C) and risk of major adverse cardiovascular events (MACE) in patients treated with high- or low-dose statin therapy.Methods: This was a prospective case-cohort study within the Randomized Evaluation of Aggressive or Moderate Lipid-Lowering Therapy with Pitavastatin in Coronary Artery Disease (REAL-CAD) study, a randomized trial of high- or low-dose (4 or 1 mg/d pitavastatin, respectively) statin therapy, in patients with stable coronary artery disease (CAD). Serum sdLDL-C was determined using an automated homogenous assay at baseline (randomization after a rule-in period, >1 month with 1 mg/d pitavastatin) and 6 months after randomization, in 497 MACE cases, and 1543 participants randomly selected from the REAL-CAD study population.Results: High-dose pitavastatin reduced sdLDL-C by 20% than low-dose pitavastatin (p for interaction <0.001). Among patients receiving low-dose pitavastatin, baseline sdLDL-C demonstrated higher MACE risk independent of LDL-C (hazard ratio [95% confidence interval], 4th versus 1st quartile, 1.67 [1.04–2.68]; p for trend=0.034). High-dose (versus low-dose) pitavastatin reduced MACE risk by 46% in patients in the highest baseline sdLDL-C quartile (>34.3 mg/dL; 0.54 [0.36–0.81]; p=0.003), but increased relative risk by 40% in patients with 1st quartile (≤ 19.5 mg/dL; 1.40 [0.94–2.09]; p=0.099) and did not alter risk in those in 2nd and 3rd quartiles (p for interaction=0.002).Conclusions: These findings associate sdLDL-C and cardiovascular risk, independent of LDL-C, in statin-treated CAD patients. Notably, high-dose statin therapy reduces this risk in those with the highest baseline sdLDL-C.

I found out that several of them considered their listing as co-authors to be inappropriate. In addition, more than half of the co-authors agreed to the retraction of this paper.These facts raise concerns about the paper. From the standpoint of maintaining the integrity of the research community, I felt that such a paper should be retracted at once.Accordingly, I sent a summary of my concerns about this paper to the corresponding author, Dr. Shin Wakui. I also had an online interview with him to discuss this matter. I told Dr. Wakui that inappropriate authorship on a non-negligible scale is a serious problem that raises concerns about the paper.I prepared a draft of this Editor’s Announcement and sent it to Dr. Wakui for review prior to revision and release. Brassicaceae vegetables contain glucosinolate (GSL), which is well known for its role in the plant defense system, bitter substances, and functional components. This study reported the GSL profiles of 18 GSLs in 14 Brassicaceae vegetables (kale, cabbage, kohlrabi, broccoli, cauliflower, komatsuna, pak choi, turnip, choy sum, Chinese cabbage, rutabaga, leaf mustard, leaf radish, and radish root) consisting of 64 cultivars grown in Japan. The GSL compositions showed specific trends among each species, and furthermore, there was considerable variation in the total and individual GSL contents among the cultivars and tissues of the same vegetable. Because each vegetable was grown under the same conditions, this variation was expected to primarily be due to the genetic background. It is desirable to collect additional data from commonly distributed cultivars to conduct more detailed studies on GSLs in Brassicaceae vegetables, such as investigating a negative correlation between GSL consumption and cancer risk.

Food extracts from spices and teas were examined for the taste altering ability on sweet, umami and bitter tastes by using the taste sensation test. The idea arose based upon the findings that 1) some food extracts from spices and teas act as salt taste enhancers and 2) the enhancing effect is directly proportional to glutamate decarboxylase 67 (GAD67) activity in vitro. Our results indicated that those extracts significantly activated or inhibited GAD67 enzyme activity were able to alter sweet, umami, and bitter tastes. However, there were no relationships between the taste potency of each of the examined extracts and the GAD67 relative activity ratio. While it is unlikely that the extracts have directly activated GAD67 activity and enhanced sweet, umami or bitter tastes, the results do not exclude the idea that GABA may participate in the taste cell-to-cell communications.

Lipid oxidation in food can result in the generation of off-odor compounds. We investigated the combined effects of α-tocopherol with rosmarinic acid and various antioxidants on the prevention and inhibition of off-odor formation. We added these combined compounds in a linoleic acid emulsion, followed by an oxidation reaction with 2'2-Azobis (2-methylpropionamidine) Dihydrochloride at 37 ℃ for 5 h. The hydroperoxide was measured by the ferric thiocyanate method, and the off-odor components were measured by gas chromatography. Median effect analysis was used to assess the combined effects. The results showed that the combinations of α-tocopherol with rosmarinic acid and caffeic acid exhibited a synergistic effect on inhibition of lipid peroxidation and some off-odor components, likely attributable to the catechol structure in these compounds. Interestingly, the combination of α-tocopherol and rosmarinic acid showed a potent synergistic effect on (E)-2-nonenal and (E)-2-octenal, indicating that the formation process of off-odor components may influence the combined effect.

The mutant allele frequency of the Pyruvate kinase (PK) gene has been investigated mostly in pure breed cats. We investigated the PK mutant gene in stray and animal hoarding mongrel cats in Hokkaido, Japan. We also investigated the kinship of individuals carrying the mutant gene. Genotyping was conducted using the previously reported real-time PCR method. Fourteen microsatellite markers were used to identify the parents and offspring of cats carrying the PK mutant gene, and some kinship such as parent-offspring and siblings was observed. Some stray and animal hoarding cats carried the PK mutation gene and that consanguinity was confirmed among these cats indicated that the PK mutation gene was spread by unregulated interbreeding.

目的:本研究の目的は,不眠症の疑いのある労働者が就寝時に「快眠音」を聞くことにより,睡眠潜時(寝つくまでの時間)が短縮するのかを検証することである。方法:研究デザインは,個々の研究対象者が「介入群(快眠音)」と「対照群(無音)」をもつランダム化比較試験とした。快眠音システムは音源内蔵スピーカー(ヤマハ社製ISX-80)とベッドマット下生体センサー(EMFIT社製EMFIT-QS)を用いた。日本企業4社の従業員1,185名を対象として事前にアテネ不眠尺度を用いてスクリーニングを行い,531名より回答を得た。不眠症の疑いのある6点以上の162名を抽出し,研究同意・データが得られた42名を対象に分析を行った。データ収集方法は,対象者が自宅に設置した快眠音システムを用いて就寝時にランダムに「快眠音」と「無音」を聞き,それぞれ平日5晩ずつ計10晩の睡眠潜時,睡眠時間,睡眠効率のデータを収集した。睡眠潜時のデータを主要評価項目とし,同様に睡眠時間および睡眠効率のデータを副次評価項目とした。分析方法は,「快眠音」と「無音」での対応のあるt検定を行った。結果:睡眠潜時,睡眠時間,睡眠効率すべての評価項目において「快眠音」と「無音」で有意な差は認められなかった。結論:「快眠音」は不眠症の疑いのある労働者に対する睡眠潜時の短縮効果は得られなかった。

Background: Total work-family conflicts (TWFCs) could associate with mental health, and having ikigai (a purpose of life) may mediate this association.Methods: In a cross-cultural study of 4,792 Japanese Aichi Workers’ Cohort study participants and 3,109 Egyptian civil workers, the Midlife Development in the United States (MIDUS) questionnaire measured TWFCs and the Center for Epidemiological Studies Depression (CES-D) 11-item scale measured depression. We used logistic regression models to estimate odds ratios (ORs) of having depression and a high-ikigai across levels of TWFCs (low, moderate, and high), and the PROCESS macro of Hayes to test the mediation effect.Results: The prevalence of high TWFCs, depression, and having a high ikigai were 17.9%, 39.4%, and 70.1% in Japanese women, 10.5%, 26.8%, and 70.1% in Japanese men, 23.7%, 58.2%, and 24.7% in Egyptian women, and 19.1%, 38.9%, and 36.9% in Egyptian men, respectively. Compared with participants with low TWFCs, the multivariable ORs of depression in Japanese women and men with high TWFCs were 4.11 (95% confidence interval [CI], 2.99–5.65) and 5.42 (95% CI, 4.18–7.02), and those in Egyptian women and men were 4.43 (95% CI, 3.30–5.95) and 4.79 (95% CI, 3.53–6.48), respectively. The respective ORs of having a high-ikigai were 0.46 (95% CI, 0.33–0.64) and 0.40 (95% CI, 0.31–0.52) in Japanese women and men and were 0.34 (95% CI, 0.24–0.48) and 0.28 (95% CI, 0.20–0.39) in Egyptian women and men. No interaction between TWFCs and country was observed for the associations with depression or ikigai. Ikigai mediated (up to 18%) the associations between the TWFCs and depression, especially in Egyptian civil workers.Conclusion: TWFCs were associated with depression, and having low ikigai mediated these associations in Japanese and Egyptian civil workers.

However, the number of small molecules known to affect the cellular spliceosome is currently limited probably because of the lack of a robust cell-based approach to identify small molecules that target the spliceosome. We have previously reported the development of a genetic reporter to detect the cellular levels of small nuclear ribonucleoproteins (snRNPs), which are subunits of the spliceosome, using a split luciferase. However, the original protocol was designed for small scale experiments and was not suitable for compound screening. Here, we found that the use of cell lysis buffer used in blue native polyacrylamide gel electrophoresis (BN-PAGE) dramatically improved the sensitivity and the robustness of the assay. Improved assay conditions were used to discover a small molecule that altered the reporter activity. Our method may be used with other cellular macromolecular complexes and may assist in the discovery of small bioactive molecules.

日本人の宗教観や宗教意識を表現する際に,しばしば使われるのが「無宗教」や「無神論」といった表現である。本稿では,「日本人=無宗教」あるいは「日本人=無神論」という言説が生まれた背景を探るとともに,日本人が無宗教・無神論であるのかについて検討する。そして,それを踏まえて,日本宗教と異文化における宗教との比較研究を深化させる視点や,グローバル化状況における日本宗教を理解する視点を提示したい。

Based on molecular phylogenetic evidence, phylogenetic analyses of both cpDNA and nrDNA sequences based on 76 (cp) and 77 (nr) samples representing 64 species, two subspecies, and three (cp) and two (nr) varieties of Lilium and six (cp) and five (nr) samples from Cardiocrinum, Fritillaria, and Notholirion as outgroups were used to better understand relationships within Lilium and to revise its infrageneric classification. The same individuals were used for both cpDNA and nrDNA analyses to avoid confusion caused by genetic polymorphism within a species. Nine thousand eight hundred thirty seven bp from 10 cpDNA regions and 1069 bp from the nuclear (ITS + ETS) regions were analyzed. The same twelve clades were formed in Lilium in both the chloroplast and nuclear trees (one clade of the former tree and one clade of the latter tree were not strictly formed because of low resolution). In light of the cpDNA and nrDNA trees obtained, 13 morphological characters, including a pair of marginal ridges along a central groove on the adaxial surface of the tepals were examined with attention given for the first time to the use of this character as having diagnostic value. The morphological evidence supported the twelve clades. Based on molecular and morphological evidence, a revised infrageneric classification of Lilium recognizing twelve clades as twelve sections is proposed.

We report a case of atypical slow-slow atrioventricular nodal reentrant tachycardia (AVNRT) utilizing a superior slow pathway as a retrograde limb. The standard electrophysiological criteria confirm the diagnosis of this AVNRT by successfully excluding a diagnosis of atrial tachycardia and atrioventricular reentrant tachycardia. The earliest atrial activation during tachycardia was found at the interatrial septum 17.5 mm superior to the site identified during retrograde conduction with the fast pathway. The tachycardia was not inducible after ablation at the right posterior septum, consistent with successful ablation of the typical slow pathway.

[Purpose] This study aimed to determine whether certain research activities improve the attitude of rehabilitation professionals towards evidence-based practice and its implementation in Japan. [Participants and Methods] We included physical, occupational, and speech therapists currently working in clinical settings. We employed hierarchical multiple regression analyses to assess the attitude of rehabilitation professionals towards evidence-based practice and research activities. Scores of the five dimensions of the Health Sciences-Evidence Based Practice questionnaire were considered the dependent variables. The five dimensions were as follows: Dimension 1, attitude towards evidence-based practice; Dimensions 2–4, evidence-based practice implementation; and Dimension 5, work environment related to evidence-based practice barriers–facilitators. The four sociodemographic variables (gender, academic degree, clinical experience, and the number of therapists at work) were initially included, following which self-reported research achievements were supplemented as independent variables (the number of case studies, literature reviews, cross-sectional studies, and longitudinal studies). [Results] We analyzed data from 167 participants. In addition to sociodemographic variables, the research achievements that statistically increased F-values of the modeling were case study achievements in Dimensions 2–3, cross-sectional study achievements in Dimensions 2 and 4, and longitudinal study achievements in Dimension 5. [Conclusion] Case studies and cross-sectional studies could improve evidence-based practice implementation among rehabilitation professionals in Japan.
