神経根ブロック,硬膜外ブロックおよびブロックを行わずに治療を行った例の3群間の治療成績の差異を明らかにすることを第一の目的とした.第二には,神経根ブロックにおけるステロイド使用の意義を明らかにすることを目的に,椎間板ヘルニアに対する神経根ブロックの治療効果をステロイド使用群と非使用群とで比較した.腰椎椎間板ヘルニアに対するブロック療法群と非ブロック療法群の成績は,治療後6カ月の時点ではブロック療法群の方が有意に成績が良かった.しかし,治療後2年では,両者の差はなかった.ブロック群では,症状軽快または消失までの期間が非ブロック群に比べ有意に短かった.また神経根ブロック群の方が硬膜外ブロック群よりも有症状期間をさらに短縮していた.神経根ブロックにおけるステロイド使用の意義は,椎間板ヘルニアでは少なかった.

2005年から2010年にかけ、苫小牧市および別海町で発見されたオオハクチョウ3例について、酪農学園大学野生動物医学センターで剖検を行った。3例とも栄養状態は良好であり、外傷や外貌の汚れは認めなかった。内部所見としては暗赤色流動性の血液、心臓周辺を主とした各臓器および血管の鬱血、口腔および気管粘膜の溢血点、水・唾液などの液状成分により著しく膨化した食パンによる喉頭部閉塞が共通所見として認められた。喉頭閉塞では神経系の反射的心停止が知られることから、このような現象も今回の死因の一部と推察された。

近年, 摂食障害と発達障害との関連が指摘されている. 今回, 発達障害の合併が疑われ, 知能検査の施行が治療方針変更の良いきっかけとなった1例を経験したので報告する. 症例は32歳女性. X−21年に過食を開始し, 過食, 自己誘発性嘔吐や食事制限, 下剤の乱用により, 体重は大きく変動した. X−6年に神経性過食症と診断され, 入退院を繰り返した. X年に2型糖尿病に伴う血糖コントロールの悪化をきっかけに食事量が著明に低下し, 1日数十回の嘔吐を認め, 当科第11回入院となった. 生育歴やこれまでの経過から, 何らかの発達障害の合併が疑われたため, ウェクスラー成人知能検査を施行した. その結果, 動作性IQが言語性IQに比して有意に低値であり, 注意欠陥多動性障害を疑う所見も認められた. 退院後atomoxetineを開始したところ, 過食・嘔吐の頻度が週に1, 2回程度に減少し, その後も安定した状態を維持している.

我々はクローン動物および体細胞由来ntES細胞を効率よく作出する方法の開発および初期化のメカニズムの解明を試みている。昨年我々はntES細胞と受精卵由来のES細胞には違いが見られないことを証明したが、19年度はさらに詳しい比較と倫理問題の解決を試みた。我々は核移植の影響を調べるために、単為発生ES細胞を用いた遺伝子発現解析法を確立し、これによってついに我々は特定の遺伝子におけるメチル化状態について、ntES細胞とES細胞の間に違いがあることを発見した。現在このntES細胞特異的なDNAメチル化状態の違いが、その後の細胞分化や応用にどのような影響を与えるのか解析中である(Hikichi, et. al., 2007,2008)。一方倫理問題の解決については2つのアプローチを試みた。体外受精に失敗した卵子は、加齢により質が低下することから廃棄されている。この廃棄卵子を用いれば健康な女性から卵子を提供してもらうという倫理問題は解決する。そこでマウスを用いたモデル実験を行った結果、体外受精に失敗した卵子でも核移植およびntES細胞の作成には問題ないことを明らかにした(Wakayama, et. al., 2007a)。この成果は多数の全国紙やテレビで紹介された。また、胚を殺さずに1割球(細胞)を取り出し、そこからES細胞を樹立する手法を開発し、生命の萌芽である胚を壊すという問題についても回避できることを明らかにした(Wakayama, et. al., 2007b)。他に核移植技術をより簡素化した手法についての研究も報告した(Kishigami, et. al., 2007;Wakayama, et. al., 2008)。

核移植技術では核そのものを操作するが、それよりも小さな単位、すなわち染色体を操作するための技術開発を試みた。特定の染色体をラベルするためsuntag systemを使用し、ES細胞中のY染色体をラベルすることに成功し任意の染色体をラベルできる可能性を示した。また胚の染色体を分散させることで、生きた胚間での染色体移植が可能であることを示した。しかしながら、胚において特定染色体をラベルして操作するには至らなかった。加えて、染色体操作の使用が想定される絶滅危惧種において、個体を傷つけずに採取可能な尿中に含まれる極わずかな細胞から、クローン個体およびntES細胞株が樹立可能であることを示した。

世界初のクローンヒツジ「ドリー」の報告以来、クローン動物の作出効率は動物種によらずわずか数%と低率であった。本研究代表者は、クローン胚をヒストン脱アセチル化酵素阻害剤(HDACi)、トリコスタチンA(trichostatin A, TSA)で処理することでクローンマウスの作出効率が大幅に改善されることを発見した。本課題ではその作出効率改善の機構解明として発生におけるHDAC酵素やタンパク質アセチル化の重要性を明らかにし、さらにこれまで不可能であったマウス系統からのクローンマウスの作出に世界で初めて成功した。

This paper addresses position and path-tracking problems of robot manipulators subject to constraints on both the magnitude and difference of the control input. Along with the model predictive control (MPC), the disturbance observer (DOB) is used to compensate for disturbances and nominalize the plant dynamics. To constrain the total control input, which is the sum of the MPC output and DOB output, we adopt time-varying input constraints and construct a useful structure of the combined DOB-MPC system. The effectiveness of the proposed control scheme is validated through a few experiments using a real two-link manipulator.

【要旨】広汎性発達障害、注意欠陥多動性障害、学習障害、発達性言語障害などの内、知能障害が無いか軽いものを軽度発達障害と呼ぶ。発達障害は基本的には行動特徴によって定義されている。したがって、詳細な行動の観察や病歴の聴取が重要であり、幅広く情報を収集する必要がある。診断の補助ツールとして種々の質問紙や構造化面接法が考案されているが、日本語に翻訳された感受性と特異性が高いものはない。多くの患者では何らかの共存症を認めることに留意する必要がある。診断には必須ではないが、療育や教育の計画に神経心理検査は有用である。障害毎のWISC-III得点プロフィールの特徴はある程度は認められるが、個人差が大きい。

A general method for the stereospecific synthesis of 2, 3-trans-3, 4-cis-trisubstituted cyclopentanones (9) is described. This synthetic method has the advantage that three functional groups can be stereospecifically introduced on a five-membered ring by the catalytic hydrogenation of 2, 3, 4-trisubstituted cyclopentenones. Furthermore, 9 could be stereospecifically converted to 1, 2-trans-2, 3-trans-3, 4-cis-trisubstituted cyclopentanols (20) and 3, 4-cis-disubstituted cyclopentanones (11) by a simple procedure. These synthetic methods may be useful for the synthesis of natural products containing a five-membered ring.

Variously functionalized cyclopentanones are important as starting materials for the synthesis of natural products. We have succeeded in a simple, highly stereospecific synthesis of five membered ring ketone using Wilkinson complex. Thus, the precursors, 1-al-4-enes(4a-e,11,13) prepared in optically active form from D-limonene, (+)-limonen-10-ol and 1-carvone in three steps were submitted for RhCl(PPh_3)_3-catalyzed cyclization in CH_2Cl_2 to afford the cis-3,4-disubstituted cyclopentanones(14-22) in good yield (Table 2). The following results were obtained in this cyclization. 1) In all cases, the cis-3,4-disubstituted cyclopentanones were stereospecifically obtained as the sole products. 2) The cyclization reaction of the six membered lactols(12,13) was found to proceed very slowly even if at high temperature. 3) The cis-3,4-disubstituted cyclopentanones obtained in this method were also found to be applicable for the synthesis of bicyclic ketone(23). 4) The configurations of 3,4-disubstituents in each compounds were clarified by the way of chemical correlation and spectral analyses. Further application for the synthesis of ent-cis,cis-dihydro-nepetalactone(39) will be discussed.

In the synthesis of natural products consisting of the basic structure of cyclopentane ring, variously functionalized cyclopentanones are required as starting materials. Previously, we succeeded in a simple, highly stereospecific synthesis of the cis-3,4-disubstituted cyclopentanones by the mild reaction with RhCl(PPh_3)_3. By the application of this newer cyclization reaction, we have succeeded in the synthesis of cis,cis-dihydronepetalactone, prostanoic acid, 8-isoprostanoic acid, (+)-brefeldin A and carbacyclin in the optically active form from limonene or Corey lactone. These compounds were synthesized via the following key steps. 1) 3-(3-Oxobutyl)cyclopentanone was easily converted to the deconjugated bicyclic enone (3→4). 2) The regioselective alkylation of limonene was performed by using s-BuLi-TMEDA (1→8). 3) The cis-3,4-disubstituted cyclopentanone from limonen-10-ol could be converted to the trans-3,4-disubstituted cyclo-pentanone by the epimerization (15→16, 22→23).

While quinoline 1-oxide itself is not capable to react with acetyl cyanide, an exothermic reaction occurred in the presence of acetic anhydride, and quinaldonitrile and 2-quinolinepyruvonitrile were obtained. The former was a sole product from the reaction in dioxane or chloroform, whereas the latter was the main product without solvent.

Six kinds of 2-aryloxazole N-oxide derivatives were prepared, and their reactions with phosphoryl chloride and acetic anhydride were studied. Methyl group located on the 4-position of 2-aryloxazole N-oxide was easily subjective to nucleophilic attack as an active methyl group in the reaction with phosphoryl chloride or acetic anhydride, whereas 5-methyl group was inert to such an attack.
