The enzyme cycloisomaltooligosaccharide glucanotransferase (CITase) was isolated from Bacillus circulans U-155, which had a yield of 25.8%, and purified to homogeneity. The Mr was approximately 98,000, similar to that for all known CITases. Specific activity of the purified enzyme was 2.11 U/mg protein, with maximal activity at approximately pH 6.0. The enzyme was stable at pH 4.5 up to pH 9.0 and at temperatures up to 50°C. The main product of the initial enzyme reaction was cycloisomalto-heptaose. The cit gene has a 2,895-bp open reading frame and encodes CITase in B. circulans U-155. We cloned this gene into a recombinant plasmid pCI811 and expressed it in Escherichia coli. A comparison between DNA sequence data from the transformant and the N-terminal amino acid sequence of the purified enzyme from B. circulans U-155 suggested that CITase was translated as a secretory precursor with a 30-amino-acid signal peptide. The mature enzyme contained 934 residues with a predicted molecular mass of 103.93 kDa. The enzyme activity in the transformants was approximately 3.0 mU/mL, similar to that of the purified enzyme secreted by B. circulans U-155. The enzyme also showed 72 and 67% identity with CITase from Paenibacillus sp. 598K and B. circulans T-3040, respectively. These results suggest that the enzyme isolated from B. circulans U-155 shares a greater similarity with CITase expressed by Paenibacillus sp. 598K than B. circulans T-3040.

本研究の目的は,閉眼および開眼片足立ち測定,Multi-Directional Reach Testの3評価結果における転倒群と非転倒群の比較,転倒回数との関連性および3評価結果間の相関について調べ,高齢者の転倒要因とされるバランス能力をどのような視点から捉えるべきか検討した。結果,いずれの評価結果においても転倒群が有意に少ない値を示し,また,転倒回数が多い者ほど値が少なかった。転倒群では,各評価結果間すべてにおいて有意な相関が認められた。以上より,転倒要因として圧中心保持能力と圧中心偏移能力の両方の視点からバランス能力を捉える必要のあることが示唆された。

Antitumor activity of Sarcodon aspratus (BERK.) S. ITO and Ganoderma lucidum (FR.) KARST. was investigated. Methanol and aqueous extracts of these Japanese mushrooms were tested for antitumor activity against solid type of sarcoma 180 by intraperitoneal or oral administration. The aqueous extract was remarkably effective for inhibition of tumor growth, but the methanol extract was not. The fraction of molecular weight more than 10000 had a high inhibitory activity against the tumor growth, but the fraction of molecular weight less than 10000 did not. Fractionation was carried out by using an ion-exchanger, and fraction S-4 having the highest carbohydrate content had the highest antitumor activity by intraperitoneal administration.

Soybean extracts (SBE) containing isoflavone glycosides were cultured with Ganoderma lucidum mycelia producing β-glucosidase. The anti-angiogenic effects of the cultivated product, containing rich in genistein, named GCP (genistein combined polysaccharide), were assessed with chick chorioallantoic membranes (CAM) and a mouse dorsal air-sac model. β-Glucosidase produced by the mycelia converted the isoflavone glycosides into aglycons. A test of volunteers showed that serum concentrations of genistein in the subjects treated with GCP (n=4) at 3 h after administration were significantly higher than those in the subjects treated with SBE (n=4). GCP inhibited angiogenesis in CAM, and the activity of GCP was greater than that of SBE. GCP inhibited the formation of new vessels induced by colon carcinoma cells in vivo.

Medicinal mushrooms occurring in South India namely Ganoderma lucidum, Phellinus rimosus, Pleurotus florida and Pleurotus pulmonaris possessed profound antioxidant and antitumor activities. This indicated that these mushrooms would be valuable sources of antioxidant and antitumor compounds. Investigations also revealed that they had significant antimutagenic and anticarcinogenic activities. Thus, Indian medicinal mushrooms are potential sources of antioxidant and anticancer compounds. However, intensive and extensive investigations are needed to exploit their valuable therapeutic use.

The effect of ganoderic acid Me (GA-Me), which was purified from the fermentation mycelia of the traditional Chinese medicinal mushroom Ganoderma lucidum as reported (Tang W, Gu TY, Zhong JJ. Biochem Eng J. 2006;32:205–210), on anti-invasion was investigated. Wound healing assay indicated that GA-Me inhibited cell migration of 95-D, a human highly metastatic lung tumor cell line, in dose- and time-dependent manners. Results of cell aggregation and adhesion assays showed that GA-Me promoted cell homotypic aggregation and inhibited cell adherence to extracellular matrix (ECM). In addition, GA-Me suppressed matrix metalloproteinases 2/9 (MMP2/9) gene expressions at both mRNA and protein levels in 95-D cells according to qRT-PCR and Western blotting, respectively. The results demonstrated that GA-Me effectively inhibited tumor invasion, and it might act as a new MMP2/9 inhibitor for anti-metastasis treatment of carcinoma cells.

Alzheimer's disease (AD) is a progressive, degenerative disorder that is characterized by a slow, progressive decline in mental and social function, impairing memory, thinking, reasoning, and the ability to learn. Although there is no cure for AD, the early detection can improve symptoms and slow progression of the disease and also can give patients and their families better care and treatment options. Recently, it has become increasingly important to detect AD at an early stage due to the advent of drugs with slowing progression. Psychologists are expected to develop effective methods for screening to be more sensitive for detecting individuals in the early stages of AD. This article reviewed existing dementia screening tools and examined the roles of psychology in assessment and measurement of cognitive function.

Eye movements have been extensively studied in Neuroscience as a model system to understand neural mechanisms of motor control and learning. Further they have been examined in clinical and psychological studies since neural circuitries involved in eye movements extend to wide brain areas that are also involved in other brain functions such as attention. In this paper, I summarize recent researches in neuroscience that employ eye movements to elucidate neuronal mechanisms of motor learning. Then, an example of application of the accumulated neuroscience evidence to real world engineering problem, namely adaptive robot control, is introduced. Another application of eye movements to monitor car driver's physiological states is also summarized. By showing these recent studies on eye movements, I propose that eye movements can be one of the most attractive model systems to bridge the engineering and basic psychology in harmony.

The history of the Red List of Japanese vascular plants is briefly reviewed for editing and research. Especially on the results of recent monitoring, the present status of information and conservation activities on the endangered plants in Japan is discussed and the dynamics of the Japanese flora are taken up, in relation to basic research on plant biodiversity on the Japanese Archipelago. The figures of endangered plants are not very variable during the past quarter of a century, but we can surmise that the conservation of threatened species in Japan has been promoted to some extent. Based on the results of such a study, proposals are made to contribute to the sustainable use of plant biodiversity on the Japanese Archipelago under a global conspectus.(Communicated by Satohiko SASAKI, M.J.A.)

症例は33歳, 男性. 体重増加(160kg)と著明な労作時息切れを主訴に来院し, 精査加療目的に入院となった. 全身浮腫, 低酸素血症, 高二酸化炭素血症, 総ビリルビン値上昇を認め, 左室拡張機能および収縮機能は低下していたが, 肺水腫や肺うっ血は認めなかったことから, 右心不全優位の肥満低換気症候群(obesity hypoventilation syndrome; OHS)と考えられた. 簡易型終夜ポリグラフ検査にて重症の閉塞型睡眠時無呼吸(obstructive sleep apnea; OSA)を認めたが, 患者は持続陽圧呼吸療法を受け入れることができなかった. 心不全に対して通常の内科治療のみを開始したところ, 心不全は徐々に増悪し, 入院第13病日に多臓器不全を呈した. 心係数低下, 肺動脈楔入圧上昇を認めたため, 両心不全の増悪と判断し, 人工呼吸器管理を行った. 経過中にTorsade de Pointes(TdP)とそれに引き続く心不全の再増悪を認め, 持続緩徐式血液濾過が必要となった. その後, 心不全は徐々に改善し, 体重は77.5kgまで減少し, カルベジロールを開始した. 第211病日にはOSAと心機能は著明に改善した. 現在カルベジロール少量投与にて経過観察中である. 重症心機能障害を合併するOHSでは, 早期より気管切開による陽圧呼吸療法も含めた積極的な加療を考慮すべきである.

Δ9-Tetrahydrocannabinol (Δ9-THC), a biologically active constituent of marijuana, possesses a wide variety of pharmacological and toxicological effects (e.g., analgesia, hypotension, reduction of inflammation, and anti-cancer effects). Among Δ9-THC’s biological activities, its recognized anti-estrogenic activity has been the subject of investigations. Since Δ9-THC is used as both a drug of abuse (marijuana) and as a preventive therapeutic to treat pain and nausea in cancer patients undergoing chemotherapy in the United States and other countries (synthesized Δ9-THC; dronabinol), it is important to investigate the mechanistic basis underlying the anti-estrogenic activity of Δ9-THC. Since Δ9-THC has “no” binding potential for estrogen receptor α (ERα) which can be activated by estrogen (E2), the question of how Δ9-THC exerts its inhibitory effect on ERα is not resolved. We have recently reported that ERβ, a second type of ER, is involved in the Δ9-THC abrogation of E2/ERα-mediated transcriptional activity. Here we discuss the possible mechanism(s) of the Δ9-THC-mediated disruption of E2/ERα signaling by presenting our recent findings as well.

This study aimed to assess the steady-state pharmacokinetics of pranlukast, a leukotriene receptor antagonist, in children with allergic rhinitis and bronchial asthma, and to clarify factors affecting apparent clearance (CL/F). A total of 192 plasma samples were obtained from 98 children (rhinitis 64, asthma 13, complications 21), aged 3-14 years in 2 clinical trials. Plasma concentration of pranlukast was determined by liquid chromatography connected with a tandem mass spectrometer and analyzed by a population approach using NONMEM program. The plasma concentration-time course of pranlukast was described by using a one-compartment model with the first-order absorption and lag time. The robustness of the population pharmacokinetic model was evaluated by using 200 bootstrap samples. The results of population pharmacokinetic analysis showed that only age was a factor affecting the CL/F per body weight, with CL/F decreasing with increasing age. No significant variation was seen in the CL/F between rhinitis and asthma. The interindividual variability in the CL/F and the residual variability were 19.7% and 48.4%, respectively. All the parameters fell within 10% of the bootstrapped mean. In conclusion, the results show that age is the most influential factor for explaining interindividual variability in CL/F, and the difference in diseases does not affect CL/F.

S.; 2) Koriyama Women’s col.; 3) Nagaokakyou City; 4) Abiko City; 5) Mie Univ.) ニセクロハツは子実体に触れたり,傷つけたり,あるいは子実体を切断すると,その部分が赤変したままで,クロハツやクロハツモドキのように赤変後,黒変しないきのこである.リボソームDNAの28SおよびITS領域に基づいたクロハツ節の解析結果から,クロハツは3型に,クロハツモドキは9型に,ニセクロハツは5型にそれぞれ分かれた.ここではニセクロハツの分子系統解析の結果,子実体の巨視的な形態的特徴・顕微鏡的な特徴に関して,報告する. 28Sを用いた分子系統解析ではニセクロハツは大きなグループを作り,遺伝的に異なった5群(型)に分かれた.しかし,ITS領域の解析ではニセクロハツの3型(B-2,B-3,B-4)は同一群に属したが,それ以外の2型(B-1,B-5)は3型と同じ群に属さなかった.タイプ標本を含むB-1はクロハツと同じ群を形成した. 子実体の巨視的な形態観察ではヒダが細かくて柄が長いB-2,ヒダの赤変性が強いB-3など,型間に違いが見られた.顕微鏡的な特徴では胞子の表面構造やカサの表皮の状態が,タイプ標本を含むB-1とB-3との間で明らかに異なった. 以上のことから,分子系統解析で得られた5型間に子実体の巨視的な,あるいは顕微鏡的な特徴の違いが見られたので,B-2やB-3はニセクロハツの別種か,あるいは変種と考えることができる.

Recently, expectations for camera-based document analysis and recognition have increased by improved performance of digital camera devices. In this paper, we propose a rotation angle estimation method using Gray-Scale Gradient Feature and Modified Quadratic Discriminant Function (MQDF). This method can recognize characters and estimate the rotation angle of those characters rapidly. As the result of the evaluation experiment using printed alphanumeric character, we have confirmed that the low dimensional feature vector is sufficient to estimate the rotation angle of characters. Also, we reduced the number of used eigenvectors of the covariance matrix to calculate the MQDF while keeping estimation accuracy.

Accurate off-line character recognition is still very important for camera based printed document analysis. Due to its inherent conceptual and technical simplicity, conventional recognition strategies relied on features extracted using square block zoning of a character image. In this paper, we propose an isotropic feature extraction method using regular hexagonal zoning and empirically confirm its effectiveness for printed and handwritten character recognition. We accomplished printed character recognition and handwritten character recognition experiments using large-scale evaluating datasets. The average accuracy was improved by 2 % in experiments using gradient features. And the effectiveness of hexagonal zoning for recognition of high stroke count characters and low-resolution characters is confirmed in both printed and handwritten character recognition by the experiments.
