BackgroundCRISPR-Cas9 gene-editing technology has facilitated the generation of knockout mice, providing an alternative to cumbersome and time-consuming traditional embryonic stem cell-based methods. An earlier study reported up to 16% efficiency in generating conditional knockout (cKO or floxed) alleles by microinjection of 2 single guide RNAs (sgRNA) and 2 single-stranded oligonucleotides as donors (referred herein as "two-donor floxing" method).ResultsWe re-evaluate the two-donor method from a consortium of 20 laboratories across the world. The dataset constitutes 56 genetic loci, 17,887 zygotes, and 1718 live-born mice, of which only 15 (0.87%) mice contain cKO alleles. We subject the dataset to statistical analyses and a machine learning algorithm, which reveals that none of the factors analyzed was predictive for the success of this method. We test some of the newer methods that use one-donor DNA on 18 loci for which the two-donor approach failed to produce cKO alleles. We find that the one-donor methods are 10- to 20-fold more efficient than the two-donor approach.ConclusionWe propose that the two-donor method lacks efficiency because it relies on two simultaneous recombination events in cis, an outcome that is dwarfed by pervasive accompanying undesired editing events. The methods that use one-donor DNA are fairly efficient as they rely on only one recombination event, and the probability of correct insertion of the donor cassette without unanticipated mutational events is much higher. Therefore, one-donor methods offer higher efficiencies for the routine generation of cKO animal models.

昨年12月下旬、東京ドームシティの「プリズムホール」はリクルートスーツ姿の学生たちで溢れ返っていた。目的は毎日コミュニケーションズ(マイコミ)が主催する日本人の海外留学生向け就職イベントに参加すること。約2900人が会場に詰めかけた。留学生の浦島太郎化を防ぐ 海外で生活する留学生にとり、クリスマス休暇で帰国する時期に就職活動ができるのは魅力的だ。

ナミハグモ属の種多様性には,著しい地理的種分化と,体サイズの分化による同所的な種の組み合わせという2つの側面がある. 日本から70種以上が記録されているが,まだ多くの未記載種が残されている.ナガトナミハグモ種群は生殖器の形態に著しい地理的分化を示し,種内の複雑な地理的変異,環状重複,近縁種の側所的分布や同所的共存といった異所的種分化のさまざまな過程がみられた.また,体サイズは種分化とその後の同所的共存に重要な役割を果たすと考えられ,カチドキナミハグモ種群では体サイズの分化が種分化を促進した可能性がある.

A series of the process of neutrophil extravasation induced by topical application of leukotriene B4 (LTB4) or formyl-methionyl-leucyl-phenylalanine (fMLP) on microvasculature of the hamster cheek pouch can be divided into five steps; I) rolling on the venular endothelium, II) adhesion on the endothelium, III) passage between the endothelial cells, IV) staying in the venular wall, V) migration from the venular wall to the interstitial space. In step II, topical application of chemoattractants on the microvasculature caused an increase in the number of neutrophils adhered to the venules. In step III, the size of the adhered neutrophils became gradually smaller and finallly disappear from the vascular lumen, as they were observed on the monitor screen. Over 80-90% of the adhered neutrophils passed through the endothelial cells. The whole process from step II to III took about 7 minutes. In step IV, the neutrophils which passed through the endothelial cells stay for about 30 minutes in the venular wall. In step V, neutrophils penetrated the basement membrane and migrated into the interstitial space.Step II and III were suppressed by a cyclic AMP phosphodiesterase inhibitor, fibronectin inhibitor and cytochalasin B. From these results, it was suggested that chemoattractant may cause the contraction of microfilament of neutrophil, and the contraction may induce the expression of adhesive glycoprotein on the neutrophil membrane. The increase of cyclic AMP in the neutrophil may inhibit the contraction of microfilament and/or the expression of adhesive glycoprotein of neutrophil. Dexamethasone did not affect the step I to IV, but suppressed the step V. Dexamethasone may inhibit synthesis and/or release of protease (s) in the neutrophil granules which degrade the basement membrane. In fact, the collagenase inhibitor suppressed the step V, although it did not the step I to IV. Thus, neutophil-derived collagenase may act on the passage of neutrophil through the basement membrane. The process of neutrophil extravasation is the chain of reaction.

韓国は日本以上の学歴社会であり,親の子どもに対する教育熱は高く,教育費の増大が社会問題となっている。英語教育に関しては,1997 年に英語が小学校に導入され,それに伴い早期英語教育が過熱化し,所得による教育格差,即ち所得が多い家庭の児童は英語塾に通い,また,早期留学に行くことができるなど親の所得が子どもたちの学校以外での英語学習への参与,さらには英語力に影響を与えるなどの問題が生じてきた。そのため韓国政府では,所得による格差から生まれる英語力の格差を軽減するために,様々な政策を行っており,近年ではその格 差も徐々に軽減されつつある。 一方,日本においても,近年不景気が続き,子どもの教育に十分投資できない家庭も増えており(小林,2008),教育格差ということが徐々に問題になりつつある。ゆえに教育格差を軽減するために様々な対策を講じている韓国の事例から日本は多くのことを学ぶことができると思われる。よって,本稿では最初に韓国の英語教育を所得による格差の観点から考察し,次に韓国政府や自治体がそのような格差をなくすためにどのような対策を行ってきたのかを論じていく。

The point of departure for the present study is that the combination of spatial dispersal and global integration has created a new strategic role for major cities. Beyond their long history as centers for international trade and banking, these cities now function in four new ways: first, as highly concentrated command points in the organizations of the world economy; second, as key locations for finance and for specialized service firms, which have replaced manufacturing as the leading economic sectors; third, as sites of production, including the production of innovations, in these leading industries; and fourth, an markets for the products and innovations produced. These changes in the functioning of cities have had a massive impact upon both international economic activity and unban form: cities concentrate control over vast resources, while finance and specialized service industries have restructured the urban social and economic order. Thus a new type of city has appeared. It is the global city. The first thesis advanced in this book is that the territorial dispersal of current economic activity creates a need for expanded central control and management. But top-level control and management of the industry has become concentrated in a few leading financial centers, especially New York, London, Tokyo, Frankfurt, and Paris. The Fundamental dynamic posited here is that the more globalized the economy becomes, the higher the agglomeration of central functions in a relatively few sites, that is, in global cities. A second major theme of this book concerns the impact of this type of these development of the national urban system in each of these countries and for the relationship of the global city to its nation-state. A third major theme explored in this book concerns the consequences of these developments for the national urban system in each of these countries and for the relationship of the global city to its nation-state. The fourth and final theme in the book concerns the impact of these new forms of, and conditions for, growth on the social order of the global city.

Recently, many cases have been reported of individuals who complain of having halitosis which no one else can smell and for which there is no local or systemic cause. We call this condition “Self-halitosis”. Self-halitosis can be caused by either psychosomatic or psychiatric disorders. We have been treating such patients with brief courses of psychotherapy without using drugs. For patients with psychiatric disorders, we emphasize the importance of psychiatric treatment, but such patients often refuse to be treated by a psychiatrist because they are convinced that they really do have bad breath.In this paper, we report a case of self-halitosis considered to have been induced by skin disease. The patient was a 31-year-old man and his symptoms were relieved by a brief course of psychotherapy.

This study will explore balls at the Imperial Hotel in Tokyo from 1891 to 1926. The first ball was held on November 3, 1891 when the Minister of Foreign Affairs sponsored a gala party at the Imperial Hotel to celebrate the Emperor's birthday. On this occasion, members of Japanese high society and foreign diplomatic officials were invited and they danced the Quadrills, the Waltz, the Caledonian and other dances. This party to celebrate the Emperor's birthday was held at the hotel every year until 1903. The second Imperial Hotel was completed on September 1, 1923 and it was designed by the American architect, Frank Lloyd Wright (1867-1959). The hotel had a beautiful ballroom called the Peacock Room and it became the venue for many balls, such as the St. Valentine's, Christmas, and various charity balls. Partygoers enjoyed dancing the One step, the Two step, and the Foxtrot which were very popular at the time. The Imperial Hotel always produced beautiful and elegant balls. There, it was a special world in Japan, far from everyday life of ordinary Japanese people.
