Background:Uric acid (UA) serves as an antioxidant in vascular endothelial cells. UA transporter 1 (URAT1) encoded by SLC22A12 is expressed in the kidney and vessels and its loss of function causes hypouricemia. The purpose of this study was to examine whether there is any endothelial dysfunction in patients with hypouricemia.Methods and Results:Twenty-six patients with hypouricemia (<2.5 mg/dl) and 13 healthy control subjects were enrolled. Endothelial function was evaluated using flow-mediated dilation (FMD). mRNA of UA transporters expressed in cultured human umbilical endothelial cells (HUVEC) was detected on RT-PCR. There was a positive correlation between FMD and serum UA in the hypouricemia group. URAT1 loss-of-function mutations were found in the genome of 21 of 26 patients with hypouricemia, and not in the other 5. In the hypouricemia groups, serum UA in homozygous and compound heterozygous patients was significantly lower than in other groups, suggesting that severity of URAT1 dysfunction may influence the severity of hypouricemia. Thirteen of 16 hypouricemia subjects with homozygous and compound heterozygote mutations had SUA <0.8 mg/dl and their FMD was lower than in other groups. HUVEC do not express mRNA of URAT1, suggesting the null role of URAT1 in endothelial function.Conclusions:Depletion of UA due to SLC22A12/URAT1 loss-of-function mutations causes endothelial dysfunction in hypouricemia patients.

The study of “traditional” young men's groups (Wakamono-gumi) in Japan has been focussed mainly on the customary institution of young men's lodging house and on the function of marital control, both from the viewpoint of Japanese folklore, and has not analyzed its transformation in relation to village structure. This report considers the traditional groups as the original pattern for new young men's associations (Seinen-dan) since late Meiji, and involves consideration of how the former was recognized, using the example of Izu fishing village. It considers first the traditional group in village structure, and then pursues how it was transformed in relation to inner factors in the village as well as nationwide conditions. The results of the research are as follows : (1) The traditional group was first organized as a substructure of the fisherman's boss group in the fishing village under the feudal system. The Community structure that was organized by the boss group and its fishing workers (the former having special fishing rights and latter not) had reflected its dual character on the traditional young men's group, and there its exclusive character had produced two groups : the one consisting of young fishermen and the other of young merchants and handicraftsmen.(2) Since the middle of Meiji the establishment of public social systems, the reformation of fisheries laws and the development of fishing tecniques relaxed gradually the solidarity of the boss group based on inshore fishing grounds. Next, united body of traditional groups was organized by governmental guidance to new young men's associations on the level of local autonomous entities that generally included several national villages. Therefore the association had dual character : that of the united body to be used for military forces, and that of each traditional group as a branch of body adhering closely to each village system. The coexistence of the two different groups was ensured by the homogeneous gemeinschaftlich character proper to their structure. This coexistence continued essentially to the end of the World War II.(3) After the War (1945) with the dissolution of the united body, the young men's associations started. But since 1955 socio-economical changes have not allowed all young men in the village to be organized as a unit. So there are many different small young men's groups now.The organization principles of young men's groups have experienced the following changes : (a) gemeinschaftlich (b) “gemeinschaftlich-nationalistic”, (c) individualistic. The change of these principles is founded on the structural character of the fishing village in each time.

A marital commitment scale was created to study marital relationships of middle-aged and elderly couples. This study examined the scale’s reliability and validity. A total of 840 middle-aged and elderly married participants ranged from 37 to 89 years completed the marital commitment questionnaire and answered questions regarding marital love, perception of the balance of power between spouses, and the concept of gender roles. The results showed that marital commitment consisted of: personal commitment, resigned and instrumental commitment and normative commitment. Personal commitment was highly correlated with marital love. Resigned and instrumental commitment was correlated with perception of poorly balanced of power between spouses. Normative commitment was correlated with the concept of traditional gender roles. Thus the internal consistency as well as the construct and criterion-related validity of the marital commitment scale were confirmed. Gender differences were also identified. Men scored higher on personal and normative commitment, and women scored higher on resigned and instrumental commitment. The results are discussed in terms of gender.

This study examined whether preference for solitude promotes emotional well-being after controlling for the influence of loneliness. We administered a questionnaire that included variables regarding preference for solitude, loneliness, positive affect, and negative affect. The sample consisted of 318 young adult university students and 253 individuals aged 65 and older. The results showed a significant negative correlation between preference for solitude and positive affect; however there was no correlation between preference for solitude and negative affect. Hierarchical multiple regression analysis confirmed a negative effect of preference for solitude on negative affect after controlling for the influence of loneliness, and there was no effect on positive affect. The results of this study supported the hypothesis concerning the relationship between preference for solitude and negative affect, and demonstrated that preference for solitude decreased negative affect and promotes emotional well-being.

Molecular filtration and purification play important roles for biomolecule analysis. However, it is still necessary to improve efficiency and reduce the filtration time. Here, we show self-assembled nanowire arrays as three-dimensional (3D) nanopores embedded in a microfluidic channel for ultrafast DNA filtration. The 3D nanopore structure was formed by a vapor-liquid-solid (VLS) nanowire growth technique, which allowed us to control pore size of the filtration material by varying the number of growth cycles. λ DNA molecules (48.5 kbp) were filtrated from a mixture of T4 DNA (166 kbp) at the entrance of the 3D nanopore structure within 1 s under an applied electric field. Moreover, we observed single DNA molecule migration of T4 and λ DNA molecules to clarify the filtration mechanism. The 3D nanopore structure has simplicity of fabrication, flexibility of pore size control and reusability for biomolecule filtration. Consequently it is an excellent material for biomolecular filtration.

症例は65歳,女性.1996年にHBV陽性を指摘されIFN治療を受けるも改善無く,その後肝硬変に進展した.2001年よりLamivudineを開始し,その後LamivudineとAdefovirの併用,更にEntecavirとAdefovirの併用へと切り替え,ALTは正常範囲内を維持するに至り,HBV-DNA量も低下した.2009年8月,AFPが26.5 ng/ml,従来法AFP-L3分画が48.0%と上昇を認め,翌9月のMRI検査にて肝S3に径10 mmの典型的な肝細胞癌を認めた.2010年2月に肝外側区切除を施行し,組織診断は高分化型肝細胞癌であった.保存血清を用いて2010年より測定可能となった高感度AFP-L3分画をretrospectiveに再測定したところ,肝細胞癌が臨床診断される3年前,2006年から高感度AFP-L3分画が上昇し続け,切除後に正常化していたことが確認された.高感度AFP-L3分画は,AFPの上昇と肝細胞癌の臨床診断に先行して異常高値となる症例もあり,肝細胞癌のhigh riskグループを抽出する腫瘍マーカーとして有用と考えられた.

The present study was undertaken to examine the effect of cerebrosides derived from the sea cucumber Acaudina molpadioides and the starfish Asterias amurensis on the anti-tumor activity in vitro and in vivo. The results indicated that both Acaudina molpadioides cerebrosides (AMC) and Asterias amurensis cerebrosides (AAC) exhibited an inhibitory effect on cell proliferation through induction of apoptosis in S180 cells. Moreover, administration of AMC and AAC (50 mg/kg BW) on S180 tumor bearing mice reduced the tumor weight by 45.24 % and 35.71 %, respectively. In S180 ascites tumor model, AMC and AAC (50 mg/kg BW) treatment exhibited a significant ascites fluid growth inhibition of 31.23 % and 22.72 %. Furthermore, the ascites tumor cell viability ratio in AMC and AAC groups reduced to 50.89 % and 51.69 %, respectively. The life span of AMC and AAC administrated groups increased by 55.28 % and 35.77 % compared to control. Quantitative real-time PCR analysis demonstrated that the administration of AMC and AAC down-regulated the expression of Bcl-2, Bcl-xL, while on the other hand, up-regulated Bax, Cytochrome c, caspase-9 and caspase-3 mRNA level of the S180 ascites tumor cells. It was concluded that AMC and AAC should have potential anti-tumor activity both in vitro and in vivo by inducing apoptosis through the mitochondria-mediated apoptosis pathway. AAC seemed to be more effective than AMC in vitro but less potent in vivo. It may depend on the structural differences in their fatty acid groups and sphingoid bases.

Poisons and drugs other than alcohol and CO were detected in 17 of all judicial autopsy cases in the Department of Legal Medicine, Okayama University Medical School during the last 12 years. We report 7 cases from the forensic toxicological viewpoint. 1) lethal overdose of Sedes® A, 2) fatal poisoning due to biperiden, thioridazine and nitrazepam taken simultaneously, 3) fatal DDVP poisoning, 4) fatal paraquat poisoning, 5) hypoxic death by inhalation of liquefied petroleum gas, 6) fatal shock induced by sulpyrine, and 7) fatal shock induced by ketoprofen.

わが国の食料の生産基盤である耕地は減少を続け,食料安全保障に多大な影響を及ぼしうる.本研究では,わが国の食料の生産力の現状を世界の各地域と比較評価するとともに,潜在的な生産力を現在ならびに将来について推定した.解析は,FAOSTATの主要作物14品目のデータを用い熱量に換算した.わが国で自給している主要作物からの一人あたり一日当たりの供給熱量は1961年の2497 kcalから低下を続け,2010年には795 kcalとなった.この水準は世界平均の26%かつアフリカ地域の37%である.供給熱量を最大化するため,すべての耕地にイネとイモ類を植え付けた場合でも1626 kcalと,アフリカ地域の76%にとどまった.同様に,将来について,人口の減少傾向に加えて,耕地と単収の変化を考慮して4シナリオを比較した.シナリオ1では耕地が減少を続け,単収が増加しない場合,シナリオ2では耕地が減少せず,単収が増加しない場合,シナリオ3は耕地が減少せず,単収が増加する場合,シナリオ4では人口減少がない場合とした.なおシナリオ1~3は人口が減少するとした.2030年において,シナリオ1では1608 kcalと現状と同程度に対し,耕地面積が減少しないシナリオ2では1826 kcalに,それに加えて現在の単収の増加傾向が続くシナリオ3では1976 kcalまで増加が推定された.一方,シナリオ4として人口の減少がないとした場合では1464 kcalまでの減少を予測した.本研究により,わが国の食料安全保障を考えた場合,現在からの耕地面積の減少を回避するとともに,単収の持続的な増加は欠かすことができないことを示した.

This study evaluated the effects of several low-concentration bleaching products on microtensile bond strength (µTBS) to enamel at different elapsed times after the bleaching treatment. One hundred and fifty bovine incisors were divided into five groups: No treatment, 10% carbamide peroxide (Opalescence), 10% hydrogen peroxide (Opalescence Treswhite Supreme), 3% carbamide peroxide plus lactoperoxidase (WhiteKin), or 3% carbamide peroxide plus 2.7% carbamide peroxide (Clysiden Kit Express). All treatments lasted 4 weeks. After bleaching treatment or non-treatment, teeth were restored immediately, at 1, 3, 7, or 14 days after bleaching and submitted to µTBS test. Data were analyzed by ANOVA and Tukey’s test (p<0.05). For WhiteKin and Clysiden, µTBS to enamel was significantly reduced immediately, at 1, 3 and 7 days after bleaching. At 14 days after bleaching, similar µTBS values were obtained regardless of bleaching product. Therefore, while over-the-counter (OTC) products might affect bond strength to enamel, this effect was no longer observed after 14 days.
