The intracellular DNA of bacteria, bacteriophage and plasmids is subject to topological constraints that function to maintain it in a superhelical conformation. For the last several years, accumulating evidence has indicated that supercoiling of DNA duplex is required for effective transcription, as well as for the replication and recombination of cellular DNA. The relevance of DNA superhehlicity to transcription in vivo has been suggested by the observations that the overall rate of transcription directed by the E. coli, bacteriophage and plasmid genomes is generally reduced when the activity of DNA gyrase is inhibited by the specific drugs, such as nalidixic acid or coumermycin. An interesting observation from our in vitro studies has been that stimulation of transcription by superhelicity of DNA is more pronounced with S 100 crude extract proteins than with pure RNA polymerase.DNA in the prokayrotic cell has a compact conformation due to its interaction with cellular proteins (Histone like proteins). Recentry, seven classes of those proteins (HLP I, HLP II, 28K protein etc.) have been isolated and characterized as essential components of the nucleoid. In this review, we focus on the effect of superhelical conformation of the DNA duplex and association of the DNA with the nucleoid proteins on transcription.

わが国における交通事故死者数は減少傾向にあるものの「歩行中」など致死率の高い事故の減少率は低く,安全対策の必要性が高まっている.愛知県豊田市では「歩行者保護モデルカー活動」と称した人対車両事故削減を目的とした交通安全啓発に取組んでいる.本研究では「横断歩道での歩行者優先」を取り上げ,ドライバーへの意識調査で得られた知見を踏まえ,地域住民らによる交通安全街頭啓発運動を活用した効果的な啓発方法を検討した. ドライバーとのアイ・コンタクトを取りながら「止まってくれてありがとう!」等のメッセージを掲示し停止したドライバーに会釈する等,街頭において「能動的な啓発活動」の実証実験を行った結果,「能動的な啓発活動」実施時には通常の街頭活動時よりも歩行者優先率が向上することが明らかとなった.

自己免疫疾患は超高齢化社会の日本と世界の先進国で増加している。自己免疫疾患の発症や増悪は病巣感染で誘因されることが知られている。高齢者でも安心に使用できる治療法の開発のため、詳細な機序の解明が急務である。これまでの研究成果を発展させて超高齢化社会で問題となる病巣感染の病態メカニズムに迫り、高齢者でも安心な合併症の少ない治療法の開発への貢献を目指す。

SBHSの中で最も遅くJIS化されたSBHS400および未だ道路橋示方書に規定のないSBHS700の機械的性質や耐荷力に関する情報は,従来鋼やSBHS500に比べ不足している.本研究では,SBHS400とSBHS700の引張試験を実施し,機械的性質に関する情報を収集するとともに,従来鋼やSBHS500との比較をおこなった.続いて幅厚比パラメータの互いに異なる短柱軸圧縮試験を実施し,両縁支持板としての耐荷力特性を詳細に検討した.その後,鋼材の応力-ひずみ関係や残留応力分布の違いが軸力-軸方向変位関係に与える影響を解析的に検討した.最後に,実験結果と解析結果を基に,SBHS400およびSBHS700製両縁支持板の軸圧縮耐荷力評価式を提案した.

起立性調節障害は今日思春期の疾患としてまた社会的問題として,その治療が注目されている。今回小児心理学会の診断基準による起立性調節障害と思われる10症例に,漢方医学的診断と治療を実施して,凡てで改善をみることができた。この10例を分析したところ,起立性調節障害を引き起こす背景として,血証,水毒,心身症的要因などが見いだされ,血証には加味逍遙散,温経湯,当帰芍薬散を処方,水毒には五苓散合九味檳榔湯,苓桂朮甘湯を処方,心身症的要因には柴胡桂枝湯,柴朴湯を処方した。現代医学的治療では治療効果が不十分であり,漢方医学的診断と治療が大変有効であると考えられる。

遷延性術後痛(chronic postsurgical pain:CPSP)は,多くの患者のQOLを損失する深刻な手術合併症である.CPSPは侵襲の大きさなどの手術因子,精神・心理的因子や遺伝素因などの患者関連因子,環境因子を含んだ複合的なメカニズムによって発症し,その病態の理解は単純ではない.これまでにCPSP対策として薬物療法,神経ブロック,低侵襲手術の普及などさまざまな治療介入が行われており一定の効果は得られているが,より効果的な治療介入の開発のためにさらなる研究が望まれる.精神・心理的因子はCPSPの明らかな危険因子であり,これらの影響を評価し適切な治療介入を行うためには,痛みの有無や強度のみの単純な評価だけではなく,痛みの多面的評価が必要である.最近では,遺伝子解析やMRIなどの先進的な手法を用いてCPSPのリスクや病態を客観的に評価する試みがなされている.本稿ではCPSP対策の現状と今後の課題を中心に概説する.

B細胞亜集団である制御性B細胞はIL-10産生を介して強い抗炎症機能を持つが、IL-10産生機構のその全貌は不明であった。申請者は、IL-10レポーターマウスを用いて、IgM+形質細胞がIL-10の主な産生源であることを見出した。また、B細胞におけるIl10の発現は形質細胞分化の必須分子であるBlimp1と強く相関し、遺伝子導入実験からBlimp1がIl10の発現を誘導することを明らかにした。また、IL-10産生性形質細胞は、B細胞が抗原刺激を受けた際にIgG+形質細胞へ分化する過程および抗原特異的IgGの産生をサポートする機能を持つことが示唆された。

When designing systems that support remote instruction on physical tasks, one must consider requirements : 1) participants should be able to use non-verbal expressions, 2) they must be able to take an appropriate body arrangement to see and show gestures, 3) the instructor should be able to monitor operators and objects, 4) they must be able to organize the arrangement of bodies and tools and gestural expression sequentially and interactively. GestureMan was developed to satisfy these four requirements by using a mobile robot that embodies a remote instructor's actions. The mobile robot mounts a camera and a remote control laser pointer on it. Based on the experiments with the system we discuss the advantage and disadvantage of the current implementation.

膵臓への異所性脂肪の蓄積である脂肪膵は、膵β細胞機能障害によって糖尿病、炎症性アディポカインによって膵臓癌を惹起すると推察されている。しかし先行研究では一致した見解が得られていない。脂肪膵が糖尿病や膵臓癌の危険因子であるかを明らかにすることを目的とする。健診施設のCT検診受診者において、CT画像を用いて測定した膵脂肪量のデータと、10年間の一般健診データを突合し、脂肪膵と糖尿病発症の関連を縦断的に解析する。また健診施設の関連病院で、10年間に診療した膵臓癌患者の膵脂肪量をCT画像から測定する。CT検診受診者と膵臓癌患者の膵脂肪量を比較し、脂肪膵と膵臓癌の関連を検討する。

西田らは、主体価値測定アプリを開発し、思春期大規模コホート研究に導入した。その結果、思春期のロールモデルの獲得・更新と自己制御性の発達が、自律性の成熟の基盤となり、さらにその自律性が基盤となって主体価値が形成されていくことを解明した。山崎らは、全英出生コホートデータを用い、思春期の主体価値と自己制御の発達の相互作用が高齢期のウェルビーイングを予測することを解明した。川上らは、国内外のコホートデータを用いた一連の研究から、思春期主体価値の2要因モデルを提唱した。

In order to mitigate seismic damage of timber houses, application of passive control devises is demanded. The authors have developed high performance shear walls with various types of dampers, which are called “energy dissipation walls”. For the future, new design method has to be proposed to generalize passively controlled timber structures. In this paper, a method to evaluate allowable strength of shear wall is proposed. Advantage of passively controlled structure is to reduce maximum deformation, and energy dissipation walls are added to a structure so that maximum deformation angle of the structure subjected to level2 earthquake is less than target deformation. Accuracy of the method is demonstrated through a lot of non linear time history analyses.

Some of the pigs introduced into the Shibaura Abattoir, Tokyo, from Nagano Prefecture on July 17, 1969, presented many irregularly round nodules of miliary or egg size scattered on the surface of peritoneal organs, including diaphragm, liver, gallbladder, omentum, colon, prostate, and ureteral muscle. Histopathologically, these nodules were composed of tissue similar to the testicular one, but contained no spermatids. It was demonstrated that hormone had been secreted from them.The cause of formation of these nodules was studied and proved to be the transplantation of such free testicular tissue as appearing after the performance of non-sanguineous castration by the ball masher method of Miyazawa.

The existence of critical impact velocity has been confirmed in experiments in which a sheet was perforated by a flying projectile. The experiments were performed to measure the values of parameters relevant to the normal impact of the cylinder against the sheets. By examining the relationship between the diameter of blanks and impact velocity, the phenomenon of the critical impact velocity was confirmed. The theoretical approach was also presented, for the same conditions as in the case of simple wave propagation, as in the Karman-Duwez solution. The comparison between theoretical examination and experimental results was also discussed.
