Self-incompatibility is the system that inhibits pollen germination and pollen tube growth by self-pollen. This trait is important for the breeding of Brassica and Raphanus species. In these species, self-incompatibility is governed by the S locus, which contains three linked genes (a set called the S haplotype), i.e., S-locus receptor kinase, S-locus cysteine-rich protein/S-locus protein 11, and S-locus glycoprotein. A large number of S haplotypes have been identified in Brassica oleracea, B. rapa, and Raphanus sativus to date, and the nucleotide sequences of their many alleles have also been registered. In this state, it is important to avoid confusion between S haplotypes, i.e., an identical S haplotype with different names and a different S haplotype with an identical S haplotype number. To mitigate this issue, we herein constructed a list of S haplotypes that are easily accessible to the latest nucleotide sequences of S-haplotype genes, together with revisions to and an update of S haplotype information. Furthermore, the histories of the S-haplotype collection in the three species are reviewed, the importance of the collection of S haplotypes as a genetic resource is discussed, and the management of information on S haplotypes is proposed.

To evaluate the safety of food for pets, the migration of chemical substances from pet tableware was investigated by mass spectrometry. The presence of polymer additives Irgafos 168 and Erucamide were suspected based on mass spectra and were confirmed to be present in polypropylene tableware. The amount of substances migrated using simulated saliva was examined by liquid chromatography-mass spectrometry after solid phase extraction and purification. Photoionization was found to be suitable for the simultaneous determination of these substances. The detection limits of the established method were 0.019 μg/mL for Irgafos 168 and 0.022 μg/mL for Erucamide. Five different types of pet tableware purchased in local markets were examined and no analytes were detected in the simulated saliva using shaking extraction. In this study, the risk to pets from the substances migrated from pet tableware was considered to be sufficiently low.

本論文はドイツ・バイエルン州における農家民宿に注目し,農村空間の商品化にともなうルーラルツーリズムの発展をプルーリアクティビティ(多就業形態)と関連づけながら検討した。バイエルン州では,農村の景観や農牧業,および生活文化が農家民宿の発展と相互に関連しながら維持され,それらのアトラクションの商品化を促進させることで農村観光の発展が図られてきた。農村空間の商品化は農村の景観や農牧業,および生活文化を維持することに貢献し,そのことが農家民宿や農村観光のさらなる発展につながった。バイエルン州ではプルーリアクティビティが機能し,酪農や林業,および農家民宿が行われることで,すべての土地基盤にわたって農村資源が利用されるようになり,そのことによって農村景観が維持される。農村景観は農家民宿の利用者や農村観光の訪問者のアトラクションとなり,農村空間の商品化が決定づけられた。

The fatty acid composition of lichens and their symbionts was determined. In lichens, linoleic and oleic acids were the most abundant and these acids were also rich in their phyco- and mycobionts. A small amount of arachidonic acid was detected in some of the lichens and their phyco- and mycobionts.

Background: We established a community-based cohort study to assess the long-term impact of the Great East Japan Earthquake on disaster victims and gene-environment interactions on the incidence of major diseases, such as cancer and cardiovascular diseases.Methods: We asked participants to join our cohort in the health check-up settings and assessment center based settings. Inclusion criteria were aged 20 years or over and living in Miyagi or Iwate Prefecture. We obtained information on lifestyle, effect of disaster, blood, and urine information (Type 1 survey), and some detailed measurements (Type 2 survey), such as carotid echography and calcaneal ultrasound bone mineral density. All participants agreed to measure genome information and to distribute their information widely.Results: As a result, 87,865 gave their informed consent to join our study. Participation rate at health check-up site was about 70%. The participants in the Type 1 survey were more likely to have psychological distress than those in the Type 2 survey, and women were more likely to have psychological distress than men. Additionally, coastal residents were more likely to have higher degrees of psychological distress than inland residents, regardless of sex.Conclusion: This cohort comprised a large sample size and it contains information on the natural disaster, genome information, and metabolome information. This cohort also had several detailed measurements. Using this cohort enabled us to clarify the long-term effect of the disaster and also to establish personalized prevention based on genome, metabolome, and other omics information.

Mitochondria play an important role in energy conversion as well as in intracellular calcium (Ca2+) storage. Ca2+ uptake from the cytosol to the mitochondria is mediated by the calcium uniporter, which functions as a Ca2+ ion channel. However, the molecular composition of this uniporter has remained unclear until recently. The Ca2+ ion channel consists of seven subunits. The yeast reconstitution technique revealed that the mitochondrial calcium uniporter (MCU) and essential MCU regulatory element (EMRE) are the core subunits of the complex. Furthermore, detailed structure-function analyses of the core subunits (MCU and EMRE) were performed. In this review, the regulatory mechanism of mitochondrial Ca2+ uptake is discussed.

Electrode reactions in electrochemical devices often consist of charge transfer at electrode/electrolyte interface and charge compensation in electrode active material. Therefore, to design electrochemical devices with high electrochemical performance, it is important to understand electronic structures of the electrode/electrolyte interface and electrode bulk during electrochemical reactions, and to design electrode materials to control them. In this paper, certain phenomena at the electrode/electrolyte interface and electrode bulk in lithium-ion batteries are clarified by using synchrotron radiation X-ray analyses. The information obtained from those X-ray analyses are applied to control the structure of the electrode/electrolyte interface and electrode bulk. Moreover, new cathode materials for all-solid-state fluoride-ion with high power and cyclic performances have been developed, compared to simple metal/metal fluoride materials, by using the findings obtained from the synchrotron radiation X-ray analyses.

L-amino acid oxidase (LAAO) is a metabolic enzyme that converts L-amino acids into ketoacids, ammonia, and hydrogen peroxide (H2O2). The generated H2O2 has previously been shown to have antibacterial and gut microbiota-modulatory properties in LAO1 knock-out (KO) mice. Since most microbial metabolites reach the liver through the portal vein, we examined gut-liver interactions in LAO1 KO mice. We found lower total cholesterol levels, higher glutamic pyruvic transaminase (GPT) levels in the serum, and higher pro-inflammatory cytokine mRNA expression in the liver tissue. In wild-type (WT) mice, LAO1 was expressed in gut tissues (ileum and colon). Microbiome analysis revealed that the abundance of some bacteria was altered in LAO1 KO mice. However, short-chain fatty acid (SCFAs) levels in cecal feces and gut permeability did not change. Fecal microbiota transplantation (FMT) revealed that feces from LAO1 KO mice slightly stimulated pro-inflammatory cytokine expression in the liver. During metabolomic analysis, 5-Aminolevulinic acid (5-ALA) was the only metabolite found to be significantly upregulated in the portal and abdominal veins of the LAO1 KO mice. Intraperitoneal administration of 5-ALA to WT mice significantly increased IL-6 mRNA expression in the liver. These observations suggest that gut LAO1 plays a role in regulating 5-ALA production and that a high level of 5-ALA stimulates the liver to increase pro-inflammatory cytokine expression by disrupting LAO1 in mice.

Reinforcement learning is promising as a machine learning paradigm in edge computing. However, its high computational cost poses a challenge when implementing in devices with limited circuit resources and power consumption. In this study, we investigated the relationship between the bit-length of floating-point operations and the learning performance of the reinforcement learning algorithm. In the case of the FrozenLake maze problem, we found that the learning performance of 8-bit floating-point arithmetic decreased, while that of 16-bit floating-point arithmetic was comparable to that of 64-bit CPU arithmetic. Our results provide a practical guideline for designing a dedicated reinforcement learning hardware with minimum circuit resources and power consumption.

Computational screening is one of the fundamental techniques in drug discovery. Each compound in a chemical database is bound to the target protein in virtual, and candidate compounds are selected from the binding scores. In this work, we carried out combinational computation of docking simulation to generate binding poses and molecular mechanics calculation to estimate binding scores. The coronavirus infectious disease has spread worldwide, and effective chemotherapy is strongly required. The viral 3-chymotrypsin-like (3CL) protease is a good target of low molecular-weight inhibitors. Hence, computational screening was performed to search for inhibitory compounds acting on the 3CL protease. As a preliminary assessment of the performance of this approach, we used 51 compounds for which inhibitory activity had already been confirmed. Docking simulations and molecular mechanics calculations were performed to evaluate binding scores. The preliminary evaluation suggested that our approach successfully selected the inhibitory compounds identified by the experiments. The same approach was applied to 8820 compounds in a database consisting of approved and investigational chemicals. Hence, docking simulations, molecular mechanics calculations, and re-evaluation of binding scores including solvation effects were performed, and the top 200 poses were selected as candidates for experimental assays. Consequently, 25 compounds were chosen for in vitro measurement of the enzymatic inhibitory activity. From the enzymatic assay, 5 compounds were identified to have inhibitory activities against the 3CL protease. The present work demonstrated the feasibility of a combination of docking simulation and molecular mechanics calculation for practical use in computational virtual screening.

The improvement of type 2 diabetes mellitus induced by naturally occurring polyphenols, known as flavonoids, has received considerable attention. However, there is a dearth of information regarding the effect of the trihydroxyflavone apigenin on pancreatic β-cell function. In the present study, the anti-diabetic effect of apigenin on pancreatic β-cell insulin secretion, apoptosis, and the mechanism underlying its anti-diabetic effects, were investigated in the INS-ID β-cell line. The results showed that apigenin concentration-dependently facilitated 11.1-mM glucose-induced insulin secretion, which peaked at 30 µM. Apigenin also concentration-dependently inhibited the expression of endoplasmic reticulum (ER) stress signaling proteins, CCAAT/enhancer binding protein (C/EBP) homologous protein (CHOP) and cleaved caspase-3, which was elevated by thapsigargin in INS-1D cells, with peak suppression at 30 µM. This was strongly correlated with the results of flow cytometric analysis of annexin V/propidium iodide (PI) staining and DNA fragmentation analysis. Moreover, the increased expression of thioredoxin-interacting protein (TXNIP) induced by thapsigargin was remarkably reduced by apigenin in a concentration-dependent manner. These results suggest that apigenin is an attractive candidate with remarkable and potent anti-diabetic effects on β-cells, which are mediated by facilitating glucose-stimulated insulin secretion and preventing ER stress-mediated β-cell apoptosis, the latter of which may be possibly mediated by reduced expression of CHOP and TXNIP, thereby promoting β-cell survival and function.

Massive MIMO is expected to realize a large capacity for wireless communication technology. To reduce the number of devices of the massive MIMO, utilizing a two-dimensional Butler matrix for the feed circuit has been investigated, but complexing its design. We have proposed a Massive MIMO configuration using a stacked Butler matrix and a staircase array antenna to reduce the circuit design cost. This letter proposes a V-shape staircase array antenna with a stacked Butler matrix to reduce antenna profile. Computer simulation clarifies the beam steering range for the proposed antenna.

本研究の目的は、健康な成人女性6名(平均年齢50.2歳±8.7歳)を対象にストレス値からみたタクティールタッチ(スウェーデン発祥の触れるケアの一種である)の有効性を明らかにすることである。対象者の施術前条件の統一を図る目的で数計算100問の解答を課した。タクティールタッチ施術グループ3名とタクティールタッチ施術を受けない安静臥位グループ3名に分け、各対象者の左胸部にヘルスパッチを貼付し、数計算時およびタクティールタッチ施術時(または安静臥位時)その後の安静10分後、20分後、30分後、飲水時のストレス値、心拍数、体温を測定しFFT(高速フーリエ変換)で算出しHRV解析した。HRV解析図およびストレス値と心拍数・体温のHRV解析結果の平均値比較から、タクティールタッチ施術を受けたグループの方がストレス値は大きく低下し心拍数も低下したのみならず持続も認められた。

A new transcriptional up-regulator designated TMC-205 was discovered from the fermentation broth of an unidentified fungal strain TC 1630 by using an SV40 promoterluciferase reporter assay. Based on spectroscopic analyses, its structure was determined to be (E)-6-(3-methyl-1, 3-butadienyl)-1H-indole-3-carboxylic acid. Expression of the luciferase activity was activated ca. 2-, 4-, and 6-fold by 1, 10, and 100μM TMC-205, respectively. TMC-205 activated the transcriptional activity in a manner dependent on the presence of the enhancer element of SV40 in its promoter region.

In this study we investigated the eye morphology and retinal topography of two owl species in relation to their visual environment. Although Short-eared Owl Asio flammeus is larger and weighs more than Japanese Scops Owl Otus semitorques, its eye dimensions (weight, corneal diameter, and axial length) are all smaller than the scops owl's. Owl retinas were examined in Nissl-stained whole-mount preparations. The total number of retinal ganglion cells (RGCs) was greater in the Japanese Scops Owl (4,703.0×103 cells) than in the Short-eared Owl (2,346.8×103 cells). The eye morphology of, and the number of RGCs in the Japanese Scops Owl indicate that it is more adapted to a nocturnal habit. An area of high-density RGCs was horizontally distributed in the temporal retina of the Short-eared Owl, with a peak density of 17.4×103 cells/mm2. In the Japanese Scops Owl's temporal retina, there was an oval-shaped arrangement with a peak density of 23.1×103 cells/mm2. These distributions indicate that whereas Short-eared Owl is adapted to open habitats, Japanese Scops Owl is adapted to enclosed habitats. The RGCs of both species were classified into three categories (small, medium, and large) based on the size and appearance of somas. In both species, medium-sized cells predominated, and the proportion of large-sized cells was smallest. The distribution and high-density areas differed among these groups, suggesting adaptation to the visual environment. The high-density areas of these groups cover the nasal visual fields, which include the overlap for binocular vision.
