C-1027 is a potent antitumor antibiotic, in which a nonprotein chromophore is tightly and specifically bound to an apoprotein. The chromophore, which has an endiyne structure and is responsible for DNA cleavage, is very labile when isolated, but greatly stabilized through binding to the apoprotein. Their binding structure and stabilizing interactions are very interesting problems in terms of molecular recognition and protein transport. The 3D structure of C-1027 apoprotein was determined by the X-PLOR calculation using 1539 experimental restrains derived from NMR spectroscopy. The apoprotein has three antiparallel β-sheets, and the hydrophobic pocket is formed by four-stranded β-sheet (DCHI) and two loops (residues 75-79, 97-100)(Fig.3). The overall shape of the apoprotein is quite similar to those of neocarzinostatin (NCS) and actinoxanthin (AXN). The binding structure of C-1027 complex was calculated based on 1539 NMR-derived constraints which include 38 intermolecular restraints between the aromatized chromophore and the apoprotein. The aromatized chromophore is bound to the hydrophobic pocket of the apoprotein. The benzodihydropentalene core locates in the center of the pocket with its molecular plane almost perpendicular to the bottom of the pocket. The β-tyrosine unit locates on the left side of the core, and both benzoxazine and aminosugar moieties on the right side (Fig.4). The hydrophobic interaction is most likely the major binding interaction between the apoprotein and the aromatized chromophore. Moreover, the 18-amino group of the chromophore locates in the proximity of either the carboxylate of Asp101 or the imidazole ring of His104, which indicates there could be a salt-bridge or a hydrogen bond type interaction between the aromatized chromophore and these side chains. To confirm the predicted binding interactions, we made several mutant apoproteins whose specific amino acid residue is replaced with the amino acid of different type and examined their binding abilities for the aromatized chromophore by NMR. The results obtained will be discussed.

The antitumor antibiotic C-1027 is a 1:1 complex of a highly labile enediyne chromophore (1) and a carrier apoprotein. C-1027 exhibited the potent cytotoxicity toward various cancer cells. The p-benzyne biradical (2), which is in equilibrium with 1, abstracts hydrogens from DNA to exert its biological activity. The apoprotein functions as both the stabilizer and the drug delivery system of 1. Recently, we found that the biradical 2 slowly abstracts α-proton of Gly96 of the apoprotein, which caused the oxidative cleavage of the peptides and led to a self-degradation of C-1027. To create a more stable analog of antibiotic C-1027, we designed a Gly96-deuterated (D-Gly) apoprotein. The D-Gly apoprotein was expressed in Escherichia coli in the presence of glycine-d_5. The unstable chromophore 1, isolated from natural C-1027, was then incorporated into the D-Gly apoprotein using HPLC techniques to obtain the D-Gly C-1027. Stability tests revealed that the D-Gly C-1027 was 1.8 and 4.9 times as stable as the natural one under solid and solution states, respectively. Cytotoxicity test also reflected the stability of D-Gly C-1027. Thus, we achieved the creation of supranatural products by rational design utilizing kinetic isotope effect. The presented work demonstrated the novel design principle to create the supra-natural products by integrating the data of physicochemical property of the small molecule and the atomic-level 3D-structure of the protein, which will be applicable to other biologically important natural products and proteins.

Antibiotic C-1027, a novel antitumor chromoprotein isolated from the broth filtrate of Streptomyces globisporus C-1027, shows extremely potent cytotoxicity against KB carcinoma cells (IC_<50> 0.1ng/ml) in vitro and antitumor activity toward tumor-bearing mice in vivo. These activities are correlated with the ability of the antibiotic to cause DNA double-strand scission. The antibiotic consists of an apoprotein and a labile chromophore (C-1027-Chr) that is responsible for the biological activity of C-1027. The chromophore is readily separated from its apo-protein by extraction, but the exceeding instability in the protein-free state hampered the structure elucidation. The similar situation has been also observed in the other chromoprotein antibiotics of this family such as neocarzinostatin (NCS), macromomycin, auromomycin (AUR), actinoxanthin and kedarcidin. Among them, NCS is the only one whose chromophore structure has been elucidated, and very recently, the structural novelty of kedarcidin chromophore has been disclosed by the Bristol-Myers Squibb group. We have characterized an inactive but more stable reaction product (2) of C-1027-Chr, which was prepared by treatment of C-1027-Chr in ethanol. It possesses a macrocyclic structure together with oxazolinate and aminosugar moieties as side chains. Its benzodihydropentalene core structure suggested to us the presence of an enediyne in the native C-1027-Chr. We disclose herein the novel structure of C-1027-Chr (1) and the cycloaromatization mechanism leading to product (2), which would explain its extreme potency in terms of cytotoxicity and ability to cause DNA double-strand scission.

近年,運転支援システムの普及が進み,交差点での一時停止支援や出会い頭事故防止支援などが要望されている.これらのシステムの実現には高精度な位置推定とともに高精度な地図情報も必要である.特に運転支援に利用可能な高精度地図の普及は進んでいない.通常,運転支援用の高精度地図の作成には,専用の計測車両で情報を収集し人手で車線や交差点位置を抽出する必要があるため,地図の生成には膨大な費用がかかる.安価な方法として,カーナビの走行履歴から大まかな交通状態や道路形状を収集するプローブシステムもあるが,運転支援用には精度が不足する.そこで,我々は運転支援用の車載カメラ画像と汎用GPSの生データを用いた高精度地図生成手法を提案する.特にGPSのドップラーから得られる高精度軌跡に基づいて相対的に精度の高いローカルな運転支援地図を生成し,複数のローカルな運転支援地図からグローバルな高精度地図を生成する手法を提案する.本報は,地図生成の基本概念とその可能性検討結果について報告する.

The purpose of this study was to construct a portal site for distributing on-line documents for students. The site aimes at not just distributing documents but also receiving replies, such as questions and comments, from students. For that purpose, a weblog system is used. The system is called 'Nucleus CMS.' In this paper, an installation and some customizations of the system are described. Further customizations are planned to use the portal site in real classes.

透明アクリル製の円柱を削り出して,"重力"レンズを製作した.アクリル樹脂だと入手もしやすく加工も容易で,職人芸的な特殊技能がなくても,十分実用に耐えるものを製作できる.設計から製作そして活用までを簡単に紹介したい.The gravitational lens is one of the major topics in the current astrophysics. Using an acryl bar,we make a "gravitational lens",wh ich is optically equivalent with a gravitational lens in the universe. We here show the design of and how to make the gravitational lens. Such a hand-made gravitationallens is quite useful###in your classrooms,scientific museums,and public observatories.

現在,日本の多くの学校で,総合的な学習の時間を利用して,地域社会に参加する活動が行われている。日本においても,子どもは「権利を行使する主体」であるとする「子どもの権利条約」の理念が浸透しつつあることは明らかであろう。しかし,学校における子どもの参加の中には,教師主導の活動であったり,イベント的で表面的な活動に終始してしまったりする実践も少なくない。地域社会を住み良い環境にするためには,子どもが責任を負い,大人と意思決定を共有する参加が必要である。本小論では,ロジャー・ハートやデヴィッド・ドリスケルの理論を手がかりとして,総合的な学習の時間の事例を分析することも含め,子どもの参加の質を高める方策について論じる。

一九四五年八月一五日、天皇の玉音放送によってアジア太平洋戦争は敗戦を迎えたが、終戦の詔勅の意味を理解した聴き手は稀であり、雑音の中、天皇の声もうまく聞きとれなかった。このような空虚さのゆえにその声はアウラを身にまとい、国体護持を図る大きな歴史の中に敗戦の個別の経験を回収すべく機能した。本稿は高村光太郎の戦争詩における戦時と敗戦に対する姿勢を検討することで、このような〈経験の歴史化〉の機構を相対化しようと試みたものである。

本州北部の2地点で吸血性ブユモドキの3新種が発見された。盛岡郊外で放牧牛に飛来したモリオカブユモドキF. (L.) moriokensis sp. n.は, 近似の習性で知られた中国産pungobovisと比べ著しく大きな触角比をもつ。那須の放牧草地内で採集されたマツムラブユモドキF. (L.) matsumurai sp. n.は触肢第3節の輪郭が亜三角形で壼形の感覚孔を有し, 他のナスブユモドキF. (L.) nasuensis sp. n.は既知種longicornisと雌ではごく近似し, 雄では交尾器の形態が相違している。

Driven by diminishing fossil fuel resources, global warming and subsequently rigid legislation on CO₂ emission, fuel economy is a major challenge for the automotive industry. Each element of the powertrain has been optimized or newly designed to increase efficiency. In this optimization process the engine oils and transmission fluids are important design elements and their contribution to improved efficiency is significant. Polyalkylmethacrylates (PAMAs) are widely used as viscosity index improvers in engine, transmission and hydraulic oils. They have been shown to adsorb from oil solution onto metal surfaces to produce thick and viscous boundary films. These films are maintained even in low speed and high temperature conditions and thus produce a reduction of friction and wear. It was found that specifically designed film-forming PAMAs can improve pitting performance of lubricant formulations. The paper describes the impact of tailor-made functionalized PAMAs on boundary film formation and explores their ability to increase the fatigue life of lubricants.

He acquired Carlo Maratta's Marchese Pallavicini conducted by Apollo to the Temple of Fame in 1758 and commissioned in 1759 a pendant to this, Augustus and Cleopatra, from Anton Raphael Mengs, arguably the most celebrated European painter of the day. This subject is uncommon in the iconography of the seventeenth and eighteenth centuries compared with other Cleopatra's subjects, such as the Death of Cleopatra and Cleopatra's Banquet. As is evident in Cortona's Augustus and Cleopatra it represents Augustus's morally overcoming Cleopatra's temptation. Given his preference for the Choice of Hercules, Henry must have liked its ethical aspect. As Henry had Guercino's Augustus and Cleopatra in mind, he was not satisfied with Mengs's picture: his Cleopatra lacked 'grandeur' or 'majesty'. Yet his criticism implies more than that. As the concept of luxury had been changing in the eighteenth century, Henry needed Cleopatra's gorgeous dress as a symbol of his own luxuries, the garden and his art collection. He did not need an element of sacredness in Cleopatra which he found in Mengs's or Reni's Cleopatra. As Henry was the founder of Stourhead garden and the patron of artists, Henry could identify himself with Augustus, the founder of Rome and the patron of Virgil, whose Aeneid was used as a theme for Henry's garden.Henry's choice of Augustus and Cleopatra may have been influenced by the following books and plays. Sara Fielding's The Lives of Cleopatra and Octavia, whose subscribers include Henry's friends, was published in 1 757. Dryden's All for Love was performed in April, 757 and March, 758. David Garrick's version of Shakespeare's Antony and Cleopatra was published in October, 758 and the play was performed in January, 759. It is reasonable to assume that Cleopatra was one of the favorite topics of conversation among the fashionable circles in London in those years.

The focus of this paper is to analysis the researches on teacher's organizational socialization and survey the research trend in the future. Many papers about teacher socialization are published. Most of those studies focus on occupational socialization. First, we review those studies. Second, we aim to clarify deference between that and organizational socialization. Difference of content which newcomer internalize is not clear. However re-socialization, realistic job preview(RJP), socialization strategies ,and effect of socialization are characteristic of the latter. Those concepts are applicable to teacher socialization studies. And they will lead new research framework. We propose applying RJP to research about teaching practice, socialization strategies to research about training and mentoring teachers, effect of socialization to research about job separation ,and re-socialization to research about personnel change. At the end, we point out some problems about teacher socialization.

抗ウマCRPウサギ血清を用いた単純放射免疫拡散(SRID)法により, 臨床的に正常なウマの加齢および周産期におけるCRPの血中動態および実験的に炎症を作出したウマにおけるCRPの血中動態を検討した. 初乳未摂取の新生子の血清CRP濃度は雌雄ともに測定限界以下であったが, 幼齢期に上昇し, 青壮年期に低値を示し, 以降やや増加して安定するという加齢性の変化を示した. 周産期には, 分娩時および分娩後2か月目から4か月目の血清CRP濃度上昇および分娩2か月前の低下を認めた. 炎症刺激後24時間目より血清CRP濃度は上昇し, 3〜5日目で処置前の3〜6倍に達し最高値を示した. その後低下し14日目には処置前値に回復した. 以上の成績から, ウマにおいでもCRPは急性相蛋白であることが判明した.
