Immobilized metal affinity chromatography (IMAC) was used to separate ovotransferrin (OVT) by a single chromatographic step. Ovotransferrin was bound strongly to immobilized copper ions, while other components of egg white passed through the column. Using acidic pH and/or a strong competing solute, imidazole, ovotransferrin was recovered in 94〜98% pure form as indicated by SDS-polyacrylamide gel electrophoresis and immunoelectrophoresis. The elution profiles from the IMAC column indicated the presence of two forms of ovotransferrin. The capacity of the IMAC column was 20mg OVT/ml copper loaded gel. Saturation of the metal binding sites of ovotransferrin with Fe^<2+> and Cu^<2+> did not interfere with the binding of ovotransferrin to the IMAC column. However, modification of the histidine residues in ovotransferrin with diethyl pyrocarbonate almost completely destroyed the binding to the IMAC column.

無線通信における情報量的安全性に基づく鍵共有方法として,エスパアンテナを用いた秘密鍵共有方式が提案されている.本稿では,秘匿条件付相互情報量Imacを高めるための方法として,指向性セット候補用意法及び雑音環境により使用する指向性セットを切り替える適応選択法を提案する.鍵長を16bitとする計算機シミュレーションの結果,無作為に指向性セットを選んだ場合の平均と比較して以下を確認した.(1)指向性セット候補用意法によりImac=0.5となる送信電力を5.7dB低く抑えられる.(2)指向性セット適応選択法により同じく7.1dB低く抑えられる.

Background The identification of protein kinase A(PKA) anchoring proteins on mitochondria implies a direct effect of PKA on mitochondrial function. However, little is known about the relationship between PKA and mitochondrial metabolism. Methods and Results The effects of PKA on the mitochondrial redox state(flavin adenine dinucleotide(FAD)), mitochondrial membrane potential(ΔΨ_m) and reactive oxygen species(ROS) production were investigated in saponin-permeabilized rat cardiomyocytes. The PKA catalytic subunit(PKA_<cat>; 50 unit/ml) increased FAD intensities by 56.6±7.9%(p<0.01), 2'7'-dichlorofluorescin diacetate(DCF) intensities by 10.5±3.3 fold(p<0.01) and depolarized ΔΨ_m to 48.1±9.5% of the control(p<0.01). Trolox(a ROS scavenger; 100μmol/L) inhibited PKA_<cat>-induced ΔΨ_m, FAD and DCF alteration. PKA_<cat>-induced ΔΨ_m depolarization was inhibited by an inhibitor of the inner membrane anion channel(IMAC), 4,4'-diisothiocyanatostilbene-2,2'-disulfonic acid(DIDS: 1μmol/L) but not by an inhibitor of mitochondrial permeability transition pore(mPTP), cyclosporine A(lOOnmol/L). Conclusions PKA_<cat> alters FAD and ΔΨ_m via mitochodrial ROS generation, and PKAcat-induced ΔΨ_m depolarization was not caused by mPTP but rather by DIDS-sensitive mechanisms, which could be caused by opening of the IMAC. The effects of PKA on mitochondrial function could be related to myocardial function under the condition of extensive β-adrenergic stimulation.

The essencial pleasure of designing can be found in this procedure. Art and Design course of Tsukuba University reopened wood and metal craft class as fundamental study in 1998. This class aims to introduce an easy way to the students who hesitate to handle actual articles. Making a desk for iMac is an example. To touch and feel the real parts and material will be a big help to improve design mind of students.

現在、最も優れたデザインブランドの一つとも言われているアップル。そのアップルはガレージファクトリーという、非クリエイティブ環境で生まれたブランドである。しかし今日ではクリエイティブ業界でアップルを知らない者はいないくらい浸透している。そのアップルにおけるブランド要素はどのようなものであるか、MacintoshというブランドとiPodというブランドを通して考えてみたい。また実際に20代の学生のアップルブランドにおけるブランド意識はどのようなものがあるか考察してみた。

頚髄損傷による四肢麻痺者は運動機能のみならず、上肢の感覚機能にも障害を有する。そのため、高い巧緻性を必要とするマウス操作が困難となる場合が多い。そこで、感覚の無い手指やスティックによりポインティング操作を行なうためマウスの代替装置を利用する。しかし、キー操作などによる代替装置では失われた感覚機能を視覚により代替することが必要であり、マウスポインタに集中するべき視線が頻回に移動せざるを得ない。そこで、キーボードなどによる選択や押し替えの必要な方法ではなく、上肢の位置に影響されず、粗大な運動によって二次元の方向を示すことが可能なポインティングデバイスを考案・試作し、評価実験を行なった。Quadriplegics cannot operate the mouse because of paralyzed hand's motor and sensory function. When they use the mouse emulators by keyboard operation or switch operation, they have to move line of sight from the display to the keyboard quite frequently. We developed a pointing device that is not infruenced by position of the upper limb and can operate with limited motor function. This pointing device, "Magic Wand" can reduce excessive line of sight movement when quadriplegics are accessing the computers.

本研究は,インドネシアの現地系自動車部品メーカーの獲得した技術水準に着目し,アジアの自動車産業の産業システム高度化進展度を分析することを目的としている。分析にあたっては自動車メーカーとサプライヤー間の取引方式に着目した。先行研究によると「承認図」方式を通じてなされる取引方式では,双方化による学習(Learning-by-Reciprocating)効果が期待され,開発途上国への技術移転速度を速めると提唱されている。しかし,一方で,「承認図」方式による取引には,1)初期開発能力(企画・設計), 2)後期開発能力(工程開発能力,VE), 3)量産能力(品質,納期など),4)改善能力(VAなど)のすべての能力が必要とされている。そのため,本研究では現地系自動車部品メーカーヘの「承認図」方式の普及状況が産業システム高度化進展度の指標になると考え,承認図に関する事例分析を行った。分析の結果,現地系の部品メーカーは自動車メーカーに対して製造技術を提供できる段階に達しているが,その開発能力は発展途上にあり,産業システムとしてはいまだ未成熟であるとの結論を得た。しかし,「承認図メーカー」に近い現地系自動車部品メーカーも存在しており,産業システムが進化しつつあることも確認された。

We created many Computer Generated Images for 2-hours drama, RAMPO -mysterious women- in last year ('94). In this paper, allow me to introduce making of Computer Generated Images and special effects for this drama. We have tried to creat effect by Motion Control Camera and Matte Painting by Hollywood's sfx in addition to CGI. Digital images were rare for Japanese TV dramas, but recently, they show an increase trend.

When we estimate a sound field in a room, it is important to grasp the spatial informa-tion, especially of the early reflection periods. In this paper we'll discuss a way to grasp the spatial information of sound fields from impulse responses measured at closely located four points, the origin and three points of the same distance (3∼5cm) from the origin on the rectangular coordinate axes. From these four impulse responses the coordinates and powers of virtual image sources are calculated by correlation technique or intensity technique. Concert halls, opera theaters and many other sound fields are measured by this technique. The distributions of virtual image sources and directivity patterns of some concert halls are shown.

消化管疾患,特に悪性疾患の早期診断に有用な検査として,胃癌については発癌リスク評価としての血清ペプシノゲン法,ヘリコバクター・ピロリ抗体検査がある.いずれも胃癌の青景因子である萎縮性胃炎を評価し,発癌高危険群の囲い込みに有用である.また大腸癌については便潜血反応の有効性が確立している.更に便中遺伝子変異を検出することで癌の局在診断に結びつけようとする研究も進んでおり,今後の進展が期待される.

Using malic adapted bacterial cells, the various conditions that induce malo-lactic fermentatzion (MLF) in grape juice, apple juice and wine sterilized with diethylpyrocarbonate (DEPC), were studied.1. DEPC showed inhibitory action against MLF with malic adapted cells of Leuc. mesenteroides at a concentration higher than 200 ppm.2. The rate of MLF in the grape juice, the grape juice containing 250 ppm of DEPC and wine was more rapid with Leuc. sp. than with other lactic acid bacteria strains.3. SO_2 extremely inhibited MLF as shown in a previous paper, but the inhibition was not influenced with DEPC.4. Yeasts in grape juice or wine under SO_2-free conditions were killed completely at above 70ppm of DEPC, but DEPC had no effect on the prevention of browning of grape and apple juice or wine.5. MLF and microbicidal action of DEPC were not influenced by sodium erithorbate (antioxidant).6. From the above results, the appropriate methods of wine and cider making using malic adapted cells are suggested as follows.a) To grape juice or apple juce containing sodium erithorbate (SO_2-free), DEPC (about 200 ppm) should be added to kill microorganism in juice. After an hour, the washed malic-adapted cells (about 10^7/ml) should be added to the juice. This should be kept at 10℃ and allowed to undergo MLF without occuring alcohol fermentation with wild type yeast. When malic acid is decomposed completely, SO_2 (100 ppm) should be added to the grape or apple must and the starter (pure culture of wine yeast, OC No. 2) inoculated. And then this must should be allowed to undergo alcoholic fermentation with the purely cultured yeast at 15℃.b) To wine or cider containing a small amount of SO_2 (20 ppm or less) should be added sodiume erithorbate (about 200 ppm) and DEPC (about 150 ppm). After an hour, the washed malic adapted cells should be added to the DEPC treated wine or cider. Incubations should be carried out at 15℃ for 4〜7 days, and then add SO_2 (about 100 ppm) after complete decomposition of malic acid in order to store safely the resultant wine or cider for long periods.

In general, plants synthesize sucrose for translocation of photosynthate. However, some horticultural crops of Rosaceae family are known to translocate sorbitol. Both types of crops are important in horticultural science. Although sucrose metabolism in source and sink tissue is well-known, regulation of sugar composition has not been investigated yet. Fruit generally contains three major sugars i.e., sucrose, glucose and fructose. As the sweetness of these three sugars is quite different, the manipulation of sugar composition in fruit could be useful for improving fruit quality. Thus genes encoding fructokinase that might be related to the level of fructose in fruit were cloned and analyzed. In contrast to sucrose, there are fewer studies on sorbitol metabolism in source and sink tissue. Therefore cloning and expression analysis of the gene encoding sorbitol-6-phosphate dehydrogenase (S6PDH) that may play a key role in sorbitol biosynthesis are also reported here. Two fructokinase cDNAs, Frk1 and Frk2, were isolated from tomato and identified using transformed yeast with those genes. The response of both genes to sugar and their expression patterns shown by northern and in situ hybridization suggest that Frk1 responds to carbon availability and plays a primary role in fructose metabolism in plant cells while Frk2 is localized specifically in cells that import and store carbohydrate. On the other hand, S6PDH cDNA was cloned from apple seedlings. The levels of S6PDH activity and protein were high in source tissue such as cotyledons and leaves in apple seedlings. Besides the S6PDH transcripts were induced during germination corresponding to sorbitol synthesis. On peach trees, the levels of S6PDH transcripts were also higher in more developed leaves compared with folded young leaves. The results strongly suggest that the S6PDH gene is expressed for biosynthesis of sorbitol in source tissue. Now some groups are executing mapping and overexpression of S6PDH.

The importance of the nonverbal aspect in all communication has been well recognized. It is also known that the code (i.e. the relationship between signs and meanings) is different from culture to culture. In this book, we will discuss and analyze the meaning of behaviour, gestures, or nonverbal actions. Verbal actions (greeting, addressing, using polite expressions, etc.) will also be taken up. Compare the following typical actions of a maid in Japanese scenes. In example A from Natsume Soseki's The Wayfarer (Kojin), the narrator is a young man. The maid at his boarding house comes to his room to announce with a smirk that there is a lady visitor. "You are standing to speak to me!" I said sharply. The maid immediately kneeled down just outside the door sill. Example B from Soseki's Botchan is in a similar situation, but the woman is the old keeper of the boarding house: She was waiting for his reply after kneeling in the corridor and saying that a student wanted to see him. The period of the two quotations above is the end of the nineteenth century and the beginning of this century. Example C is taken from Tanizaki's The Fine-flaked Snow or The Makioka Sisters. The time is the 1930s. The mistress of an upper-middle class family sends for her maid, O-haru. "Did you call me, Madam?" O-haru opened the paper door fearfully. She had kneeled just outside the door sill, her hands on the floor. The color had left her face, since she had expected that she would be scolded. The positioning and posture of these women are similar to each other, but different in a meaningful way. They are sitting in the wood-floor corridor, just outside the doorsill, on which paper doors slide. When they speak to a superior, they are supposed to kneel, not keep standing. Note that superiors are also sitting on the tatami-floor. The woman in A has forgotten this rule, so she is reminded of that. In A and B, the women kneel down, but not sit back, implying that their business is something of minor importance and to be finished soon. But in C, the maid sits on her heels, and put her hands on the floor, so that she leans forwards. This is a polite and obedient posture expressing that one is ready to listen to one's superior. In "The Human Element", Somerset Maugham makes a good use of the meaning of positioning. Carruthers, a diplomat and novelist, visited Betty, whom he loved, on Rhode Island. He was met at the pier by Betty's chauffeur-a young British man. He thought that "it would be nicer of him to offer to sit by the chauffeur rather than behind by himself and was just going to suggest it, when the matter was taken out of his hands," since "the chauffeur told the porters to put Carruthers' bags at the back, and settling himself at the wheel said: 'Now if you'll hop in, we'll get along.'" Carruthers was Betty's guest. So it was proper for him to sit on the back seat, but he tried to show that he was a "nice" person by sitting next to the driver's seat. The matter proceeded as he had wanted, but not of his own will. He felt discontented and uncomfortable. Through these examination and analysis of examples from Japanese and British/American literature, we try to describe the meaning of verbal and nonverbal actions and the difference between the two language-cultures.
