This study was to investigate the influences of habitual mouth breathing on memory and learning ability during the growth period. At age 5 weeks, the experimental rats were subjected to surgery to close completely one side of the nasal cavity. An 8-arm radial maze was used to evaluate memory and learning ability at age 7, 11, and 15 weeks. Moreover, the brain was extracted at age 7, 11, and 15 weeks, and subjected to histomorphometric examination for the distribution and number of pyramidal cells in the hippocampal CA1 and CA3 regions after Nissl staining. The trial time to accomplish each task was significantly longer in the experimental group than in the control group throughout the experimental period. The number of pyramidal cells was significantly less in the experimental rats than in controls in both the CA1 and CA3 regions for the entire experimental period. Thus, the functional deterioration of the respiratory system during the growth phase exerts a substantial effect on the growth and development of the central nervous system.

Background: Kawasaki disease is suspected to be triggered by previous infection. The prevention measures for coronavirus disease 2019 (COVID-19) have reportedly reduced transmission of certain infectious diseases. Under these circumstances, the prevention measures for COVID-19 may reduce the incidence of Kawasaki disease.Methods: We conducted a retrospective study using registration datasets of patients with Kawasaki disease who were diagnosed in all 11 inpatient pediatric facilities in Yamanashi Prefecture. The eligible cases were 595 cases that were diagnosed before the COVID-19 pandemic (from January 2015 through February 2020) and 38 cases that were diagnosed during the COVID-19 pandemic (from March through November 2020). Incidence of several infectious disease were evaluated using data from the Infectious Disease Weekly Report conducted by the National Institute of Infectious Diseases.Results: Epidemics of various infectious diseases generally remained at low levels during the first 9 months (March through November 2020) of the COVID-19 pandemic. Moreover, the incidence of COVID-19 was 50–80 times lower than the incidence in European countries and the United States. The total number of 38 cases with Kawasaki disease for the 9 months during the COVID-19 pandemic was 46.3% (−3.5 standard deviations [SDs] of the average [82.0; SD, 12.7 cases] for the corresponding 9 months of the previous 5 years. None of the 38 cases was determined to be triggered by COVID-19 based on their medical histories and negative results of severe acute respiratory syndrome coronavirus 2 testing at admission.Conclusion: These observations provide a new epidemiological evidence for the notion that Kawasaki disease is triggered by major infectious diseases in children.

We had aimed to determine whether homeostasis model assessment–insulin resistance (HOMA-IR) reflects insulin resistance-sensitivity during treatment with a sodium-glucose cotransporter 2 inhibitor (SGLT2i). Hyperinsulinemic-euglycemic clamp analysis was performed in 22 patients with type 2 diabetic patients taking dapagliflozin (5 mg/day before or after breakfast). Propensity score matching of these individuals (SGLT2i group) for age, sex, body mass index, and clamp-derived tissue glucose uptake rate with 44 type 2 diabetic patients who had undergone clamp analysis without SGLT2i treatment (control group) identified 17 paired subjects in each group for further analysis of the relation between HOMA-IR and a clamp-derived insulin sensitivity index (ISI). Natural log–transformed HOMA-IR was negatively correlated with ISI in both SGLT2i (r = –0.527, p = 0.030) and control (r = –0.534, p = 0.027) groups. The simple regression lines for log-transformed HOMA-IR and ISI in the two groups showed similar slopes but differed in their intercepts. Multivariate analysis revealed that HOMA-IR for patients with the same ISI in the two groups was related by the formula: HOMA-IRcontrol = HOMA-IRSGLT2i × 2.45. In conclusion, HOMA-IR was well correlated with ISI during SGLT2i treatment, but values corresponding to the same ISI were lower in the SGLT2i group than in the control group.

本稿では、これまで編著した「あ~と」の続編として、「な~」で始まる用語について、対訳語が複数あるものを取り上げ、類義語との異同に関する注釈を付け、用例は実例を調査して、できるだけ多く提示することを試みた。法律用語の中には「内水」のように、国際法と国内法とで定義が異なるものがあり、このような場合は対訳語の使い分けができるように工夫することを心掛けた。また、あらぬ誤訳が生じることが懸念される場合は、特殊な法律専門用語よりも、対訳語として一般的に用いられる表現を優先的に取り上げるように配慮した。著者らは商事法和英辞典の作成途上にあり、本稿には不十分なところも多々あると考えている。お気づきの点があれはご教示願いたい。

The euryhaline mysid Neomysis awatschensis occurs widely in estuaries and brackish lakes in Japan. Although N. awatschensis may serve as an essential energy channel to higher trophic levels, its feeding ecology has been studied only in sheltered and desalinated Lake Kasumigaura. To determine major food sources for N. awatschensis, monthly sampling was conducted during the day along the microtidal Yura River estuary from March 2014 to July 2015. Additional sampling was conducted in May 2019 to assess diel vertical migration in relation to feeding. Microscopic examinations revealed that benthic diatoms (e.g. Bacillaria paxillifer and Navicula spp.) dominated in the stomach irrespective of developmental stage, diel cycle, or season. Stable carbon and nitrogen isotope ratios in N. awatschensis and its potential food sources indicated great contributions to the diet of N. awatschensis from benthic microalgae (mainly benthic diatoms) in 2014 and from particulate organic matter (mainly phytoplankton) in 2015. Although N. awatschensis always concentrated close to the bottom in 2019, diel vertical migration might have happened in 2015, when fresh water occupied the whole estuary possibly due to river mouth clogging. Under the stagnant and oligohaline conditions, N. awatschensis could have swum up into the water column and fed on phytoplankton at night in 2015. This hypothesis may resolve the apparent contradiction between the microscopy and stable isotope analysis in 2015. In the Yura River estuary, N. awatschensis feeds primarily on benthic diatoms, although it can utilize phytoplankton flexibly in response to prevailing environmental conditions.

Background: Skipping breakfast has been suspected as a risk factor for type 2 diabetes (T2DM), but the associations are not entirely consistent across ethnicities or sexes, and the issue has not been adequately addressed in the Japanese population.Methods: We followed 4631 participants (3600 men and 1031 women) in a work-site cohort of participants aged 35–66 years in 2002 through 2011 for T2DM development. Frequency of eating breakfast was self-reported and was subsequently dichotomized to breakfast skippers, who eat breakfast 3–5 times/week or less, and to eaters. Cox proportional hazards models were used to adjust for potential confounding factors, including dietary factors, smoking and other lifestyles, body mass index (BMI), and fasting blood glucose (FBG) at baseline.Results: During 8.9 years of follow-up, 285 T2DM cases (231 men and 54 women) developed. Compared to participants who reported eating breakfast every day, maximally-adjusted hazard ratios and 95% confidence intervals (CI) of those with the frequency of almost every day and 3–5, 1–2, and 0 days/week were: 1.06 (95% CI, 0.73–1.53), 2.07 (95% CI, 1.20–3.56), 1.37 (95% CI, 0.82–2.29), and 2.12 (95% CI, 1.19–3.76), respectively. In a dichotomized analysis, breakfast skipping was positively associated with T2DM incidence (maximally-adjusted hazard ratio 1.73; 95% CI, 1.24–2.42). The positive associations were found in both men and women, current and non-current smokers, normal weight and overweight (BMI ≥25 kg/m2), and normal glycemic status and impaired fasting glycemic status (FBG 110 to <126 mg/dL) individuals at baseline (Ps for interaction all >0.05).Conclusions: The present study in middle-aged Japanese men and women suggests that skipping breakfast may increase the risk of T2DM independent of lifestyles and baseline levels of BMI and FBG.

Forward genetics is a powerful approach based on chromosomal mapping of phenotypes and has successfully led to the discovery of many mouse mutations in genes responsible for various phenotypes. Although crossing between genetically remote strains can produce F2 and backcross mice for chromosomal mapping, the phenotypes are often affected by background effects from the partner strains in genetic crosses. Genetic crosses between substrains might be useful in genetic mapping to avoid genetic background effects. In this study, we investigated single nucleotide polymorphisms (SNPs) available for genetic mapping using substrains of C57BL/6 and BALB/c mice. In C57BL/6 mice, 114 SNP markers were developed and assigned to locations on all chromosomes for full utilization for genetic mapping using genetic crosses between the C57BL/6J and C57BL/6N substrains. Moreover, genetic differences were identified in the 114 SNP markers among the seven C57BL/6 substrains from five production breeders. In addition, 106 SNPs were detected on all chromosomes of BALB/cAJcl and BALB/cByJJcl substrains. These SNPs could be used for genotyping in BALB/cJ, BALB/cAJcl, BALB/cAnNCrlCrlj, and BALB/cCrSlc mice, and they are particularly useful for genetic mapping using crosses between BALB/cByJJcl and other BALB/c substrains. The SNPs characterized in this study can be utilized for genetic mapping to identify the causative mutations of the phenotypes induced by N-ethyl-N-nitrosourea mutagenesis and the SNPs responsible for phenotypic differences between the substrains of C57BL/6 and BALB/c mice.

Background: There is scarce data evaluating the current practice pattern and clinical outcomes for patients with severe aortic stenosis (AS), including both those who underwent surgical aortic valve replacement (SAVR) or transcatheter aortic valve implantation (TAVI) and those who were managed conservatively in the TAVI era.Methods and Results: The Contemporary outcomes after sURgery and medical tREatmeNT in patients with severe Aortic Stenosis (CURRENT AS) Registry-2 is a prospective, physician-initiated, multicenter registry enrolling consecutive patients who were diagnosed with severe AS between April 2018 and December 2020 among 21 centers in Japan. The rationale for the prospective enrollment was to standardize the assessment of symptomatic status, echocardiographic evaluation, and other recommended diagnostic examinations such as computed tomography and measurement of B-type natriuretic peptide. Moreover, the schedule of clinical and echocardiographic follow up was prospectively defined and strongly recommended for patients who were managed conservatively. The entire study population consisted of 3,394 patients (mean age: 81.6 years and women: 60%). Etiology of AS was degenerative in 90% of patients. AS-related symptoms were present in 60% of patients; these were most often heart failure symptoms. The prevalence of high- and low-gradient AS was 58% and 42%, respectively, with classical and paradoxical low-flow low-gradient AS in 4.5% and 6.7%, respectively.Conclusions: The CURRENT AS Registry-2 might be large and meticulous enough to determine the appropriate timing of intervention for patients with severe AS in contemporary clinical practice.

Invasive Aspergillus infection is a major factor for poor prognosis in patients receiving lung transplantation (LT). An antifungal agent, itraconazole (ITCZ), that has antimicrobial activity against Aspergillus species, is used as a prophylactic agent against Aspergillus infection after LT. ITCZ and its metabolite, hydroxyitraconazole (OH-ITCZ), potently inhibit CYP3A and P-glycoprotein that metabolize or excrete calcineurin inhibitors (CNIs), which are the first-line immunosuppressants used after LT; thus, concomitant use of ITCZ and CNIs could induce an increase in the blood concentration of CNIs. However, no criteria for dose reduction of CNIs upon concomitant use with ITCZ in LT recipients have been defined. In this study, the effect of ITCZ and OH-ITCZ on the blood concentrations of two CNIs, tacrolimus and cyclosporine, after LT were retrospectively evaluated. A total of 39 patients who received LT were evaluated. Effects of ITCZ and OH-ITCZ on the concentration/dosage (C/D) ratio of tacrolimus and cyclosporine were analyzed using linear mixed-effects models. The plasma concentrations of OH-ITCZ were about 2.5-fold higher than those of ITCZ. Moreover, there was a significant correlation between the plasma concentrations of ITCZ and OH-ITCZ. Based on parameters obtained in the linear regression analysis, the C/D ratios of cyclosporine and tacrolimus increase by an average of 2.25- and 2.70-fold, respectively, when the total plasma concentration of ITCZ plus OH-ITCZ is 1000 ng/mL. In conclusion, the plasma levels of ITCZ and OH-ITCZ could be key factors in drawing up the criterion for dose reduction of CNIs.

Spatial metabolomics uses imaging mass spectrometry (IMS) to localize metabolites within tissue section. Here, we performed matrix-assisted laser desorption/ionization-Fourier transform ion cyclotron resonance-IMS (MALDI-FTICR-IMS) to identify the localization of asparaptine A, a naturally occurring inhibitor of angiotensin-converting enzyme, in green spears of asparagus (Asparagus officinalis). Spatial metabolome data were acquired in an untargeted manner. Segmentation analysis using the data characterized tissue-type-dependent and independent distribution patterns in cross-sections of asparagus spears. Moreover, asparaptine A accumulated at high levels in developing lateral shoot tissues. Quantification of asparaptine A in lateral shoots using liquid chromatography-tandem mass spectrometry (LC-MS/MS) validated the IMS analysis. These results provide valuable information for understanding the function of asparaptine A in asparagus, and identify the lateral shoot as a potential region of interest for multiomics studies to examine gene-to-metabolite associations in the asparaptine A biosynthesis.

Cigarette smoke induces skeletal muscle wasting by a mechanism not yet fully elucidated. Branched-chain amino acids (BCAA) in the skeletal muscles are useful energy sources during exercise or systemic stresses. We investigated the relationship between skeletal muscle wasting caused by cigarette smoke and changes in BCAA levels in the plasma and skeletal muscles of rats. Furthermore, the effects of BCAA-rich diet on muscle wasting caused by cigarette smoke were also investigated. Wistar Kyoto (WKY) rats that were fed with a control or a BCAA-rich diet were exposed to cigarette smoke for four weeks. After the exposure, the skeletal muscle weight and BCAA levels in plasma and the skeletal muscles were measured. Cigarette smoke significantly decreased the skeletal muscle weight and BCAA levels in both plasma and skeletal muscles, while a BCAA-rich diet increased the skeletal muscle weight and BCAA levels in both plasma and skeletal muscles that had decreased by cigarette smoke exposure. In conclusion, skeletal muscle wasting caused by cigarette smoke was related to the decrease of BCAA levels in the skeletal muscles, while a BCAA-rich diet may improve cases of cigarette smoke-induced skeletal muscle wasting.

The tornadic debris signatures (TDSs) of the Ichihara Tornado associated with Typhoon Hagibis (2019) were observed using two operational C-band dual-polarization weather radars and an X-band phased-array weather radar (PAWR). This TDS observation was the first to be made over Japan in a typhoon environment. The TDS bins spread over time, and the maximum vertical and aerial extents reached 2.4 km and 9.41 km2, respectively. The estimated rise velocity of the TDS was 8 m s−1. The first TDS was detected ∼1 min before damage reporting began. The TDSs became clearer along with the rotational velocity of the near-surface vortex detected by PAWR. The copolar correlation coefficient reached a minimum (0.27) ∼1 min after the tornado passed the most severely damaged area and increased gradually over 4 min. This suggests that heavy and/or dense debris was lofted and immediately fell out and/or diffused, whereas light debris remained aloft for ≥ 4 min. By comparing the PAWR-detected vortex signatures with aerial photographs, we inferred that the first TDS comprised vegetated debris, the clearest TDS mainly comprised destroyed manmade structures. These results indicate that TDS detection is effective both for investigating damage and for recognizing tornado's occurrence even in a typhoon environment.

The taxonomic subdivision of certain Japanese freshwater amphipods into Jesogammarus jesoensis (Schellenberg, 1937), J. hokurikuensis Morino, 1985, J. fujinoi Tomikawa and Morino, 2003, and J. shonaiensis Tomikawa and Morino, 2003 was reassessed using both morphological and molecular data. Putative diagnostic morphological characters did not exhibit consistent geographic distributions, but eight geographically consistent monophyletic clades were detected based on COI and COI+12S rRNA sequence data. Uncorrected COI nucleotide divergences between the clades ranged from 7.0% to 16.4%, a level usually considered to represent inter-specific differences in crustaceans. No morphological features that differentiate the molecular clades were found. While the formal nomenclatural handling of these findings remains a problem for the future, for now we suggest the use of the term “J. jesoensis complex” to represent taxonomic entity consisting of the nominal taxa J. jesoensis, J. fujinoi, J. shonaiensis, and J. hokurikuensis, and potentially a number of hitherto cryptic taxa as well.
