Evaluating the function of genes expressed in fruit tissues of fruit tree species using a genetic transformation approach is a long process because the trees are generally recalcitrant to genetic transformation and cannot bear fruit during their long juvenile phases. Transient gene expression in fruit enables the functional analysis of genes associated with fruit traits, which may accelerate the study of fruit physiology. Here, by using the recently developed “Tsukuba system”, we successfully established an efficient transient expression system in harvested fruit tissues. The “Tsukuba system” utilizes a combination of the geminiviral replication system and a double terminator, which ensures sufficient levels of transgene expression. We used blueberry fruit as a model to characterize the applicability of this system for transient expression in fruit tissue. The pTKB3-EGFP vector was introduced by agroinfiltration into the fruit tissues of several blueberry cultivars. We found that transient GFP fluorescence in fruit peaked 4–6 days after agroinfiltration. Agrobacterium suspensions were easily injected into soft, mature fruit, and GFP was strongly expressed; however, hard, immature fruit were not penetrable by Agrobacterium suspensions, and GFP was rarely detected. We then tested the applicability of the developed system to other fruit tree species: six families, 17 species, and 26 cultivars. GFP fluorescence was detected in all species, except for Japanese apricot. In blueberry, bilberry, sweet cherry, apricot, and satsuma mandarin, GFP was highly expressed and observed in a large proportion of the flesh. In kiwifruit, hardy kiwifruits, persimmon, peach, apple, European pear, and grape, GFP fluorescence was limited to certain parts of the fruits. Finally, transient VcMYBA1 overexpression in blueberry was tested as a model for gene functional analysis in fruit. Transient VcMYBA1 overexpression induced red pigmentation in the flesh, suggesting that VcMYBA1 expression caused anthocyanin accumulation. This study provides a technical basis for the rapid evaluation of genes expressed in fruit, which will be useful for gene function evaluation studies in fruit crops with long juvenile phases.

We investigated bluish-purple Platycodon grandiflorus flowers for yet unidentified flavonoid-related compounds and their flower coloration mechanisms. We identified a new polyacylated anthocyanin, delphinidin 3-O-[6-O-(α-rhamnopyranosyl)-β-glucopyranoside]-7-O-[6-O-(4-O-(6-O-(4-O-(6-O-(4-O-(β-glucopyranosyl)-trans-caffeoyl)-β-glucopyranosyl)-trans-caffeoyl)-β-glucopyranosyl)-trans-caffeoyl)-β-glucopyranoside] (3) as the minor anthocyanin component along with four known anthocyanins, four known flavones, and chlorogenic acid. The major anthocyanin of the bluish-purple P. grandiflorus flowers is platyconin. While platyconin has two caffeic acids linked in series via glucose molecules at the 7-position of delphinidin, anthocyanin 3 has three caffeic acids linked in series via glucose molecules at the 7-position of delphinidin. To investigate the effects of the number of aromatic acyl groups in polyacylated anthocyanin on bluing and stability, the color and stability of anthocyanin 3 in diluted aqueous solution (5 × 10−5 M) at a weak acidic condition (pH 5.7, the same pH as the petal sap) were compared with those of platyconin. Results showed that even if the three caffeic acids were bound in series at the 7-position of delphinidin, there was no further bluing or stabilizing effects of anthocyanin 3 compared to platyconin, but a tendency toward weakening color intensity was observed. Investigation of the change in anthocyanin content at the flower developmental stages showed that delphinidin 3-rutinoside-7-glucoside, which is the deacylated form of platyconin, first accumulated up to sufficient amounts at the bud stage, followed by an increase in platyconin content. At the bud stage, the petals were not purple or violet, except for veins, despite the presence of a good amount of delphinidin 3-rutinoside-7-glucoside, indicating the delphinidin 3-rutinoside-7-glucoside was present in some colorless state.

This paper presents a new efficient real-coded genetic algorithms for constrained optimization. In general, it is efficient to use the gradient methods for the constrained optimization. However, because actual design problems often include extremely nonlinear or discontinuous characters, the gradient methods do not necessarily work well. Therefore, the present authors developed the "Center Neighborhood Crossover (CNX)" for real-coded genetic algorithms and applied to truss structure optimization. But, GAs have some fatal problems in handling the constraints. In the constrained optimization using GAs, designers have to translate the constrained problem to a basic unconstrained problem by using some methods such as the penalty function methods. It is, however, difficult to decide appropriate penalty parameters. In the present study, we developed "Real-coded Genetic Algorithms with Active Constraints (RGAAC)" In RGAAC, the points outside feasible area is pulled back to active constraints by the gradient method. The present method enables an efficient search.

土子金四郎著「洒落哲学」は日本最初の笑い学研究である。「洒落哲学」の内容、成立事情を説明する。「洒落哲学」を批判した大西祝の「洒落哲学を評す」の摘要を示し、最後に大西祝の論文「滑稽の本性」の内容を示して、「洒落哲学」が近代日本での笑い学研究の魁であることを証明する。付では「洒落哲学」のうち洒落の種類を表にして紹介する。

Internal carotid artery (ICA) ligation for placing a high-flow extracranial-intracranial (EC-IC) bypass is used in patients with aneurysms on the cavernous portion of the ICA. Recanalization and rupture after proximal ICA ligation can occur. We present four patients who underwent endovascular distal ICA occlusion and report our surgical technique and treatment results. We ligated the ICA to place an EC-IC bypass using a radial artery (RA) graft. Failure to obtain spontaneous occlusion in the distal region required endovascular treatment an average of 219 days later. A guide catheter was placed in the common carotid artery, a guide or distal access catheter was introduced in the RA graft from the external carotid artery, and a microcatheter was navigated into the cavernous aneurysm through the RA graft. Using detachable coils, endovascular ICA occlusion was from just distal to the aneurysmal neck to a site proximal to the origin of the ophthalmic artery. Aneurysmal occlusion was completed by endovascular occlusion of the distal ICA. Complications were RA graft stenosis and transient consciousness disturbance due to local subarachnoid hemorrhage. Outpatient follow-up for a mean of 109.5 months revealed no recurrences. Distal occlusion of the ICA through the implanted RA graft is simple and presents a low risk for cerebral infarction due to thrombus formation during the procedure. To treat cavernous carotid aneurysms that do not disappear after placing the EC-IC bypass after ICA ligation at the aneurysmal neck, we offer our procedure as a treatment option.

小児に対する前投薬として,ミダゾラム注腸投与の効果を,1~6歳の小児109例を対象とし,投与時間(導入30分前と60分前),投与量(0.2mg/kgと0.4mg/kg)および年齢(1~3歳と4~6歳)に分けて検討した.鎮静スコア1(導入時にマスクを嫌がるもの)を無効,スコア2~4を有効,スコア5を過剰鎮静として評価すると,30分前投与群で,投与量に関係なく,年長児の有効率(92~100%)は年少児(54~58%)に比較して有意に高く,この傾向は投与時間を導入60分前にしても同様であった.以上の結果からミダゾラム0.2~0.4mg/kgの30~60分前注腸投与法は,年少児での有効性は十分でないが,4~6歳の就学前児童の前投薬としては有用な方法と思われた.

生産量の増加が期待される施設栽培のパプリカを対象とし,同一園芸施設で栽培された2014年産,2015年産の適熟果を5品種ずつ用いて,Na,K,Ca,Mg,P,Fe,Zn,CuおよびMnを分析し,品種と収穫時期の違いによる含有量の変動を検討した.二元配置分散分析の結果,2014年産はすべての無機元素の品種間,収穫日間およびCa以外の無機元素の交互作用で有意な差が認められ,2015年産はすべての無機元素の品種間,収穫日間および交互作用で有意な差が認められた.ただし,品種別に整理すると,品種間や色による明らかな違いが見られなかったこと,および品種によらず,収穫時期の違いによる無機元素含有量の変動状況が似ていることなどから,パプリカに含まれる無機元素は,品種や色の違いより,収穫時期の影響が大きいと判断された.本報告は,施設栽培パプリカにおける品種と収穫時期の違いによる成分変動の一例であるが,本報告の2015年産パプリカ試料では,CaやMnは夏季から初秋にかけて低い傾向があり,また夏季から初秋にかけてKおよびZnが高い傾向があった.

食用ウニ類は外観から生殖巣重量や生殖巣指数を予測することが困難である.本研究では,青果物の内部品質の非破壊評価に広く用いられている透過方式の近赤外分光装置を用いて,キタムラサキウニ(Strongylocentrotus nudus)の生殖巣重量および生殖巣指数の評価手法の開発を試みた.試料は,2021年および2022年に宮城県石巻市または女川町で水揚げされたキタムラサキウニ216個体を用いた.得られた近赤外吸収スペクトルを主成分分析で解析したところ,2次微分処理を行ったスペクトルの第1主成分の寄与率は90 %であり,そのローディングベクトルでは,主に脂質の吸収帯である930 nm付近に負のピークが観察された.また,Partial Least Squares回帰分析を行った結果,2次微分スペクトルを用いた生殖巣重量の検量モデルの評価結果は,因子数4,相関係数0.78,標準誤差4.4 gであった.同様に生殖巣指数では,因子数4,相関係数0.76,標準誤差4.6 g/100 gであった.これらの結果から,生殖巣重量および生殖巣指数を非破壊・非接触的に評価できることが示唆され,これは生殖巣の脂質が検知されることによるものと考えられた.本手法を用いた非破壊評価システムの構築により,天然については身入りの良好な個体を出荷し,身入りの悪い個体は海に放流あるいは養殖に向ける等,新たな判断基準として活用することができる.

A cyclone over the Arabian Sea has lots of effect on people's life in India and it sometimes bring Indian summer monsoon. In the present study, cyclogenesis and its development over the Arabian Sea are investigated by using satellite observation data and atmospheric reanalysis data. There is large temperature gradient around the coastal area of the Arabian Peninsula between warmer air over land and cooler air over ocean. Baroclinic instability by the horizontal temperature gradient in that region easily generates a seed of cyclone at 850hPa. Regarding development of cyclone over the Arabian Sea, water vapor over the region of a seed of cyclone does not show significant difference between development and non-development case. On the other hand, occurrence of air disturbance in middle-upper troposphere and coupling of upper and lower air disturbances are considered to be more important for the development of cyclone over the Arabian Sea.

本研究では精油の抽出にあたり,簡易な方法で製作,使用できる水蒸気蒸留装置を試作し,その性能を評価した。また,試作機を用いて,兵庫県丹波市の里山の樹木から分析事例の少ないヒメクロモジ(Lindera lancea),アブラチャン(Lindera praecox),コクサギ(Orixa japonica)の3種を選定し,精油の抽出および成分分析(GC–MS分析)を行った。結果,水蒸気蒸留装置は,市販品よりも安価(製作費約6万円)かつ短時間(約3時間)で製作された。装置の重量は約13.5 kgで,植物材料は1.5 kg,水は1.5 Lまでの蒸留が可能で,屋内外でも使用できる持ち運び可能な装置である。得られた3種類の精油の含有成分は,ヒメクロモジ35種類,アブラチャン40種類,コクサギ39種類であった。精油の主成分は,ヒメクロモジではリナロール(41.26%),アブラチャンではカンファー(22.60%),コクサギではα-ピネン(23.81%)であった。近年の分析結果が少ないその成分が本研究で明らかになった。これらのことから,教育現場や里山などさまざまな場所で地域資源を利用した和精油による嗅覚教育の可能性が高まった。

2005年に開催された愛・地球博に対する会場近隣大学学生の評価を明らかにするため,愛知工業大学学生の意識調査を実施した。学生の愛・地球博への関わり方に対して6つの仮説を立て,それらに沿って意識調査の質問項目を作成した。得られた回答を分析し,前回報告された愛知県立大学学生の評価と比較したところ,近隣大学と言えども,2大学間の学生の評価にはいつかの差異が見られた。ただ両大学学生とも,愛・地球博開催期間中,それが近隣で行われた故の悪影響を大きく受けており,総合的評価が低いことでは一致を見た。これらの評価を参考にし,今後,万博や大規模なイベントが開催される際,主催者が近隣大学に対して配慮すべき点について言及した。

筆者はアジア地域研究連携機構研究紀要の第1号に「秋田犬の国際化戦略」と題して秋田犬が世界的に人気を集めていることを紹介したが、その後秋田犬は「高貴な犬」として各国で人気がさらに高まっている。海外の人気が国内にブーメラン効果で還流する動きもあり、秋田県大館市は福原市長の下、秋田犬利用プロジェクト「ハチ公の駅」を設置し、観光の目玉にする構想を進めている。国際教養大でも4月、秋田犬ブランドの活用法をめぐり読売新聞主催のシンポジウムが行われた。秋田犬は秋田が独占可能な唯一のブランドでもあり、官民一体で秋田犬人気の相乗効果を狙うべきだ。

In this paper, we review the four processes in the psychological flexibility model of Acceptance and Commitment Therapy (ACT) as mindfulness in the context of ACT. We will discuss in the following order: (1) human psychopathology from the perspective of ACT, (2) human health from the perspective of ACT, and (3) mindfulness from the perspective of ACT. The “purpose” and “method” of using mindfulness training or technique in ACT involves originality and will be discussed by introducing concrete examples of practice.
